    Occurrence of the Old World bug Megacopta cribraria (Fabricius) (Heteroptera: Plataspidae) in Georgia: a serious home invader and potential legume pest

    Specimens of Megacopta cribraria (Fabricius) were collected in northern Georgia in late October 2009, where they were invading homes in large numbers. This is the first known occurrence of this species and the family Plataspidae in the New World. Megacopta cribraria was previously known from Asia and Australia. A key is provided to separate Plataspidae from other families of Pentatomoidea in America North of Mexico. A diagnosis and figures are provided to facilitate recognition of M. cribraria. Reported host plants and other aspects of the biology of this species are reviewed. Megacopta cribraria is considered a pest of numerous legumes in Asia, has the potential to provide biological control of kudzu, Pueraria montana var. lobata (Willd.) Ohwi, (Fabaceae) and likely will continue to be a household pest in the vicinity of kudzu fields as well as become a pest of North American legume crops

    A national registry for juvenile dermatomyositis and other paediatric idiopathic inflammatory myopathies: 10 years' experience; the Juvenile Dermatomyositis National (UK and Ireland) Cohort Biomarker Study and Repository for Idiopathic Inflammatory Myopathies

    Objectives: The paediatric idiopathic inflammatory myopathies (IIMs) are a group of rare chronic inflammatory disorders of childhood, affecting muscle, skin and other organs. There is a severe lack of evidence base for current treatment protocols in juvenile myositis. The rarity of these conditions means that multicentre collaboration is vital to facilitate studies of pathogenesis, treatment and disease outcomes. We have established a national registry and repository for childhood IIM, which aims to improve knowledge, facilitate research and clinical trials, and ultimately to improve outcomes for these patients. Methods: A UK-wide network of centres and research group was established to contribute to the study. Standardized patient assessment, data collection forms and sample protocols were agreed. The Biobank includes collection of peripheral blood mononuclear cells, serum, genomic DNA and biopsy material. An independent steering committee was established to oversee the use of data/samples. Centre training was provided for patient assessment, data collection and entry. Results: Ten years after inception, the study has recruited 285 children, of which 258 have JDM or juvenile PM; 86% of the cases have contributed the biological samples. Serial sampling linked directly to the clinical database makes this a highly valuable resource. The study has been a platform for 20 sub-studies and attracted considerable funding support. Assessment of children with myositis in contributing centres has changed through participation in this study. Conclusions: This establishment of a multicentre registry and Biobank has facilitated research and contributed to progress in the management of a complex group of rare muscloskeletal conditions

    Status and Relative Abundance of Alabama Shad, \u3ci\u3eAlosa alabamae\u3c/i\u3e, in Alabama

    Alabama Shad, Alosa alabamae, an anadromous fish found historically from the Mississippi River basin eastward to the Suwanee River, has experienced population declines and even extirpation in some States. In Alabama, A. alabamae have been found in rivers of the Mobile River basin and Conecuh, Yellow, and Choctawhatchee rivers in the coastal Gulf Plain. We report on our directed and targeted efforts to assess the current status and relative abundance of A. alabamae in Alabama and compare our results to past A. alabamae surveys in Alabama. We completed 52 sampling trips and expended 129.5 hours of boat-electrofishing effort targeting A. alabamae. Sampling was conducted during the spring to coincide with the spring-spawning migration at historical sites and sites conducive for the collection of A. alabamae. No A. alabamae was collected from the Mobile River basin (i.e., Alabama and Tombigbee rivers) and only one A. alabamae was collected from the Conecuh River. We collected seven A. alabamae in 2011 and three in 2018 from the Choctawhatchee River. For the Choctawhatchee River population, our results indicated a precipitous decline in abundance by 71% and 98% from 1999/2000 to 2011 and 2018, respectively. In addition, our results support the extirpation of A. alabamae from the Mobile River basin and a severely depressed population in the Conecuh River. Although A. alabamae was recently denied listing under the Endangered Species Act by the National Marine Fisheries Service due to lack of apparent range-wide extinction, our results indicate what was once considered the second largest population of A. alabamae from the Choctawhatchee River is on the verge of extirpation. Alosa alabamae could become extirpated from Alabama in the near future, which is a significant portion of its range

