Pn wave velocity and anisotropy underneath the central segment of the North-South Seismic Belt in China
We present a Pn wave velocity and anisotropy model of the central segment of the North-South Seismic Belt in China, where there are numerous stable basins and active faults, making this segment attractive for extensive studies. The model was obtained by a tomographic analysis of 49,973 Pn wave phase readings collected by the China Earthquake Networks Center and temporary stations in Yunnan and Sichuan. The tomographic velocity model shows that the average Pn wave velocity is 8.06km/s; prominent high-velocity (high-V) anomalies are visible under the Sichuan Basin, the Zoige Basin and the Ordos block, which clearly outline their tectonic mar- gins. A pronounced low-velocity (low-V) zone is observed from the Songpan-Ganzi block to the Chuan-Dian and Daliangshan blocks, suggesting the presence of hot material upwelling. The station delay data show a gradual variation from negative to positive values, possibly reflecting a crustal thickness variation from the southwest to the northeast of the study area. A correlation between the Pn wave anisotropy and the distribution of velocity anomalies is observed: anisotropy is relatively weaker in the high-V anomaly zones beneath stable basins, while it is stronger in the low-V anomaly zones and the high-to-low-V anomaly transition zones. The high-resolution velocity and anisotropy tomographic model that we obtained could also provide a better understanding of the study area seismicity, since the occurrence of strong earthquakes seems to be related to the presence and strength of lateral heterogeneities at the uppermost mantle level
Hydrous melting driven upwelling from the mantle transition zone in the Mongolia plateau revealed by receiver function analysis
The detailed images of the mantle transition zone (MTZ) give insights into the nature of mantle upwelling from the deep Earth. We use receiver function to investigate the MTZ structure beneath the Mongolia Plateau using earthquakes recorded by 223 stations deployed in the Central Asian Orogenic Belt. The topographies of the 410 km (D410) and 660 km (D660) discontinuities are both characterized by slight depressions in most parts of the Mongolia Plateau. An obvious low-velocity layer (LVL) atop the MTZ with an average thickness of ∼40 km is observed beneath this study area, indicating the hydrous partial molten layer atop the D410 and a hydrous MTZ. The D410 depth of ∼410 km and the LVL up to ∼60 km thick beneath the Dariganga volcano imply that there is a chemically distinct hydrous upwelling related to the subducted Pacific slab rather than a classic thermal-dominated upwelling. The ∼10 km thinned MTZs are found beneath the western Hangay Dome and the Middle Gobi volcano. A narrow NS-trending MTZ thickening zone extending from the Hovsgol rift to the central Hangay Dome is mainly caused by a ∼10 km depression of D660. We speculate that the mantle upwelling beneath the Hangay Dome may be related to the stagnant slab descending through the MTZ, rather than being rooted from the deep lower mantle. Effects of Dietary Crude Protein Levels on Fecal Crude Protein, Amino Acids Flow Amount, Fecal and Ileal Microbial Amino Acids Composition and Amino Acid Digestibility in Growing Pigs
The purpose of this experiment was to evaluate the effects of low protein corn-soybean meal-based diets on fecal CP, amino acid (AA) flow amount, AA digestibility and fecal and ileal microbial AA composition in growing pigs. Eighteen pigs (initial body weight = 30 ± 1.35) were randomly divided into three groups and fed with basal diets with CP levels of 12%, 15% and 18%, respectively. The Lys, Met + Cys, Thr and Trp level in the 12% CP and 15% CP groups is the same as 18% CP group by the addition of four crystalline Lys, Met + Cys, Thr and Trp to the diet. The results showed that with the decrease of dietary CP level from 18% to 12%, the fecal total nitrogen (N), CP and total AA (TAA) flow amount decreased linearly (p < 0.05). Dry matter (DM) digestibility, CP digestibility, TAA digestibility, essential amino acid (EAA) digestibility and non-essential amino acid (NEAA) digestibility increased linearly with the decrease of dietary CP concentration from 18% to 12%. Compared with 18% CP group, the flow amount of Asp, Ser, Glu, Gly, Tyr, Val, Leu and Phe in feces of pigs in the 15% CP group and 