    High fidelity simian immunodeficiency virus reverse transcriptase mutants have impaired replication in vitro and in vivo

    The low fidelity of HIV replication facilitates immune and drug escape. Some reverse transcriptase (RT) inhibitor drug-resistance mutations increase RT fidelity in biochemical assays but their effect during viral replication is unclear. We investigated the effect of RT mutations K65R, Q151N and V148I on SIV replication and fidelity in vitro, along with SIV replication in pigtailed macaques. SIVmac239-K65R and SIVmac239-V148I viruses had reduced replication capacity compared to wild-type SIVmac239. Direct virus competition assays demonstrated a rank order of wild-type>K65R>V148I mutants in terms of viral fitness. In single round in vitro-replication assays, SIVmac239-K65R demonstrated significantly higher fidelity than wild-type, and rapidly reverted to wild-type following infection of macaques. In contrast, SIVmac239-Q151N was replication incompetent in vitro and in pigtailed macaques. Thus, we showed that RT mutants, and specifically the common K65R drug-resistance mutation, had impaired replication capacity and higher fidelity. These results have implications for the pathogenesis of drug-resistant HIV

    Virologically Suppressed HIV Patients Show Activation of NK Cells and Persistent Innate Immune Activation

    FcRγ is an ITAM-containing adaptor required for CD16 signaling and function in NK cells. We have previously shown that NK cells from HIV patients receiving combination antiretroviral therapy (cART) have decreased FcRγ expression, but the factors causing this are unknown. We conducted a cross-sectional study of cART-naive viremic patients (ART(-)), virologically suppressed patients receiving cART (ART(+)), and HIV-uninfected controls. CD8(+) T cells were activated, as assessed by CD38(+)HLA-DR(+) expression, in ART(-) patients (p < 0.0001), which was significantly reduced in ART(+) patients (p = 0.0005). In contrast, CD38(+)HLA-DR(+) NK cells were elevated in ART(-) patients (p = 0.0001) but did not decrease in ART(+) patients (p = 0.88). NK cells from both ART(-) and ART(+) patients showed high levels of spontaneous degranulation in ex vivo whole blood assays as well as decreased CD16 expression (p = 0.0001 and p = 0.0025, respectively), FcRγ mRNA (p < 0.0001 for both groups), FcRγ protein expression (p = 0.0016 and p < 0.0001, respectively), and CD16-dependent Syk phosphorylation (p = 0.0001 and p = 0.003, respectively). HIV-infected subjects showed alterations in NK activation, degranulation, CD16 expression and signaling, and elevated plasma markers of inflammation and macrophage activation, that is, neopterin and sCD14, which remained elevated in ART(+) patients. Alterations in NK cell measures did not correlate with viral load or CD4 counts. These data show that in HIV patients who achieve viral suppression following cART, NK cell activation persists. This suggests that NK cells respond to factors different from those driving T cell activation, but which are associated with inflammation in HIV patients

The assay uses shrimp nuclease (SNuc) to eliminate DNA contamination, giving high sensitivity and low variability.     The BCB project has provided a model of an open, modular transport concept working on the basis of information and logistics, for the operation of private-sector vessels. The test phase has provided presentation of the economical, ecological and safety aspects of inland navigation, demonstrating an economically viable model of the operating process in the trials phase. This addressed the major goals of utilising cost-savings potentials by improved information processing customer service enhancement and increased flexibility, for better customer loyalty. In order to achieve more acceptance for BCB, it would be necessary to conduct long-term fleet trials. Shore-based information and traffic guidance systems will become extremely important in future for Europe-wide transport of dangerous goods, in view of their political and social relevance. It is important to follow up developments here. The Integrated Navigation system for Inland Waterways was aimed at simplifying the man-machine interface and the sensors, so as to reduce the components of an Inland Vessel Control Center (BCC=Binnenschiff Control Center). This BCC would have to be developed and tested.
