    Optimasi Amobilisasi Bromelin Menggunakan Matriks Pendukung Kitosan

    Optimasi amobilisasi bromelin dengan matriks pendukung kitosan telah diteliti. Enzim bromelin yang diisolasi dari buah nanas mentah maupun matang diperoleh aktivitas tertinggi pada pengendapan 40% dengan ammonium sulfat jenuh. Kondisi optimum amobilisasi enzim bromelin didapatkan pada konsentrasi kitosan 240,4 mg dan bromelin 10 mg dengan jumlah enzim teramobil 9,5417 mg dan aktivitas dalam 10 mg enzim amobil yang diuji adalah 86,2067 Unit. Uji aktivitas enzim amobil terhadap pengaruh substrat kasein didapatkan aktivitas tertinggi pada kasein 3000 ppm. Uji Perulangan enzim amobil dapat digunakan hingga enam kali dengan efisiensi 57,69%

    Assessment of the quality of life in parents of children suffering from epilepsy

    Background: In case of paediatric epilepsy, parents are responsible for providing medicines, round the clock care to the child, while also sustaining their own needs and expectations from life. Socioeconomic status, interaction with healthcare workers, and the level of monetary and emotional support from external agencies play an important role in modelling the quality of life of these parents. The authors’ objective was to assess the quality of life of parents of children suffering from epilepsy and evaluate the influence of demographic and clinical variables. Methods: We conducted a cross-sectional analytical study at the out-patient department of our hospital. We recruited parents of epileptic children and those of children not suffering from any chronic illness and elicited demographic data, clinical history of illness, and QOL using the Quality of Life in the Child’s Chronic Disease Questionnaire of parents in both groups followed by analysis of the data by applying appropriate statistical tests. We used Student t- test to find the significance of data on continuous scale and Chi-square test for discrete variables. One-way ANOVA test was performed to find the influence of various variables on QOL of parents. Results: Male child, female caregiver, Hindu religion, illiterate parent, low monthly family income, residence in rural areas, GTCS type of seizure, frequent seizures during the day, and multiple medications negatively affected the QOL of parents. Conclusions: To provide holistic care to epileptic children, we need to integrate and provide social, occupational, emotional, and financial help to their parents in an acceptable, affordable, and sustainable way.

    Numerous cosmetic enhancements and augmentations to the natural appearance of the periorbital area are readily available today. Due to the increasing popularity of these cosmetic procedures, it is important for ophthalmologists to be aware of their potential risks, complications and adverse effects. The aim of this literature review was to introduce some of the most common ocular cosmetic enhancements and provide a comprehensive overview of their associated adverse effects reported in various medical journals. PubMed, Embase, and Google Scholar were used to identify articles related to the following ocular cosmetic procedures; eyelash extensions, permanent eyelid tattooing, and eyelash dyeing. The most common complication associated with eyelash extensions was allergic blepharitis (79%). Allergic granulomatous reactions were the predominant complication in patients who underwent eyelid tattooing (56%). Besides, 60% of subjects who underwent eyelash dyeing experienced allergic contact dermatitis as the most common adverse effect. Although millions of these procedures are performed annually without any adverse effects, reports of complications continue to increase in the literature. Knowledge of the possible adverse effects associated with these enhancements is important for eye care providers and licensed estheticians to be aware of given both the direct and indirect effects they may have on ocular health and visual outcomes

    GROWTH MINDSET IN ENGLISH LANGUAGE LEARNING OF COLLEGE STUDENTS

    This study investigated the growth mindset of college students in English language learning and the challenges they encounter in an online setting. Using a researcher-made questionnaire about the growth mindset of college students in English language learning and the challenges they encounter in an online setting, data from the 60 respondents in the study coming from the different colleges or departments of the Notre Dame of Midsayap College were analyzed quantitatively. Findings indicated that students have a high growth mindset and that factors such as motivation, criticism, environment, and effort had played a major role in their English language learning. Major challenges of students in an online setting include finding difficulty in accessing online materials due to poor internet connection, experiencing headaches and body pains due to prolonged exposure to gadgets, and getting bored easily when class time is too lengthy. The least challenges encountered by students in an online setting yet also hampered their learning are lacking gadgets such as a cellphone and laptops, feeling disconnected from the instructor, and excessive workloads. Furthermore, there is no significant difference in a growth mindset in terms of sex and college or department. However, it was found out that College of Nursing (CN) got the highest mean when it comes to their growth mindset. Meanwhile, College of Information Technology, and Engineering (CITE) got the lowest mean. Moreover, there is a significant relationship between the growth mindset of English language learning and the challenges they encountered in an online setting.  Article visualizations

    Ageing enhances cellular immunity to myeloperoxidase and experimental anti-myeloperoxidase glomerulonephritis

