    Twist1 enhances hypoxia induced radioresistance in cervical cancer cells by promoting nuclear EGFR localization

    Twist1 is a crucial transcription factor that regulates epithelial mesenchymal transition and involves in metastasis. Recent evidence suggests that Twist1 plays important role in hypoxia-induced radioresistance, but the underlying mechanism remains elusive. Here we investigated the change of Twist1 expression in human cervical squamous cancer cell line SiHa after hypoxia treatment. We also explored the role of Twist1 in radioresistance by manipulating the expression level of Twist1.We observed that hypoxia treatment elevated the expression of Twist1 in SiHa cells. Knockdown of Twist1 with siRNA increased the radiosensitivity of SiHa cells under hypoxia condition, accompanied by reduced levels of nuclear Epidermal Growth Factor Receptor (EGFR) and DNA-dependent protein kinase (DNA-PK). Conversely, overexpression of Twist1 led to increased radioresistance of SiHa cells, which in turn increased nuclear EGFR localisation and expression levels of nuclear DNA-PK. Moreover, concomitant high expression of hypoxia-inducible factor-1? (HIF-1?) and Twist1 in primary tumors of cervical cancer patients correlated with the worse prognosis after irradiation treatment. Taken together, these data provide new insights into molecular mechanism underlying hypoxia-induced radio resistance in cervical cancer cells, and suggest that Twist1 is a promising molecular target to improve the efficacy of cancer radiotherapy

    Picroside I inhibits asthma phenotypes by regulating Tbet/ GATA-3 ratio and Th1/Th2 balance in a murine model of asthma

    Purpose: To examine the anti-asthmatic activity of picroside I in murine asthma model, and to elucidate the mechanism(s) involved.Methods: The study involved systematic sensitization of acclimatized BALB/c mice with ovalbumin (OVA), and subsequent exposure to aerosol allergens. The effect of picroside I on associated IgE formation was determined. All assays were performed using standard protocols. Protein expression was assessed using western blotting.Results: Picroside I inhibited allergic airway inflammation, AHR, and the production of OVA-associated IgE and Th2 cytokines. Moreover, it altered the T-bet/GATA3 ratio by suppressing the phosphorylation of STAT6 in a dose-dependent manner.Conclusion: These results indicate that the anti-asthmatic effect of picroside I occurs via a mechanism involving inhibition of Th2 cytokines by suppression of the expressions of pSTAT6 and GATA-3, and upregulation of the expression of T-bet.     High expression of transcriptional coactivator p300 correlates with aggressive features and poor prognosis of hepatocellular carcinoma

    <p>Abstract</p> <p>Background</p> <p>It has been suggested that p300 participates in the regulation of a wide range of cell biological processes and mutation of p300 has been identified in certain types of human cancers. However, the expression dynamics of p300 in hepatocellular carcinoma (HCC) and its clinical/prognostic significance are unclear.</p> <p>Methods</p> <p>In this study, the methods of reverse transcription-polymerase chain reaction (RT-PCR), Western blotting and immunohistochemistry (IHC) were utilized to investigate protein/mRNA expression of p300 in HCCs. Receiver operating characteristic (ROC) curve analysis, spearman's rank correlation, Kaplan-Meier plots and Cox proportional hazards regression model were used to analyze the data.</p> <p>Results</p> <p>Up-regulated expression of p300 mRNA and protein was observed in the majority of HCCs by RT-PCR and Western blotting, when compared with their adjacent non-malignant liver tissues. According to the ROC curves, the cutoff score for p300 high expression was defined when more than 60% of the tumor cells were positively stained. High expression of p300 was examined in 60/123 (48.8%) of HCCs and in 8/123 (6.5%) of adjacent non-malignant liver tissues. High expression of p300 was correlated with higher AFP level, larger tumor size, multiplicity, poorer differentiation and later stage (<it>P </it>< 0.05). In univariate survival analysis, a significant association between overexpression of p300 and shortened patients' survival was found (<it>P </it>= 0.001). In different subsets of HCC patients, p300 expression was also a prognostic indicator in patients with stage II (<it>P </it>= 0.007) and stage III (<it>P </it>= 0.011). Importantly, p300 expression was evaluated as an independent prognostic factor in multivariate analysis (<it>P </it>= 0.021). Consequently, a new clinicopathologic prognostic model with three poor prognostic factors (p300 expression, AFP level and vascular invasion) was constructed. The model could significantly stratify risk (low, intermediate and high) for overall survival (<it>P </it>< 0.0001).</p> <p>Conclusions</p> <p>Our findings provide a basis for the concept that high expression of p300 in HCC may be important in the acquisition of an aggressive phenotype, suggesting that p300 overexpression, as examined by IHC, is an independent biomarker for poor prognosis of patients with HCC. The combined clinicopathologic prognostic model may become a useful tool for identifying HCC patients with different clinical outcomes.</p

    Identify submitochondria and subchloroplast locations with pseudo amino acid composition: Approach from the strategy of discrete wavelet transform feature extraction

    AbstractIt is very challenging and complicated to predict protein locations at the sub-subcellular level. The key to enhancing the prediction quality for protein sub-subcellular locations is to grasp the core features of a protein that can discriminate among proteins with different subcompartment locations. In this study, a different formulation of pseudoamino acid composition by the approach of discrete wavelet transform feature extraction was developed to predict submitochondria and subchloroplast locations. As a result of jackknife cross-validation, with our method, it can efficiently distinguish mitochondrial proteins from chloroplast proteins with total accuracy of 98.8% and obtained a promising total accuracy of 93.38% for predicting submitochondria locations. Especially the predictive accuracy for mitochondrial outer membrane and chloroplast thylakoid lumen were 82.93% and 82.22%, respectively, showing an improvement of 4.88% and 27.22% when other existing methods were compared. The results indicated that the proposed method might be employed as a useful assistant technique for identifying sub-subcellular locations.     Response of temperate forest ecosystem services to rainfall: A case study in the forest nature reserves of northern China

    In the context of global climate change, temperate forests in climate-sensitive areas are inevitably affected. To deepen the understanding of the impact on precipitation changes into the relationship between key ecosystem services (ESs), this study selected net primary productivity (NPP), soil conservation (SC) and water yield (WY) of temperate forest in northern China as objects, and the Spearman correlation test and redundancy analysis were applied to analyze the response of ESs relationship to precipitation gradient. The results show that precipitation is the meteorological factor with the greatest impact (contribution 21.2%, p&lt;0.01) on ESs and their relationships in temperate forests. The 600-700 mm precipitation gradient is the key turning point in the change of ESs relationship of WY with NPP and SC. This indicates that attention should be paid to the spatial variation of the 600-700 mm precipitation region in the future warm-wet in northern China, which should be used as a dividing line of forest management and policy development. Based on the results, future restoration projects in northern temperate forest should focus on (1) in areas with less than 600-700 mm of precipitation, attention should be paid to the selection of tree species for afforestation to maintain regional water balance; (2) in areas with more than 700 mm of precipitation, soil and water conservation projects need to be planned, especially in mountainous area. The research can not only support the management of temperate forest ecosystems in northern China, but also provide reference to other forest ecosystems to cope with climate change
