    Jasmonic acid pretreatment improves salt tolerance of wheat by regulating hormones biosynthesis and antioxidant capacity

    Salt stress is a severe environmental factor that detrimentally affects wheat growth and production worldwide. Previous studies illustrate that exogenous jasmonic acid (JA) significantly improved salt tolerance in plants. However, little is known about the underlying molecular mechanisms of JA induced physiochemical changes in wheat seedlings under salt stress conditions. In this study, biophysiochemical and transcriptome analysis was conducted to explore the mechanisms of exogenous JA induced salt tolerance in wheat. Exogenous JA increased salt tolerance of wheat seedlings by alleviating membrane lipid oxidation, improving root morphology, enhancing the contents of ABA, JA and SA and increasing relative water content. In the RNA-seq profiles, we identified a total of 54,263 unigenes and 1,407 unigenes showed differentially expressed patterns in JA pretreated wheat seedlings exposed to salt stress comparing to those with salt stress alone. Subsequently, gene ontology (GO) and KEGG pathway enrichment analysis characterized that DEGs involved in linoleic acid metabolism and plant hormone signal transduction pathways were up-regulated predominantly in JA pretreated wheat seedlings exposed to salt stress. We noticed that genes that involved in antioxidative defense system and that encoding transcription factors were mainly up- or down-regulated. Moreover, SOD, POD, CAT and APX activities were increased in JA pretreated wheat seedlings exposed to salt stress, which is in accordance with the transcript profiles of the relevant genes. Taken together, our results demonstrate that the genes and enzymes involved in physiological and biochemical processes of antioxidant system, plant hormones and transcriptional regulation contributed to JA-mediated enhancement of salt tolerance in wheat. These findings will facilitate the elucidation of the potential molecular mechanisms associated with JA-dependent amelioration of salt stress in wheat and lay theoretical foundations for future studies concerning the improvement of plant tolerance to abiotic environmental stresses

    Distribution, characterization, and induction of CD8+ regulatory T cells and IL-17-producing CD8+ T cells in nasopharyngeal carcinoma

    <p>Abstract</p> <p>Background</p> <p>CD8<sup>+ </sup>effector cells often have an antitumor function in patients with cancer. However, CD8<sup>+</sup>Foxp3<sup>+ </sup>regulatory T cells (Tcregs) and interleukin (IL)-17-producing CD8<sup>+ </sup>T cells (Tc17 cells) also derive from the CD8<sup>+ </sup>T cell lineage. Their role in the antitumor response remains largely unknown. In the present study, we aimed to investigate the distribution, characterization, and generation of CD8<sup>+ </sup>Tcregs and Tc17 cells in NPC patients.</p> <p>Methods</p> <p>Peripheral blood and tumor biopsy tissues from 21 newly diagnosed patients with nasopharyngeal carcinoma (NPC) were collected, along with peripheral blood from 21 healthy donors. The biological characteristics of Tcregs and Tc17 cells from blood and tumor tissues were examined by intracellular staining, tetramer staining and fluorescence-activated cell sorting (FACS) analysis. The suppressive function of Tcregs was investigated using a proliferation assay that involved co-culture of sorted CD8<sup>+</sup>CD25<sup>+ </sup>T cells with naïve CD4<sup>+ </sup>T cells <it>in vitro</it>.</p> <p>Results</p> <p>We observed an increased prevalence of Tcregs and Tc17 cells among tumor-infiltrating lymphocytes (TILs) and different distribution among peripheral blood mononuclear cells (PBMCs) in NPC patients. Cytokine profiles showed that the Tcregs expressed a high level of IL-10 and low level of transforming growth factor β, whereas Tc17 cells expressed a high level of tumor necrosis factor α. Interestingly, both subsets expressed a high level of interferon γ in TILs, and the Tcregs suppressed naïve CD4<sup>+ </sup>T cell proliferation by a cell contact-dependent mechanism <it>in vitro</it>. Moreover, we demonstrated the existence of Epstein-Barr virus latent membrane protein (LMP) 1 and LMP2 antigen-specific Tcregs in NPC.</p> <p>Conclusions</p> <p>Our data provide new insights into the composition and function of CD8<sup>+ </sup>T-cell subsets in NPC, which may have an important influence on NPC immunotherapy.</p

    Properties of a Fading AGN from SDSS-IV MaNGA

    We identify a fading AGN SDSS J220141.64+115124.3 from the internal Product Launch-11 (MPL-11) in Mapping Nearby Galaxies at Apache Point Observatory (MaNGA) survey. The central region with a projected radius of \sim2.4 kpc is characterized as LINER-like line ratios while the outskirts extended to \sim15 kpc show Seyfert-like line ratios. The [OIII]λ\lambda5007 luminosity of the Seyfert regions is a factor of 37 (2) higher than the LINER regions without (with) dust attenuation correction, suggesting that the AGN activity decreases at least \sim8 ×\times 103^3 yrs (\sim2.4 kpc/light-speed) ago. We model the emission line spectra in the central region with double Gaussian components (a narrow core and a broad wing) and analyze the properties of each component. The narrow core component mostly co-rotates with the stellar disc, whereas the broad wing component with a median of the velocity dispersion \sim300 km s1^{-1} is related to a wind outflow. The kinematic position angle (PA) of the ionized gas shows a \sim20{\deg} twist from the galaxy center to 1.5 effective radius. The median of the PA difference between the gas and stellar components is as large as \sim50{\deg} within 0.4 effective radius. The tidal feature in DESI image and star-gas misalignment suggest this galaxy is a merger remnant. Combining all these observational results as well as public available X-ray and MIR luminosities, we confirm this is a fading AGN, the merger process kick-started the central engine to quasar phase which ionized gas composed of tidal debris, and now the activity of the central black hole decreases. The discontinuity in [OIII]λ\lambda5007 flux and EQW maps is due to multiple AGN outbursts triggered by merger remnant gas inflows.Comment: Accepted for publication in MNRAS. 12 pages, 10 figures, 1 tabl