    OBJECTIVES: Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis is an autoimmune disease characterised by small blood vessel inflammation, commonly affecting the kidneys and respiratory tract. It is unclear why the incidence of this condition increases with age. Previous studies in a passive antibody transfer system in aged mice have implicated innate effectors. To test the hypothesis that autoimmunity to myeloperoxidase, an autoantigen responsible for ANCA-associated vasculitis, increases with age, anti-myeloperoxidase autoimmunity was studied in murine models of active autoimmunity and disease induced by cellular immunity. METHODS: Young (8 weeks) and aged (either 15 or 22 month) mice were immunised with whole proteins or peptides from ovalbumin, as a model foreign antigen, or myeloperoxidase protein or peptides. Mice were subjected to a model of active anti-myeloperoxidase glomerulonephritis. Cellular and humoral immune responses and tissue inflammation were assessed. RESULTS: While cellular immunity to ovalbumin was diminished in aged mice, cellular autoimmunity to myeloperoxidase and its immunodominant CD4+ and CD8+ T cell epitopes was increased after immunization with either MPO peptides or whole MPO protein, assessed by peptide and antigen specific production of the pro-inflammatory cytokines interferon-γ and interleukin-17A. MPO-ANCA titres were not increased in aged mice compared with young mice. In experimental anti-MPO glomerulonephritis, cell mediated injury was increased, likely due to CD4+ and CD8+ T cells, innate immunity and the increased vulnerability of aged kidneys. CONCLUSION: Heightened cellular immunity to MPO develops with ageing in mice and may contribute to the increased incidence and severity of ANCA-associated vasculitis in older people

    On Passion and Sports Fans:A Look at Football

    The purpose of the present research was to test the applicability of the Dualistic Model of Passion (Vallerand et al., 2003) to being a sport (football) fan. The model posits that passion is a strong inclination toward an activity that individuals like (or even love), that they value, and in which they invest time and energy. Furthermore, two types of passion are proposed: harmonious and obsessive passion. While obsessive passion entails an uncontrollable urge to engage in the passionate activity, harmonious passion entails a sense of volition while engaging in the activity. Finally, the model posits that harmonious passion leads to more adaptive outcomes than obsessive passion. Three studies provided support for this dualistic conceptualization of passion. Study 1 showed that harmonious passion was positively associated with adaptive behaviours (e.g., celebrate the team’s victory), while obsessive passion was rather positively associated with maladaptive behaviours (e.g., to risk losing one’s employment to go to the team’s game). Study 2 used a short Passion Scale and showed that harmonious passion was positively related to the positive affective life of fans during the 2006 FIFA World Cup, psychological health (self-esteem and life satisfaction), and public displays of adaptive behaviours (e.g., celebrating one’s team victory in the streets), while obsessive passion was predictive of maladaptive affective life (e.g., hating opposing team’s fans) and behaviours (e.g., mocking the opposing team’s fans). Finally, Study 3 examined the role of obsessive passion as a predictor of partner’s conflict that in turn undermined partner’s relationship satisfaction. Overall, the present results provided support for the Dualistic Model of Passion. The conceptual and applied implications of the findings are discussed

    Separate F-Type Plasmids Have Shaped the Evolution of the H30 Subclone of Escherichia coli Sequence Type 131.

    The extraintestinal pathogenic Escherichia coli (ExPEC) H30 subclone of sequence type 131 (ST131-H30) has emerged abruptly as a dominant lineage of ExPEC responsible for human disease. The ST131-H30 lineage has been well described phylogenetically, yet its plasmid complement is not fully understood. Here, single-molecule, real-time sequencing was used to generate the complete plasmid sequences of ST131-H30 isolates and those belonging to other ST131 clades. Comparative analyses revealed separate F-type plasmids that have shaped the evolution of the main fluoroquinolone-resistant ST131-H30 clades. Specifically, an F1:A2:B20 plasmid is strongly associated with the H30R/C1 clade, whereas an F2:A1:B− plasmid is associated with the H30Rx/C2 clade. A series of plasmid gene losses, gains, and rearrangements involving IS26 likely led to the current plasmid complements within each ST131-H30 sublineage, which contain several overlapping gene clusters with putative functions in virulence and fitness, suggesting plasmid-mediated convergent evolution. Evidence suggests that the H30Rx/C2-associated F2:A1:B− plasmid type was present in strains ancestral to the acquisition of fluoroquinolone resistance and prior to the introduction of a multidrug resistance-encoding gene cassette harboring blaCTX-M-15. In vitro experiments indicated a host strain-independent low frequency of plasmid transfer, differential levels of plasmid stability even between closely related ST131-H30 strains, and possible epistasis for carriage of these plasmids within the H30R/Rx lineages. IMPORTANCE A clonal lineage of Escherichia coli known as ST131 has emerged as a dominating strain type causing extraintestinal infections in humans. The evolutionary history of ST131 E. coli is now well understood. However, the role of plasmids in ST131’s evolutionary history is poorly defined. This study utilized real-time, single-molecule sequencing to compare plasmids from various current and historical lineages of ST131. From this work, it was determined that a series of plasmid gains, losses, and recombinational events has led to the currently circulating plasmids of ST131 strains. These plasmids appear to have evolved to acquire similar gene clusters on multiple occasions, suggesting possible plasmid-mediated convergent evolution leading to evolutionary success. These plasmids also appear to be better suited to exist in specific strains of ST131 due to coadaptive mutations. Overall, a series of events has enabled the evolution of ST131 plasmids, possibly contributing to the lineage’s success