12% CP group decreased significantly, while the flow amount of Arg in the 15% CP group was lower than that in the 18% CP group and 12% CP group. The fecal microbial N and AA of the 15% CP group were higher than those of the 18% CP and 12% CP groups. Fecal TAA flow amount decreased linearly with the decrease of the dietary CP levels from 18% to 12%. Fecal TAA and NEAA flow amount also decreased linearly with the decrease of dietary CP level from 18% to 12%. Except for Glu, Gly, Met, Tyr, Thr and Phe, there were significant differences among the three groups in the composition of 17 kinds of AAs in fecal microorganisms. Among the 17 AA compositions of ileal microorganisms, except Tyr and Lys, the other AAs were significantly different among the three groups (p < 0.05
Mirror-Image Thymidine Discriminates against Incorporation of Deoxyribonucleotide Triphosphate into DNA and Repairs Itself by DNA Polymerases
DNA
polymerases are known to recognize preferably d-nucleotides
over l-nucleotides during DNA synthesis. Here, we report
that several general DNA polymerases catalyze polymerization reactions
of nucleotides directed by the DNA template containing an l-thymidine (l-T). The results display that the 5′–3′
primer extension of natural nucleotides get to the end at chiral modification
site with Taq and Phanta Max DNA polymerases, but the primer extension
proceeds to the end of the template catalyzed by Deep Vent (exo<sup>–</sup>), Vent (exo<sup>–</sup>), and Therminator DNA
polymerases. Furthermore, templating l-nucleoside displays
a lag in the deoxyribonucleotide triphosphate (dNTP) incorporation
rates relative to natural template by kinetics analysis, and polymerase
chain reactions were inhibited with the DNA template containing two
or three consecutive l-Ts. Most interestingly, no single
base mutation or mismatch mixture corresponding to the location of l-T in the template was found, which is physiologically significant
because they provide a theoretical basis on the involvement of DNA
polymerase in the effective repair of l-T that may lead to
cytotoxicity
Rational Modulation of Single Atom Coordination Microenvironments in a BCN Monolayer for Multifunctional Electrocatalysis
Single-atom (SA) catalysts (SACs) have demonstrated outstanding catalytic performances toward plenty of relevant electrochemical reactions. Nevertheless, controlling the coordination microenvironment of catalytically active SAs to further enhance their catalytic oerformences has remained elusive up to now. Herein, a systematic investigation of 20 transition metal atoms that are coordinated with 20 different microenvironments in a boroncarbon-nitride monolayer (BCN) is conducted using high-throughput density functional theory calculations. The experimentally synthesized ternary BCN monolayer contains carbon, nitrogen, and boron atoms in its 2D network, thus providing a lot of new coordination environments than those of the current CxNy nanoplatforms. By exploring the structural/electrochemical stability, catalytic activity, selectivity, and electronic properties of 400 (20 × 20) TM-BCN moieties, it is discovered that specific SA coordination environments can achieve superior stability and selectivity for different electrocatalytic reactions. Moreover, a universal descriptor to accelerate the experimental process toward the synthesis of BCN-SACs is reported. These findings not only provide useful guidance for the synthesis of efficient multifunctional BCN-SACs but also will immediately benefit researchers by levering up their understanding of the mechanistic effects of SA coordination microenvironments on electrocatalytic reactions.</p
Antibody-Free Colorimetric Detection of Total Aflatoxins in Rice Based on a Simple Two-Step Chromogenic Reaction
The prevalently used immunoassays
for fast screening of aftatoxins
(AFs) usually cannot meet the requirement for simultaneous determination
of total AFs (aflatoxin B<sub>1</sub> + aflatoxin B<sub>2</sub> +
aflatoxin G<sub>1</sub> + aflatoxin G<sub>2</sub>) due to the deficiency
of highly group-specific antibodies. This paper describes a two-step
chromogenic reaction based method to quantitatively detect total AFs
in rice using colorimetric measurement without antibody. In the method,
colorless AFs transform into green-colored indophenol products through
the reaction with sodium hydroxide and 2,6-dibromoquinone-4-chloroimide