    Calcifediol is a safe and effective metabolite for raising vitamin D status and improving growth and feed conversion in rainbow trout

    The vitamin D endocrine system is required for the transcriptional regulation of a myriad of vertebrate genes including those involved in bone health, growth, nutrient metabolism and immunity. The requirements of salmonids for vitamin D are amongst the highest for any aquaculture species. With nuances, the metabolism of the pre-vitamin cholecalciferol (D3) via calcifediol (25-OH-D3), required to produce the physiologically active hormone calcitriol (1,25-OH-D3) is conserved in fish. The composition of modern aquafeeds, growth in seawater and production challenges, such as disease, may result in the suboptimal biochemical activation of vitamin D hormone in fish. A 91-day experimental feeding trial was used to test the safety and efficacy of calcifediol for the supply of vitamin D to rainbow trout with an initial body weight of 57.6 g. A practical diet containing cholecalciferol within recommended levels (5240 IU) was supplemented with either 69.8, 687 or 6854 ÎĽg/kg calcifediol. The efficacy of calcifediol was determined by the assessment of zootechnical performance and the appearance of vitamin D metabolites in the blood. The safety of the dietary interventions was assessed from generic health indices, examination of gross pathologies, hematology, and blood chemistry. Test fish increased body weight at least 5.6-fold to 323.5 g over the experimental feeding period. The supplementation of 687 or 6854 ÎĽg/kg calcifediol resulted in significant improvements in growth rate and feed conversion (FCR). Whilst not detectable in control fish, calcifediol increased linearly according to dietary levels in the blood and to a lesser extent in the white muscle. The increases of calcifediol in the blood were accompanied by saturable increases of circulating active vitamin D. At the end of the 91-day feeding period, survival was 100%, no gross pathologies relating to the diets were observed, and health indices, hematology, and blood chemistry, including calcium and phosphorus, were not significantly altered. The supplementation of calcifediol to practical diets containing recommended levels of cholecalciferol improves zootechnical performance and ensures that maximal levels of active vitamin D are present in the blood to meet physiological demands. With a lack of significant effects on health indices, hematology, and blood chemistry, including calcium and phosphorus, the tested high doses of calcifediol are concluded to be safe for salmonids

    Alterations in vascular function in primary aldosteronism - a cardiovascular magnetic resonance imaging study

    Introduction: Excess aldosterone is associated with increased cardiovascular risk. Aldosterone has a permissive effect on vascular fibrosis. Cardiovascular magnetic resonance imaging (CMR) allows study of vascular function by measuring aortic distensibility. We compared aortic distensibility in primary aldosteronism (PA), essential hypertension (EH) and normal controls and explored the relationship between aortic distensibility and pulse wave velocity (PWV).<p></p> Methods: We studied PA (n=14) and EH (n=33) subjects and age-matched healthy controls (n=17) with CMR, including measurement of aortic distensibility, and measured PWV using applanation tonometry. At recruitment, PA and EH patients had similar blood pressure and left ventricular mass.<p></p> Results: Subjects with PA had significantly lower aortic distensibilty and higher PWV compared to EH and healthy controls. These changes were independent of other factors associated with reduced aortic distensibility, including aging. There was a significant relationship between increasing aortic stiffness and age in keeping with physical and vascular aging. As expected, aortic distensibility and PWV were closely correlated.<p></p> Conclusion: These results demonstrate that PA patients display increased arterial stiffness compared to EH, independent of vascular aging. The implication is that aldosterone invokes functional impairment of arterial function. The long-term implications of arterial stiffening in aldosterone excess require further study.<p></p&gt
