Identity in crisis : the politics of humanitarian intervention
This thesis examines the theory and practice of humanitarian intervention in the early post-Cold War era. Taking as its basis US policy towards Somalia, Rwanda and Haiti between
1992 and 1994, it develops a theory of humanitarian intervention based on constructivist
and scientific realist principles. Using identity as the organising concept, the thesis
examines the meta-theoretical precepts of constructivism and scientific realism, which are
developed into a methodology for analysing questions of foreign policy. Incorporating
critical insights from sequential path analysis, morphogenetic social analysis - the notion
of a dynamic mutual constitution of structure and agency - and constructivist social theory,
the case studies provide a useful new means of conceptualising humanitarian intervention
as a foreign policy practice through an identity-driven analysis. The findings of the
research shed much light on this practice and its future prospects. They also suggest new
directions for a scientific realist/constructivist research agenda
Metallocene catalysts for the ring-opening co-polymerisation of epoxides and cyclic anhydrides
The ring-opening co-polymerisation (ROCOP) of epoxides and cyclic anhydrides is a versatile route to new polyesters. The vast number of monomers that are readily available means that an effectively limitless number of unique polymeric materials can be prepared using a common synthetic route, thereby paving the way to polymers that are more recyclable and could be used in applications that currently lie exclusively within the remit of polyolefins. Metallocene complexes [Cp2MCl2] (M = Ti, Zr, Hf, Cp = η5-C5H5) are known to be excellent pre-catalysts for olefin polymerisation and yet have not been reported for ROCOP, despite their supreme pedigree in the polymerisation realm. Herein, we report the first application of [Cp2MCl2] as catalysts for the ROCOP of epoxides and anhydrides and show that they are effective catalysts for this reaction. The catalytic performances are good with standard epoxides such as cyclohexene oxide but are excellent with more challenging sterically demanding limonene oxide, and this therefore represents a highly effective catalyst system for the preparation of bio-derived recyclable polymers

Global Analysis of Palmitoylated Proteins in Toxoplasma gondii
SummaryPost-translational modifications (PTMs) such as palmitoylation are critical for the lytic cycle of the protozoan parasite Toxoplasma gondii. While palmitoylation is involved in invasion, motility, and cell morphology, the proteins that utilize this PTM remain largely unknown. Using a chemical proteomic approach, we report a comprehensive analysis of palmitoylated proteins in T. gondii, identifying a total of 282 proteins, including cytosolic, membrane-associated, and transmembrane proteins. From this large set of palmitoylated targets, we validate palmitoylation of proteins involved in motility (myosin light chain 1, myosin A), cell morphology (PhIL1), and host cell invasion (apical membrane antigen 1, AMA1). Further studies reveal that blocking AMA1 palmitoylation enhances the release of AMA1 and other invasion-related proteins from apical secretory organelles, suggesting a previously unrecognized role for AMA1. These findings suggest that palmitoylation is ubiquitous throughout the T. gondii proteome and reveal insights into the biology of this important human pathogen
Observational Constraints on the Ages of Molecular Clouds and the Star-Formation Timescale: Ambipolar-Diffusion--Controlled or Turbulence-Induced Star Formation?
We revisit the problem of the star formation timescale and the ages of
molecular clouds. The apparent overabundance of star-forming molecular clouds
over clouds without active star formation has been thought to indicate that
molecular clouds are "short-lived" and that star formation is "rapid". We show
that this statistical argument lacks self-consistency and, even within the
rapid star-formation scenario, implies cloud lifetimes of approximately 10 Myr.