(DBQC) successively, allowing selectively determining total AFs up
to 3.9 μg/kg over other competitive mycotoxins under optimal
conditions by a UV–vis spectrophotometer. In addition, the
colorimetric measurement results of the rice samples agree well with
that of a standard HPLC method, demonstrating the good reliability
and applicability of the method. Uniquely, the method has potential
for on-site detection of total AFs in rice when using a nylon membrane-based

Temporal dynamics of SARS-CoV-2 genome mutations that occurred in vivo on an aircraft
We analyzed variations in the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) genome during a flight-related cluster outbreak of coronavirus disease 2019 (COVID-19) in Shenzhen, China, to explore the characteristics of SARS-CoV-2 transmission and intra-host single nucleotide variations (iSNVs) in a confined space. Thirty-three patients with COVID-19 were sampled, and 14 were resampled 3–31 days later. All 47 nasopharyngeal swabs were deep-sequenced. iSNVs and similarities in the consensus genome sequence were analyzed. Three SARS-CoV-2 variants of concern, Delta (n = 31), Beta (n = 1), and C.1.2 (n = 1), were detected among the 33 patients. The viral genome sequences from 30 Delta-positive patients had similar SNVs; 14 of these patients provided two successive samples. Overall, the 47 sequenced genomes contained 164 iSNVs. Of the 14 paired (successive) samples, the second samples (T2) contained more iSNVs (median: 3; 95% confidence interval [95% CI]: 2.77–10.22) than did the first samples (T1; median: 2; 95% CI: 1.63–3.74; Wilcoxon test, P = 0.021). 38 iSNVs were detected in T1 samples, and only seven were also detectable in T2 samples. Notably, T2 samples from two of the 14 paired samples had additional mutations than the T1 samples. The iSNVs of the SARS-CoV-2 genome exhibited rapid dynamic changes during a flight-related cluster outbreak event. Intra-host diversity increased gradually with time, and new site mutations occurred in vivo without a population transmission bottleneck. Therefore, we could not determine the generational relationship from the mutation site changes alone

Lack of Remuscularization Following Transplantation of Human Embryonic Stem Cell-Derived Cardiovascular Progenitor Cells in Infarcted Nonhuman Primates
Human pluripotent stem cell-derived cardiovascular progenitor cells (hPSC-CVPCs) should be thoroughly investigated in large animal studies before testing in clinical trials.
The main of this study is to clarify whether hPSC-CVPCs can engraft for long time in the heart of primates after myocardial infarction (MI) and compare the effectiveness and safety of immunosuppression with cyclosporine alone or multiple-drug regimen (MDR) containing cyclosporine, methylprednisolone, and basiliximab in cynomolgus monkeys that had received intramyocardial injections of 1×10
EGFP (enhanced green fluorescent protein)-expressing hPSC-CVPCs after MI. A third group of animals received the immunosuppression MDR but without cell therapy after MI (MI+MDR group).
Measurements of EGFP gene levels and EGFP immunofluorescence staining indicated that the hPSC-CVPC engraftment rate was greater in the MI+MDR+CVPC group than that in the MI+cyclosporine+CVPC group. However, even in the MI+MDR+CVPC group, no transplanted cells could be detected at 140 days after transplantation. Concomitantly, immunofluorescent analysis of CD3, CD4, and CD8 expression indicated that T-lymphocyte infiltration in the CVPC-transplanted hearts was less in the MDR-treated animals than in the cyclosporine-alone-treated animals. The recovery of left ventricular function on day 28 post-MI in the MI+MDR+CVPC group was better than that in the MI+MDR group. Apoptotic cardiac cells were also less common in the MI+MDR+CVPC group than in the MI+MDR group, although both immunosuppression regimens were associated with transient hepatic dysfunction.
This is the largest study of hPSCs in nonhuman primates in cardiovascular field to date (n=32). Compared with cyclosporine alone, MDR attenuates immune rejection and improves survival of hPSC-CVPCs in primates; this is associated with less apoptosis of native cardiac cells and better recovery of left ventricular function at 28 days. However, even with MDR, transplanted hPSC-CVPCs do not engraft and do not survive at 140 days after transplantation, thereby excluding remuscularization as a mechanism for the functional effect
