    We present a new computational technique (a software implementation, data sets, and supplementary information are available at http://www.enm.bris.ac.uk/lpd/) which enables the probabilistic analysis of cDNA microarray data and we demonstrate its effectiveness in identifying features of biomedical importance. A hierarchical Bayesian model, called latent process decomposition (LPD), is introduced in which each sample in the data set is represented as a combinatorial mixture over a finite set of latent processes, which are expected to correspond to biological processes. Parameters in the model are estimated using efficient variational methods. This type of probabilistic model is most appropriate for the interpretation of measurement data generated by cDNA microarray technology. For determining informative substructure in such data sets, the proposed model has several important advantages over the standard use of dendrograms. First, the ability to objectively assess the optimal number of sample clusters. Second, the ability to represent samples and gene expression levels using a common set of latent variables (dendrograms cluster samples and gene expression values separately which amounts to two distinct reduced space representations). Third, in contrast to standard cluster models, observations are not assigned to a single cluster and, thus, for example, gene expression levels are modeled via combinations of the latent processes identified by the algorithm. We show this new method compares favorably with alternative cluster analysis methods. To illustrate its potential, we apply the proposed technique to several microarray data sets for cancer. For these data sets it successfully decomposes the data into known subtypes and indicates possible further taxonomic subdivision in addition to highlighting, in a wholly unsupervised manner, the importance of certain genes which are known to be medically significant.     Sensitivity of mixing layers to three-dimensional forcing

    It is well known that turbulent mixing layers are dominated by large scale, fairly coherent structures, and that these structures are related to the stability characteristics of the flow. These facts have led researchers to attempt controlling such flows by selectively forcing certain unstable modes, which can in addition have the effect of suppressing other modes. Much of the work on controlling the mixing layer has relied on forcing 2-D instabilities. The results of forcing 3-D instabilities are addressed. The objectives of the work are twofold: to understand how a mixing layer responds to 3-D perturbations, and to test the validity of an amplitude expansion in predicting the mixing layer development. The amplitude expansion could be very useful in understanding and predicting the 3-D response of the flow to a variety of initial conditions

    Dynamics of coherent structures in a plane mixing layer

    An incompressible, time developing 3-D mixing layer with idealized initial conditions was simulated numerically. Consistent with the suggestions from experimental measurements, the braid region between the dominant spanwise vortices or rolls develops longitudinal vortices or ribs, which are aligned upstream and downstream of a roll and produce spanwise distortion of the rolls. The process by which this distortion occurs is explained by studying a variety of quantities of dynamic importance (e.g., production of enstrophy, vortex stretching). Other quantities of interest (dissipation, helicity density) are also computed and discussed. The currently available simulation only allows the study of the early evolution (before pairing) of the mixing layer. New simulations in progress will relieve this restriction

    Spatial electron density and electric field strength measurements in microwave cavity experiments

    Measurements of electron density and electric field strength have been made in an argon plasma contained in a resonant microwave cavity at 2.45 GHz. Spatial measurements of electron density, n sub e, are correlated with fluorescence observations of the discharge. Measurements of n sub e were made with Stark broadening and compared with n sub 3 calculated from measured plasma conductivity. Additional measurements of n sub 3 as a function of pressure and in mixtures of argon and oxygen are presented for pressures from 10 Torr to 1 atm. Measurements in flowing gases and in static systems are presented. In addition, limitations of these measurements are identified

The corresponding Ito integrals are defined and the existence and uniqueness of solutions to a class of stochastic differential equations is established.     Asking the experts : developing and validating parental diaries to assess children's minor injuries

    The methodological issues involved in parental reporting of events in children's everyday lives are discussed with reference to the development and validation of an incident diary, collecting concurrent data on minor injuries in a community study of children under eight years old. Eighty-two mothers participated in a comparison over nine days of daily telephone interviews and structured incident diaries. Telephone methods resulted in more missing data, and participants in both groups expressed a preference for the diary method. This diary was then validated on a sample of 56 preschool and school-aged children by comparing injury recording by a research health visitor with that of their mothers. Each failed to report some injuries, but there was good agreement overall, and in descriptive data on injuries reported by both. Parental diaries have the potential to provide rich data, of acceptable validity, on minor events in everyday life
