    Studies on the Biosynthesis of Some Furocoumarins Present in Ruta graveolens

    The biosynthesis of some furocoumarins present in Ruta graveolens has been studied administering labeled psoralen, xanthotoxin, rutaretin, marmesin and 7-hydroxycoumarin-derivatives. The results obtained indicate that the biosynthetic pathway involves not only psoralen, but also xanthotoxin and bergapten, by means of methoxylation or demethoxylation reactions. Moreover results achieved from administering rutaretin demonstrate that this substance is a new very effective natural precursor for xanthotoxin; in a parallel way marmesin is incorporated into psoralen. The reported results indicate that also at the stage of natural 4′,5′-dihydrofurocoumarins intermediates an interconversion can occur. Finally the essential role of umbelliferone in the biosynthesis of furocoumarins has been confirmed

    The past to unravel the future: Deoxygenation events in the geological archive and the anthropocene oxygen crisis

    Despite the observation that we are witnessing a true oxygen crisis, the ocean deoxygenation theme is getting less attention from the media and population compared to other environmental stressors concerning climate change. The current ocean oxygen crisis is characterized by a complex interplay of climatic, biological, and oceanographic processes acting at different time scales. Earth system models offer insights into future deoxygenation events and their potential extent; however, their capacity to precisely constrain these events is complicated by the intricate interplay of various interconnected feedback mechanisms. The Earth's geological history has been punctuated by regional and global deoxygenation events, which are usually expressed by organic-rich sediment in the geological record and can be useful past analogues of the present-day and future oxygenation crisis related to current climatic stress. Accordingly, we provide an overview of the key elements characterizing past deoxygenation events, aiming for a better understanding of the Anthropocene oxygen crisis and its potential evolution. We suggest that past global deoxygenation events during hypethermals may bear similarities to present-day dynamics in the open ocean. Additionally, we explore the significance of regional deoxygenation events with cyclical occurrences for better constraining environmental dynamics and ecological impacts in semi-enclosed, restricted, and marginal basins. Despite the unprecedented magnitude and rate of current anthropogenic pressures, it is essential to consider the comparison of triggers and feedbacks from ancient deoxygenation events when investigating the future of this concealed but ecologically impactful problem

    Conservative treatment for hypervascularised placental polyp with secondary haemoperitoneum: A case report

    Objective: We describe the first case to our knowledge of hypervascularised placental polyp (HPP) presenting with acute pelvic pain and hemoperitoneum. Case Report: A 33 years-old woman with a history of medical abortion three months earlier came to our attention complaining acute pelvic pain and vaginal bleeding. Transvaginal (TV) and transabdominal (TA) ultrasound (US) demonstrated a highly vascular intrauterine lesion and intra-abdominal free fluid consistent with a diagnosis of haemoperitoneum. Emergency laparoscopy yielded no intra-abdominal bleeding and was followed by bilateral selective embolization of the uterine arteries due to persistent vaginal bleeding. Hysteroscopy and pathology findings were consistent with a final diagnosis of HPP. Conclusion: HPP may occur months or years after pregnancy or abortion and the clinical picture of abnormal vaginal bleeding associated with acute abdominal pain and haemoperitoneum should warrant to consider HPP among the differential diagnosis. Clinical and imaging findings need to be considered when planning the conservative management of HPP. Our experience suggests that uterine artery embolization is a safe and effective for the conservative treatment of highly vascularized HPP. (www. actabiomedica.it)

    Observational indications of magneto-optical effects in the scattering polarization wings of the Ca I 4227 \AA\ line

