    AbstractBased on multiple-index comprehensive evaluation, the evaluation system of agricultural modernization was constructed, and the level of agricultural modernization in central China was evaluated. As results showed, central China as a whole is still in the beginning stages of the modernization of agriculture, lagging behind the eastern region significantly. To promote agricultural modernization of central China, differently local resource endowments and levels of development on current situation must be considered, and measures should be taken in a line with the local condition and step-by-step, exerting their comparative advantages and selecting the dominant mode, strategic focus and implementation path of agricultural modernization

    Risk factors and the value of microbiological examinations of COVID-19 associated pulmonary aspergillosis in critically ill patients in intensive care unit: the appropriate microbiological examinations are crucial for the timely diagnosis of CAPA

    IntroductionDuring the Omicron pandemic in China, a significant proportion of patients with Coronavirus Disease 2019 (COVID-19) associated pulmonary aspergillosis (CAPA) necessitated admission to intensive care unit (ICU) and experienced a high mortality. To explore the clinical risk factors and the application/indication of microbiological examinations of CAPA in ICU for timely diagnosis are very important.MethodsThis prospective study included patients with COVID-19 admitted to ICU between December 1, 2022, and February 28, 2023. The clinical data of influenza-associated pulmonary aspergillosis (IAPA) patients from the past five consecutive influenza seasons (November 1, 2017, to March 31, 2022) were collected for comparison. The types of specimens and methods used for microbiological examinations were also recorded to explore the efficacy in early diagnosis.ResultsAmong 123 COVID-19 patients, 36 (29.3%) were diagnosed with probable CAPA. CAPA patients were more immunosuppressed, in more serious condition, required more advanced respiratory support and had more other organ comorbidities. Solid organ transplantation, APACHEII score ≥20 points, 5 points ≤SOFA score <10 points were independent risk factors for CAPA. Qualified lower respiratory tract specimens were obtained from all patients, and 84/123 (68.3%) patients underwent bronchoscopy to obtain bronchoalveolar lavage fluid (BALF) specimens. All patients’ lower respiratory tract specimens underwent fungal smear and culture; 79/123 (64.2%) and 69/123 (56.1%) patients underwent BALF galactomannan (GM) and serum GM detection, respectively; metagenomic next-generation sequencing (mNGS) of the BALF was performed in 62/123 (50.4%) patients. BALF GM had the highest diagnostic sensitivity (84.9%), the area under the curve of the mNGS were the highest (0.812).ConclusionThe incidence of CAPA was extremely high in patients admitted to the ICU. CAPA diagnosis mainly depends on microbiological evidence owing to non-specific clinical manifestations, routine laboratory examinations, and CT findings. The bronchoscopy should be performed and the BALF should be obtained as soon as possible. BALF GM are the most suitable microbiological examinations for the diagnosis of CAPA. Due to the timely and accuracy result of mNGS, it could assist in early diagnosis and might be an option in critically ill CAPA patients

    Diffusion Model is Secretly a Training-free Open Vocabulary Semantic Segmenter

    Recent research has explored the utilization of pre-trained text-image discriminative models, such as CLIP, to tackle the challenges associated with open-vocabulary semantic segmentation. However, it is worth noting that the alignment process based on contrastive learning employed by these models may unintentionally result in the loss of crucial localization information and object completeness, which are essential for achieving accurate semantic segmentation. More recently, there has been an emerging interest in extending the application of diffusion models beyond text-to-image generation tasks, particularly in the domain of semantic segmentation. These approaches utilize diffusion models either for generating annotated data or for extracting features to facilitate semantic segmentation. This typically involves training segmentation models by generating a considerable amount of synthetic data or incorporating additional mask annotations. To this end, we uncover the potential of generative text-to-image conditional diffusion models as highly efficient open-vocabulary semantic segmenters, and introduce a novel training-free approach named DiffSegmenter. Specifically, by feeding an input image and candidate classes into an off-the-shelf pre-trained conditional latent diffusion model, the cross-attention maps produced by the denoising U-Net are directly used as segmentation scores, which are further refined and completed by the followed self-attention maps. Additionally, we carefully design effective textual prompts and a category filtering mechanism to further enhance the segmentation results. Extensive experiments on three benchmark datasets show that the proposed DiffSegmenter achieves impressive results for open-vocabulary semantic segmentation