    Cytochrome P450 27C1 Level Dictates Lung Cancer Tumorigenicity and Sensitivity towards Multiple Anticancer Agents and Its Potential Interplay with the IGF-1R/Akt/p53 Signaling Pathway

    Cytochrome P450 enzymes (CYP450s) exert mighty catalytic actions in cellular metabolism and detoxication, which play pivotal roles in cell fate determination. Preliminary data shows differential expression levels of CYP27C1, one of the "orphan P450s" in human lung cancer cell lines. Here, we study the functions of CYP27C1 in lung cancer progression and drug endurance, and explore its potential to be a diagnostic and therapeutic target for lung cancer management. Quantitative real-time PCR and immunoblot assays were conducted to estimate the transcription and protein expression level of CYP27C1 in human lung cancer cell lines, which was relatively higher in A549 and H1975 cells, but was lower in H460 cells. Stable CYP27C1-knockdown A549 and H1975 cell lines were established, in which these cells showed enhancement in cell proliferation, colony formation, and migration. In addition, aberrant IGF-1R/Akt/p53 signal transduction was also detected in stable CYP27C1-knockdown human lung cancer cells, which exhibited greater tolerance towards the treatments of anticancer agents (including vinorelbine, picropodophyllin, pacritinib, and SKLB610). This work, for the first time, reveals that CYP27C1 impacts lung cancer cell development by participating in the regulation of the IGF-1R/Akt/p53 signaling pathway, and the level of CYP27C1 plays indispensable roles in dictating the cellular sensitivity towards multiple anticancer agents

    A simulation study on the measurement of D0-D0bar mixing parameter y at BES-III

    We established a method on measuring the \dzdzb mixing parameter yy for BESIII experiment at the BEPCII e+ee^+e^- collider. In this method, the doubly tagged ψ(3770)D0D0\psi(3770) \to D^0 \overline{D^0} events, with one DD decays to CP-eigenstates and the other DD decays semileptonically, are used to reconstruct the signals. Since this analysis requires good e/πe/\pi separation, a likelihood approach, which combines the dE/dxdE/dx, time of flight and the electromagnetic shower detectors information, is used for particle identification. We estimate the sensitivity of the measurement of yy to be 0.007 based on a 20fb120fb^{-1} fully simulated MC sample.Comment: 6 pages, 7 figure

    Social Support, Resilience, and Mental Health Among Three High-Risk Groups in Hong Kong: A Mediation Analysis

    ObjectivesTo compare the prevalence of anxiety/depression, resilience, and social support among nurses, foreign domestic helpers (FDHs), and residents living in subdivided units (SDUs), and to examine their associations in these high-risk groups in Hong Kong during Omicron waves.MethodsWe recruited 1,014 nurses, 621 FDHs, and 651 SDU residents from December 2021 to May 2022 in this cross-sectional survey. The depression, anxiety, social support, and resilience levels were measured by the validated scales. The multivariate binary logistic regression and causal mediation analysis were applied to examine the associations.ResultsWe observed a prevalence of 17.7% in anxiety and 21.6% in depression which were the highest in SDU residents, followed by FDHs, and lowest in nurses. Social support was associated with increased resilience levels and decreased risks of anxiety/depression. The association of social support with mental disorders was partly mediated by resilience, accounting for 30.9% and 20.9% of the total effect of social support on anxiety and depression, respectively.ConclusionPublic health strategies should target improving social support and providing resilience-promoting interventions to help reduce mental disorders in vulnerable groups

    Reliable Detection of Paternal SNPs within Deletion Breakpoints for Non-Invasive Prenatal Exclusion of Homozygous α0-Thalassemia in Maternal Plasma

    Reliable detection of large deletions from cell-free fetal DNA (cffDNA) in maternal plasma is challenging, especially when both parents have the same deletion owing to a lack of specific markers for fetal genotyping. In order to evaluate the efficacy of a non-invasive prenatal diagnosis (NIPD) test to exclude α-thalassemia major that uses SNPs linked to the normal paternal α-globin allele, we established a novel protocol to reliably detect paternal SNPs within the (−−SEA) breakpoints and performed evaluation of the diagnostic potential of the protocol in a total of 67 pregnancies, in whom plasma samples were collected prior to invasive obstetrics procedures in southern China. A group of nine SNPs identified within the deletion breakpoints were scanned to select the informative SNPs in each of the 67 couples DNA by multiplex PCR based mini-sequencing technique. The paternally inherited SNP allele from cffDNA was detected by allele specific real-time PCR. A protocol for reliable detection of paternal SNPs within the (−−SEA) breakpoints was established and evaluation of the diagnostic potential of the protocol was performed in a total of 67 pregnancies. In 97% of the couples one or more different SNPs within the deletion breakpoint occurred between paternal and maternal alleles. Homozygosity for the (−−SEA) deletion was accurately excluded in 33 out of 67 (49.3%, 95% CI, 25.4–78.6%) pregnancies through the implementation of the protocol. Protocol was completely concordant with the traditional reference methods, except for two cases that exhibited uncertain results due to sample hemolysis. This method could be used as a routine NIPD test to exclude gross fetal deletions in α-thalassemia major, and could further be employed to test for other diseases due to gene deletion
