    Effect of Price and Quality of Service Customer Loyalty (Study on Railway Passenger Blora Jaya PT. Kereta Api Indonesia (Persero) Daop IV Semarang)

    Development and transportation services business competition is increasing rapidly. They compete in providing the best service to retain customers. This research was conducted at PT.Kereta Api Indonesia (Persero) Regional Operations IV Semarang object KA (train) Blora Jaya. Of user data KA Blora Jaya known of the fluctuations in the number of service users KA Blora Jaya and their number of user complaints Blora Jaya train services relating to facilities, infrastructure, and operational support services. As a company engaged in the field of services, PT.KAI (Persero) DAOP IV Semarang must provide good service to customers, good service offset by price according to what is given by PT. KAI (Persero) DAOP IV Semarang.This study aims to determine the effect of price (X1) and quality of service (X2) on customer loyalty (Y) railway services Blora Jaya. This type of research uses explanatory research with data collection method using interviews and data collection tools used questionnaires to a sample of 100 respondents KA customers Blora Jaya using purposive sampling method. Using analytical techniques validity, reliability, simple linear regression, multiple linear regression, t test and F test by means of SPSS 16.The results showed that the quality of service has a strong influence on customer loyalty by 49.4%, while the price is quite a strong effect on customer loyalty by 21,7%. This indicates that the variable quality of service to customer loyalty variable effect is higher than the variable price.Advice can be given to increase customer loyalty is to improve the quality of railway services Blora Jaya which is considered important by the service user, but still less attention from the company such as the availability of bins, seating capacity, the slow ticket officer in providing service that is the least number of counters that opened and have not been applied to tickets purchased online services

Credits provided by customers will help the bank in maintaining liquidity and the impact on interest income to be received so that it will affect the financial soundness of the bank.     The aim of this study was to determine the effect of leadership style variable and employee environment on the performance of the chocolate production of PT. Pusan Manis Mulia Tangerang. The population of this study were 400 employees of chocolate production division of PT. Pusan Manis Mulia Tangerang, slovin samples formula was used resulted 80 people for sample and the spreading using the method of Nonprobability Sampling (Purposive Sampling). Likert scale was used as measurement scale. Simple linear regression test and regression test with SPSS 15.0 program were used in the data analysis. From the results of data analysis known that in the category of Leadership Style 62.5 percent of the respondence said that it was good. Employee Environment categories 72.5 percent of the respondence said it was good. Category of employee performance most of of the respondence about 68.75 percent said it was good. Leadership Style variables affect the performance of employees by 73.2 percent. Employee Environment variable effect on employee performance by 75.3 percent. Taken together Leadership Style and Employee Environment of Employee Performance by 75.9 percent. This means the better the Leadership Style and Employee Environment is expected that the better the performance. Based on these results the management of PT. Pusan Manis Mulia Tangerang needs to consider the application of Leadership Style by head division of chocolate production.     Identification of S100A8-correlated genes for prediction of disease progression in non-muscle invasive bladder cancer

    <p>Abstract</p> <p>Background</p> <p><it>S100 calcium binding protein A8 </it>(<it>S100A8</it>) has been implicated as a prognostic indicator in several types of cancer. However, previous studies are limited in their ability to predict the clinical behavior of the cancer. Here, we sought to identify a molecular signature based on <it>S100A8 </it>expression and to assess its usefulness as a prognostic indicator of disease progression in non-muscle invasive bladder cancer (NMIBC).</p> <p>Methods</p> <p>We used 103 primary NMIBC specimens for microarray gene expression profiling. The median follow-up period for all patients was 57.6 months (range: 3.2 to 137.0 months). Various statistical methods, including the leave-one-out cross validation method, were applied to identify a gene expression signature able to predict the likelihood of progression. The prognostic value of the gene expression signature was validated in an independent cohort (n = 302).</p> <p>Results</p> <p>Kaplan-Meier estimates revealed significant differences in disease progression associated with the expression signature of <it>S100A8</it>-correlated genes (log-rank test, <it>P </it>< 0.001). Multivariate Cox regression analysis revealed that the expression signature of <it>S100A8</it>-correlated genes was a strong predictor of disease progression (hazard ratio = 15.225, 95% confidence interval = 1.746 to 133.52, <it>P </it>= 0.014). We validated our results in an independent cohort and confirmed that this signature produced consistent prediction patterns. Finally, gene network analyses of the signature revealed that <it>S100A8</it>, <it>IL1B</it>, and <it>S100A9 </it>could be important mediators of the progression of NMIBC.</p> <p>Conclusions</p> <p>The prognostic molecular signature defined by <it>S100A8</it>-correlated genes represents a promising diagnostic tool for the identification of NMIBC patients that have a high risk of progression to muscle invasive bladder cancer.</p