    Ultradispersed Electro-explosive Iron Powders as Catalysts for Synthesis of Liquid Hydrocarbons of CO and H[2]

    Catalytic activity of ultradispersed iron powders (UDIP) obtained by electric explosion of the conductor in media of nitrogen, carbon monoxide, and carbon dioxide in synthesis of liquid hydrocarbons by Fischer-Tropsch method was studied. It was shown that iron powder obtained in media of CO[2] has the highest specific surface area. A sample of powder was pelletized at pressure of 21 MPa during 30 seconds with use of 10 mass% polyvinyl alcohol as adhesive for experimentation in catalytic system. Catalyst fraction of 1-2 mm was selected for study. The experiments were carried out under conditions of 1 MPa, 300 mln/min of total reactants consumption, and varied values of temperature and reactants ratio. The maximal conversion level of CO was reached at 290°С and reactants ratio of H[2]:CO=2 in the initial mixture. It was shown that UDIP has high activity at lowered concentration of hydrogen in the initial mixture. The obtained mixture of liquid hydrocarbons is applicable for further refining for upgrading and improving of operating features

    GRIM-19 Disrupts E6/E6AP Complex to Rescue p53 and Induce Apoptosis in Cervical Cancers

    BACKGROUND: Our previous studies showed a down-regulation of GRIM-19 in primary human cervical cancers, and restoration of GRIM-19 induced tumor regression. The induction of tumor suppressor protein p53 ubiquitination and degradation by E6 oncoportein of high risk-HPV through forming a stable complex with E6AP is considered as a critical mechanism for cervical tumor development. The aims of this study were to determine the potential role of GRIM-19 in rescuing p53 protein and inducing cervical cancer cell apoptosis. METHODOLOGY/PRINCIPAL FINDINGS: The protein levels of GRIM-19 and p53 were detected in normal cervical tissues from 45 patients who underwent hysterectomy for reasons other than neoplasias of either the cervix or endometrium, and cervical cancer tissues from 60 patients with non-metastatic squamous epithelial carcinomas. Coimmunoprecipitation and GST pull-down assay were performed to examine the interaction of GRIM-19 with 18E6 and E6AP in vivo and in vitro respectively. The competition of 18E6 with E6AP in binding GRIM-19 by performing competition pull-down assays was designed to examine the disruption of E6/E6AP complex by GRIM-19. The augment of E6AP ubiquitination by GRIM-19 was detected in vivo and in vitro ubiquitination assay. The effects of GRIM-19-dependent p53 accumulation on cell proliferation, cell cycle, apoptosis were explored by MTT, flow cytometry and transmission electron microscopy respectively. The tumor suppression was detected by xenograft mouse model. CONCLUSION/SIGNIFICANCE: The levels of GRIM-19 and p53 were concurrently down regulated in cervical cancers. The restoration of GRIM-19 can induce ubiquitination and degradation of E6AP, and disrupt the E6/E6AP complex through the interaction of N-terminus of GRIM-19 with both E6 and E6AP, which protected p53 from degradation and promoted cell apoptosis. Tumor xenograft studies also revealed the suppression of p53 degradation in presence of GRIM-19. These data suggest that GRIM-19 can block E6/E6AP complex; and synergistically suppress cervical tumor growth with p53