    Several strong resonance lines, such as H I Ly-α\alpha, Mg II k, Ca II K, Ca I 4227 \AA\, which are characterized by deep and broad absorption profiles in the solar intensity spectrum, show conspicuous linear scattering polarization signals when observed in quiet regions close to the solar limb. Such signals show a characteristic triplet-peak structure, with a sharp peak in the line core and extended wing lobes. The line core peak is sensitive to the presence of magnetic fields through the Hanle effect, which however is known not to operate in the line wings. Recent theoretical studies indicate that, contrary to what was previously believed, the wing linear polarization signals are also sensitive to the magnetic field through magneto-optical effects (MO). We search for observational indications of this recently discovered physical mechanism in the scattering polarization wings of the Ca I 4227 \AA\ line. We performed a series of spectropolarimetric observations of this line using the Zurich IMaging POLarimeter (ZIMPOL) camera at the Gregory-Coud\'e telescope of IRSOL (Switzerland) and at the GREGOR telescope in Tenerife (Spain). Spatial variations of the total linear polarization degree and of the linear polarization angle are clearly appreciable in the wings of the observed line. We provide a detailed discussion of our observational results, showing that the detected variations always take place in regions where longitudinal magnetic fields are present, thus supporting the theoretical prediction that they are produced by MO effects.Comment: Accepted by A&

    Radical-free hyperpolarized MRI using endogenously occurring pyruvate analogues and UV-induced nonpersistent radicals.

    It was recently demonstrated that nonpersistent radicals can be generated in frozen solutions of metabolites such as pyruvate by irradiation with UV light, enabling radical-free dissolution dynamic nuclear polarization. Although pyruvate is endogenous, the presence of pyruvate may interfere with metabolic processes or the detection of pyruvate as a metabolic product, making it potentially unsuitable as a polarizing agent. Therefore, the aim of the current study was to characterize solutions containing endogenously occurring alternatives to pyruvate as UV-induced nonpersistent radical precursors for in vivo hyperpolarized MRI. The metabolites alpha-ketovalerate (αkV) and alpha-ketobutyrate (αkB) are analogues of pyruvate and were chosen as potential radical precursors. Sample formulations containing αkV and αkB were studied with UV-visible spectroscopy, irradiated with UV light, and their nonpersistent radical yields were quantified with electron spin resonance and compared with pyruvate. The addition of <sup>13</sup> C-labeled substrates to the sample matrix altered the radical yield of the precursors. Using αkB increased the <sup>13</sup> C-labeled glucose liquid-state polarization to 16.3% ± 1.3% compared with 13.3% ± 1.5% obtained with pyruvate, and 8.9% ± 2.1% with αkV. For [1- <sup>13</sup> C]butyric acid, polarization levels of 12.1% ± 1.1% for αkV, 12.9% ± 1.7% for αkB, 1.5% ± 0.2% for OX063 and 18.7% ± 0.7% for Finland trityl, were achieved. Hyperpolarized [1- <sup>13</sup> C]butyrate metabolism in the heart revealed label incorporation into [1- <sup>13</sup> C]acetylcarnitine, [1- <sup>13</sup> C]acetoacetate, [1- <sup>13</sup> C]butyrylcarnitine, [5- <sup>13</sup> C]glutamate and [5- <sup>13</sup> C]citrate. This study demonstrates the potential of αkV and αkB as endogenous polarizing agents for in vivo radical-free hyperpolarized MRI. UV-induced, nonpersistent radicals generated in endogenous metabolites enable high polarization without requiring radical filtration, thus simplifying the quality-control tests in clinical applications

    Advances in the Pathophysiology of Thrombosis in Antiphospholipid Syndrome. Molecular Mechanisms and Signaling through Lipid Rafts

    The pathological features of antiphospholipid syndrome (APS) are related to the activity of circulating antiphospholipid antibodies (aPLs) associated with vascular thrombosis and obstetric complications. Indeed, aPLs are not only disease markers, but also play a determining pathogenetic role in APS and exert their effects through the activation of cells and coagulation factors and inflammatory mediators for the materialization of the thromboinflammatory pathogenetic mechanism. Cellular activation in APS necessarily involves the interaction of aPLs with target receptors on the cell membrane, capable of triggering the signal transduction pathway(s). This interaction occurs at specific microdomains of the cell plasma membrane called lipid rafts. In this review, we focus on the key role of lipid rafts as signaling platforms in the pathogenesis of APS, and propose this pathogenetic step as a strategic target of new therapies in order to improve classical anti-thrombotic approaches with "new " immunomodulatory drugs