We discuss additional observational evidence from external galaxies that
indicate lifetimes of molecular clouds and a timescale of star formation of
approximately 10 Myr . These long cloud lifetimes in conjunction with the rapid
(approximately 1 Myr) decay of supersonic turbulence present severe
difficulties for the scenario of turbulence-controlled star formation. By
contrast, we show that all 31 existing observations of objects for which the
linewidth, the size, and the magnetic field strength have been reliably
measured are in excellent quantitative agreement with the predictions of the
ambipolar-diffusion theory. Within the ambipolar-diffusion-controlled star
formation theory the linewidths may be attributed to large-scale non-radial
cloud oscillations (essentially standing large-amplitude, long-wavelength
Alfven waves), and the predicted relation between the linewidth, the size, and
the magnetic field is a natural consequence of magnetic support of

The molecular basis of color vision in colorful fish: Four Long Wave-Sensitive (LWS) opsins in guppies (Poecilia reticulata) are defined by amino acid substitutions at key functional sites
<p>Abstract</p> <p>Background</p> <p>Comparisons of functionally important changes at the molecular level in model systems have identified key adaptations driving isolation and speciation. In cichlids, for example, long wavelength-sensitive (LWS) opsins appear to play a role in mate choice and male color variation within and among species. To test the hypothesis that the evolution of elaborate coloration in male guppies (<it>Poecilia reticulata</it>) is also associated with opsin gene diversity, we sequenced long wavelength-sensitive (LWS) opsin genes in six species of the family Poeciliidae.</p> <p>Results</p> <p>Sequences of four LWS opsin genes were amplified from the guppy genome and from mRNA isolated from adult guppy eyes. Variation in expression was quantified using qPCR. Three of the four genes encode opsins predicted to be most sensitive to different wavelengths of light because they vary at key amino acid positions. This family of LWS opsin genes was produced by a diversity of duplication events. One, an intronless gene, was produced prior to the divergence of families Fundulidae and Poeciliidae. Between-gene PCR and DNA sequencing show that two of the guppy LWS opsins are linked in an inverted orientation. This inverted tandem duplication event occurred near the base of the poeciliid tree in the common ancestor of <it>Poecilia </it>and <it>Xiphophorus</it>. The fourth sequence has been uncovered only in the genus <it>Poecilia</it>. In the guppies surveyed here, this sequence is a hybrid, with the 5' end most similar to one of the tandem duplicates and the 3' end identical to the other.</p> <p>Conclusion</p> <p>Enhanced wavelength discrimination, a possible consequence of opsin gene duplication and divergence, might have been an evolutionary prerequisite for color-based sexual selection and have led to the extraordinary coloration now observed in male guppies and in many other poeciliids.</p
Chromosome 20q Amplification Regulates in Vitro Response to Kinesin-5 Inhibitor
We identified gene expression signatures predicting responsiveness to a Kinesin-5 (KIF11) inhibitor (Kinesin-5i) in cultured colon tumor cell lines. Genes predicting resistance to Kinesin-5i were enriched for those from chromosome 20q, a region of frequent amplification in a number of tumor types. siRNAs targeting genes in this chromosomal region identified AURKA, TPX2 and MYBL2 as genes whose disruption enhances response to Kinesin-5i. Taken together, our results show functional interaction between these genes, and suggest that their overexpression is involved in resistance to Kinesin-5i. Furthermore, our results suggest that patients whose tumors overexpress AURKA due to amplification of 20q will more likely resist treatment with Kinesin-5 inhibitor, and that inactivation of AURKA may sensitize these patients to treatment
Incidence and Outcomes Associated With Clostridium difficile Infections: A Systematic Review and Meta-analysis
Importance:
An understanding of the incidence and outcomes of Clostridium difficile infection (CDI) in the United States can inform investments in prevention and treatment interventions.
Objective:
To quantify the incidence of CDI and its associated hospital length of stay (LOS) in the United States using a systematic literature review and meta-analysis.
Data Sources:
MEDLINE via Ovid, Cochrane Library Databases via Wiley, Cumulative Index of Nursing and Allied Health Complete via EBSCO Information Services, Scopus, and Web of Science were searched for studies published in the United States between 2000 and 2019 that evaluated CDI and its associated LOS.
Study Selection:
Incidence data were collected only from multicenter studies that had at least 5 sites. The LOS studies were included only if they assessed postinfection LOS or used methods accounting for time to infection using a multistate model or compared propensity score-matched patients with CDI with control patients without CDI. Long-term-care facility studies were excluded. Of the 119 full-text articles, 86 studies (72.3%) met the selection criteria.
Data Extraction and Synthesis:
Two independent reviewers performed the data abstraction and quality assessment. Incidence data were pooled only when the denominators used the same units (eg, patient-days). These data were pooled by summing the number of hospital-onset CDI incident cases and the denominators across studies. Random-effects models were used to obtain pooled mean differences. Heterogeneity was assessed using the I2 value. Data analysis was performed in February 2019.
Main Outcomes and Measures:
Incidence of CDI and CDI-associated hospital LOS in the United States.
Results:
When the 13 studies that evaluated incidence data in patient-days due to hospital-onset CDI were pooled, the CDI incidence rate was 8.3 cases per 10 000 patient-days. Among propensity score-matched studies (16 of 20 studies), the CDI-associated mean difference in LOS (in days) between patients with and without CDI varied from 3.0 days (95% CI, 1.44-4.63 days) to 21.6 days (95% CI, 19.29-23.90 days).
Conclusions and Relevance:
Pooled estimates from currently available literature suggest that CDI is associated with a large burden on the health care system. However, these estimates should be interpreted with caution because higher-quality studies should be completed to guide future evaluations of CDI prevention and treatment interventions
