    Changes in Body Condition of Hibernating Bats Support the Thrifty Female Hypothesis and Predict Consequences for Populations with White-Nose Syndrome

    White-nose syndrome (WNS) is a new disease of bats that has devastated populations in eastern North America. Infection with the fungus, Geomyces destructans, is thought to increase the time bats spend out of torpor during hibernation, leading to starvation. Little is known about hibernation in healthy, free-ranging bats and more data are needed to help predict consequences of WNS. Trade-offs presumably exist between the energetic benefits and physiological/ecological costs of torpor, leading to the prediction that the relative importance of spring energy reserves should affect an individual's use of torpor and depletion of energy reserves during winter. Myotis lucifugus mate during fall and winter but females do not become pregnant until after spring emergence. Thus, female reproductive success depends on spring fat reserves while male reproductive success does not. Consequently, females should be “thrifty” in their use of fat compared to males. We measured body condition index (BCI; mass/forearm length) of 432 M. lucifugus in Manitoba, Canada during the winter of 2009/2010. Bats were captured during the fall mating period (n = 200), early hibernation (n = 125), and late hibernation (n = 128). Adult females entered hibernation with greater fat reserves and consumed those reserves more slowly than adult males and young of the year. Consequently, adult females may be more likely than males or young of the year to survive the disruption of energy balance associated with WNS, although surviving females may not have sufficient reserves to support reproduction

    Efficient Environment Guided Approach for Exploration of Complex Environments

    Remote inspection of a complex environment is a difficult, time consuming task for human operators to perform. The need to manually avoid obstacles whilst considering other performance factors i.e. time taken, joint effort and information gained represents significant challenges to continuous operation. This paper proposes an autonomous robotic solution for exploration of an unknown, complex environment using a high DoF robot arm with an eye in hand depth sensor. The main contribution of this work is a new strategy to find the next best view by evaluating frontier regions of the map to maximise coverage, in contrast to many current approaches which densely sample joint or workspace configurations of the robot. Multiple utility functions were evaluated that showed different behaviours. Our results indicated that the presented algorithm can explore an arbitrary environment efficiently while optimising various performance criteria based on the utility function chosen, application constraints and the desires of the user

    Design and development of a low temperature, inductance based high frequency ac susceptometer

    We report on the development of an induction based low temperature high frequency ac susceptometer capable of measuring at frequencies up to 3.5 MHz and at temperatures between 2 K and 300 K. Careful balancing of the detection coils and calibration have allowed a sample magnetic moment resolution of 5×1010Am25\times10^{-10} Am^2 at 1 MHz. We will discuss the design and characterization of the susceptometer, and explain the calibration process. We also include some example measurements on the spin ice material CdEr2_2S4_4 and iron oxide based nanoparticles to illustrate functionality

    Strict inequalities of critical values in continuum percolation

    We consider the supercritical finite-range random connection model where the points x,yx,y of a homogeneous planar Poisson process are connected with probability f(yx)f(|y-x|) for a given ff. Performing percolation on the resulting graph, we show that the critical probabilities for site and bond percolation satisfy the strict inequality pcsite>pcbondp_c^{\rm site} > p_c^{\rm bond}. We also show that reducing the connection function ff strictly increases the critical Poisson intensity. Finally, we deduce that performing a spreading transformation on ff (thereby allowing connections over greater distances but with lower probabilities, leaving average degrees unchanged) {\em strictly} reduces the critical Poisson intensity. This is of practical relevance, indicating that in many real networks it is in principle possible to exploit the presence of spread-out, long range connections, to achieve connectivity at a strictly lower density value.Comment: 38 pages, 8 figure

    Hibernation energetics of free-ranging little brown bats

    Hibernation physiology and energy expenditure have been relatively well studied in large captive hibernators, especially rodents, but data from smaller, free-ranging hibernators are sparse. We examined variation in the hibernation patterns of free-ranging little brown bats (Myotis lucifugus) using temperature-sensitive radio-transmitters. First, we aimed to test the hypothesis that age, sex and body condition affect expression of torpor and energy expenditure during hibernation. Second, we examined skin temperature to assess whether qualitative differences in the thermal properties of the hibernacula of bats, compared with the burrows of hibernating rodents, might lead to different patterns of torpor and arousal for bats. We also evaluated the impact of carrying transmitters on body condition to help determine the potential impact of telemetry studies. We observed large variation in the duration of torpor bouts within and between individuals but detected no effect of age, sex or body condition on torpor expression or estimates of energy expenditure. We observed the use of shallow torpor in the midst of periodic arousals, which may represent a unique adaptation of bats for conservation of energy during the most costly phase of hibernation. There was no difference in the body condition of hibernating bats outfitted with transmitters compared with that of control bats captured from the same hibernaculum at the same time.     Chromosome alterations and E-cadherin gene mutations in human lobular breast cancer

    We have studied a set of 40 human lobular breast cancers for loss of heterozygosity (LOH) at various chromosome locations and for mutations in the coding region plus flanking intron sequences of the E-cadherin gene. We found a high frequency of LOH (100%, 31/31) at 16q21–q22.1. A significantly higher level of LOH was detected in ductal breast tumours at chromosome arms 1p, 3p, 9p, 11q, 13q and 18q compared to lobular breast tumours. Furthermore, we found a significant association between LOH at 16 q containing the E-cadherin locus and lobular histological type. Six different somatic mutations were detected in the E-cadherin gene, of which three were insertions, two deletions and one splice site mutation. Mutations were found in combination with LOH of the wild type E-cadherin locus and loss of or reduced E-cadherin expression detected by immunohistochemistry. The mutations described here have not previously been reported. We compared LOH at different chromosome regions with E-cadherin gene mutations and found a significant association between LOH at 13 q and E-cadherin gene mutations. A significant association was also detected between LOH at 13q and LOH at 7q and 11q. Moreover, we found a significant association between LOH at 3 p and high S phase, LOH at 9p and low ER and PgR content, LOH at 17p and aneuploidy. We conclude that LOH at 16q is the most frequent chromosome alteration and E-cadherin is a typical tumour suppressor gene in lobular breast cancer. © 1999 Cancer Research Campaig

    Evaluation of a Commercial Enzyme Linked Immunosorbent Assay (ELISA) for the Determination of the Neurotoxin BMAA in Surface Waters

    The neurotoxin ß-N-methylamino-L-alanine (BMAA) is suspected to play a role in Alzheimer’s disease, Parkinson’s disease and amyotrophic lateral sclerosis. Because BMAA seems to be produced by cyanobacteria, surface waters are screened for BMAA. However, reliable analysis of BMAA requires specialized and expensive equipment. In 2012, a commercial enzyme-linked immunosorbent assay (ELISA) for determination of BMAA in surface waters was released. This kit could enable fast and relatively cheap screening of surface waters for BMAA. The objective of this study was to determine whether the BMAA ELISA kit was suitable for the determination of BMAA concentrations in surface waters. We hypothesised that the recovery of spiked samples was close to 100% and that the results of unspiked sample analysis were comparable between ELISA and liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis. However, we found that recovery was higher than 100% in most spiked samples, highest determined recovery was over 400%. Furthermore, the ELISA gave a positive signal for nearly each tested sample while no BMAA could be detected by LC-MS/MS. We therefore conclude that in its current state, the kit is not suitable for screening surface waters for BMAA