    YOLOv6: A Single-Stage Object Detection Framework for Industrial Applications

    For years, the YOLO series has been the de facto industry-level standard for efficient object detection. The YOLO community has prospered overwhelmingly to enrich its use in a multitude of hardware platforms and abundant scenarios. In this technical report, we strive to push its limits to the next level, stepping forward with an unwavering mindset for industry application. Considering the diverse requirements for speed and accuracy in the real environment, we extensively examine the up-to-date object detection advancements either from industry or academia. Specifically, we heavily assimilate ideas from recent network design, training strategies, testing techniques, quantization, and optimization methods. On top of this, we integrate our thoughts and practice to build a suite of deployment-ready networks at various scales to accommodate diversified use cases. With the generous permission of YOLO authors, we name it YOLOv6. We also express our warm welcome to users and contributors for further enhancement. For a glimpse of performance, our YOLOv6-N hits 35.9% AP on the COCO dataset at a throughput of 1234 FPS on an NVIDIA Tesla T4 GPU. YOLOv6-S strikes 43.5% AP at 495 FPS, outperforming other mainstream detectors at the same scale~(YOLOv5-S, YOLOX-S, and PPYOLOE-S). Our quantized version of YOLOv6-S even brings a new state-of-the-art 43.3% AP at 869 FPS. Furthermore, YOLOv6-M/L also achieves better accuracy performance (i.e., 49.5%/52.3%) than other detectors with a similar inference speed.     Andersson‐Magnéli phases TinO2n‐1: recent progress inspired by Swedish scientists

The characteristic properties of these materials depend strongly on the compositional deviation from TiO2 and the way in which the structure accommodates the deviation. Thus, an urgent requirement is to overcome difficulties in characterizing such materials at atomic resolution. Here, we trace the discovery of the Andersson‐Magnéli phases, and report the application of recent developments in electron microscopy to reveal the relation, at the local level, between structural characteristics and electronic states, specifically for the materials TinO2n‐1 with n=4–8. The electrical conductivity of Ti4O7 has been reported previously to show three clearly distinct states on decreasing temperature from 300 K. For this reason, we focus on Ti4O7 as a representative example of the TinO2n‐1 phases and report structural characteristics at temperatures corresponding to each of the three different phases, focusing on the distribution of Ti3+ and Ti4+ cations from analysis of single‐crystal XRD data.     Role of ethylene in the regulatory mechanism underlying the abortion of ovules after fertilization in Xanthoceras sorbifolium

    Key message Genes related to the MAPK cascade, ethylene signaling pathway, Pi starvation response, and NAC TFs were differentially expressed between normal and abortive ovules. Receptor-mediated ethylene signal perception and transmission play an important role in regulating fruit and ovule development. Xanthoceras sorbifolium, a small to medium-sized tree endemic to northern China, is an emerging dedicated oilseed crop designed for applications in advanced biofuel, engine oil, and functional food, as well as for pharmaceutical and cosmetic applications. Despite the importance of Xanthoceras seed oil, low seed productivity has constricted commercial exploitation of the species. The abortion of developing seeds (ovules after fertilization) is a major factor limiting fruit and seed production in the plant. To increase fruit and seed yields, a better understanding of the mechanisms underlying the abortion of fertilized ovules is critical. This study revealed differences in nucellus degeneration, endosperm development, and starch grain content between normally and abnormally developing ovules after fertilization. We constructed 6 RNA-sequencing (RNA-seq) libraries from normally and abnormally developing ovules at the onset of their abortion process. Comparative transcriptome analysis between the normal and abnormal ovules identified 818 differentially expressed genes (DEGs). Among DEGs, many genes involved in mitogen-activated protein kinase (MAPK) cascades, ethylene signaling pathway, and NAC transcription factor genes showed up-regulated expression in abnormal ovules. The RNA-seq data were validated using quantitative reverse-transcription PCR. Using virus-induced gene silencing (VIGS) methods, evaluation of an ethylene receptor gene (XsERS) function indicated that the gene was closely related to early development of fruits and seeds. Based on the data presented here, we propose a model for a MAPK-ethylene signaling-NAC2 gene regulatory cascade that plays an important role in the regulation of the ovule abortion process in X. sorbifolium. The present study is imperative for understanding the mechanisms of ovule abortion after fertilization and identifying the critical genes and gene networks involved in determining the fate of ovule development