    Gene expression profiles of prostate cancer reveal involvement of multiple molecular pathways in the metastatic process

    BACKGROUND: Prostate cancer is characterized by heterogeneity in the clinical course that often does not correlate with morphologic features of the tumor. Metastasis reflects the most adverse outcome of prostate cancer, and to date there are no reliable morphologic features or serum biomarkers that can reliably predict which patients are at higher risk of developing metastatic disease. Understanding the differences in the biology of metastatic and organ confined primary tumors is essential for developing new prognostic markers and therapeutic targets. METHODS: Using Affymetrix oligonucleotide arrays, we analyzed gene expression profiles of 24 androgen-ablation resistant metastatic samples obtained from 4 patients and a previously published dataset of 64 primary prostate tumor samples. Differential gene expression was analyzed after removing potentially uninformative stromal genes, addressing the differences in cellular content between primary and metastatic tumors. RESULTS: The metastatic samples are highly heterogenous in expression; however, differential expression analysis shows that 415 genes are upregulated and 364 genes are downregulated at least 2 fold in every patient with metastasis. The expression profile of metastatic samples reveals changes in expression of a unique set of genes representing both the androgen ablation related pathways and other metastasis related gene networks such as cell adhesion, bone remodelling and cell cycle. The differentially expressed genes include metabolic enzymes, transcription factors such as Forkhead Box M1 (FoxM1) and cell adhesion molecules such as Osteopontin (SPP1). CONCLUSION: We hypothesize that these genes have a role in the biology of metastatic disease and that they represent potential therapeutic targets for prostate cancer

    S100A14 Stimulates Cell Proliferation and Induces Cell Apoptosis at Different Concentrations via Receptor for Advanced Glycation End Products (RAGE)

    S100A14 is an EF-hand containing calcium-binding protein of the S100 protein family that exerts its biological effects on different types of cells. However, exact extracellular roles of S100A14 have not been clarified yet. Here we investigated the effects of S100A14 on esophageal squamous cell carcinoma (ESCC) cell lines. Results demonstrated that low doses of extracellular S100A14 stimulate cell proliferation and promote survival in KYSE180 cells through activating ERK1/2 MAPK and NF-ΞΊB signaling pathways. Immunoprecipitation assay showed that S100A14 binds to receptor for advanced glycation end products (RAGE) in KYSE180 cells. Inhibition of RAGE signaling by different approaches including siRNA for RAGE, overexpression of a dominant-negative RAGE construct or a RAGE antagonist peptide (AmphP) significantly blocked S100A14-induced effects, suggesting that S100A14 acts via RAGE ligation. Furthermore, mutation of the N-EF hand of S100A14 (E39A, E45A) virtually reduced 10 Β΅g/ml S100A14-induced cell proliferation and ERK1/2 activation. However, high dose (80 Β΅g/ml) of S100A14 causes apoptosis via the mitochondrial pathway with activation of caspase-3, caspase-9, and poly(ADP-ribose) polymerase. High dose S100A14 induces cell apoptosis is partially in a RAGE-dependent manner. This is the first study to demonstrate that S100A14 binds to RAGE and stimulates RAGE-dependent signaling cascades, promoting cell proliferation or triggering cell apoptosis at different doses
