    A target guided subband filter for acoustic event detection in noisy environments using wavelet packets

    This paper deals with acoustic event detection (AED), such as screams, gunshots, and explosions, in noisy environments. The main aim is to improve the detection performance under adverse conditions with a very low signal-to-noise ratio (SNR). A novel filtering method combined with an energy detector is presented. The wavelet packet transform (WPT) is first used for time-frequency representation of the acoustic signals. The proposed filter in the wavelet packet domain then uses a priori knowledge of the target event and an estimate of noise features to selectively suppress the background noise. It is in fact a content-aware band-pass filter which can automatically pass the frequency bands that are more significant in the target than in the noise. Theoretical analysis shows that the proposed filtering method is capable of enhancing the target content while suppressing the background noise for signals with a low SNR. A condition to increase the probability of correct detection is also obtained. Experiments have been carried out on a large dataset of acoustic events that are contaminated by different types of environmental noise and white noise with varying SNRs. Results show that the proposed method is more robust and better adapted to noise than ordinary energy detectors, and it can work even with an SNR as low as -15 dB. A practical system for real time processing and multi-target detection is also proposed in this work

    Determining layer number of two dimensional flakes of transition-metal dichalcogenides by the Raman intensity from substrate

    Transition-metal dichalcogenide (TMD) semiconductors have been widely studied due to their distinctive electronic and optical properties. The property of TMD flakes is a function of its thickness, or layer number (N). How to determine N of ultrathin TMDs materials is of primary importance for fundamental study and practical applications. Raman mode intensity from substrates has been used to identify N of intrinsic and defective multilayer graphenes up to N=100. However, such analysis is not applicable for ultrathin TMD flakes due to the lack of a unified complex refractive index (n~\tilde{n}) from monolayer to bulk TMDs. Here, we discuss the N identification of TMD flakes on the SiO2_2/Si substrate by the intensity ratio between the Si peak from 100-nm (or 89-nm) SiO2_2/Si substrates underneath TMD flakes and that from bare SiO2_2/Si substrates. We assume the real part of n~\tilde{n} of TMD flakes as that of monolayer TMD and treat the imaginary part of n~\tilde{n} as a fitting parameter to fit the experimental intensity ratio. An empirical n~\tilde{n}, namely, n~eff\tilde{n}_{eff}, of ultrathin MoS2_{2}, WS2_{2} and WSe2_{2} flakes from monolayer to multilayer is obtained for typical laser excitations (2.54 eV, 2.34 eV, or 2.09 eV). The fitted n~eff\tilde{n}_{eff} of MoS2_{2} has been used to identify N of MoS2_{2} flakes deposited on 302-nm SiO2_2/Si substrate, which agrees well with that determined from their shear and layer-breathing modes. This technique by measuring Raman intensity from the substrate can be extended to identify N of ultrathin 2D flakes with N-dependent n~\tilde{n} . For the application purpose, the intensity ratio excited by specific laser excitations has been provided for MoS2_{2}, WS2_{2} and WSe2_{2} flakes and multilayer graphene flakes deposited on Si substrates covered by 80-110 nm or 280-310 nm SiO2_2 layer.Comment: 10 pages, 4 figures.     Expression pattern and activity of six glutelin gene promoters in transgenic rice*

    The shortage of strong endosperm-specific expression promoters for driving the expression of recombinant protein genes in cereal endosperm is a major limitation in obtaining the required level and pattern of expression. Six promoters of seed storage glutelin genes (GluA-1, GluA-2, GluA-3, GluB-3, GluB-5, and GluC) were isolated from rice (Oryza sativa L.) genomic DNA by PCR. Their spatial and temporal expression patterns and expression potential in stable transgenic rice plants were examined with β-glucuronidase (GUS) used as a reporter gene. All the promoters showed the expected spatial expression within the endosperm. The GluA-1, GluA-2, and GluA-3 promoters directed GUS expression mainly in the outer portion (peripheral region) of the endosperm. The GluB-5 and GluC promoters directed GUS expression in the whole endosperm, with the latter expressed almost evenly throughout the whole endosperm, a feature different from that of other rice glutelin gene promoters. The GluB-3 promoter directed GUS expression solely in aleurone and subaleurone layers. Promoter activities examined during seed maturation showed that the GluC promoter had much higher activity than the other promoters. These promoters are ideal candidates for achieving gene expression for multiple purposes in monocot endosperm but avoid promoter homology-based gene silencing. The GluC promoter did not contain the endosperm specificity-determining motifs GCN4, AACA, and the prolamin-box, which suggests the existence of additional regulatory mechanism in determining endosperm specificity

    The role of Omi/HtrA2 protease in neonatal postasphyxial serum-induced apoptosis in human kidney proximal tubule cells

    Omi/HtrA2, a proapoptotic mitochondrial serine protease, is involved in both caspase-dependent and caspaseindependent apoptosis. A growing body of evidence indicates that Omi/HtrA2 plays an important role in the pathogenesis of a variety of ischemia-reperfusion (I/R) injuries. However, the role of Omi/HtrA2 in renal injuries that occur in neonates with asphyxia remains unknown. The present study was designed to investigate whether Omi/HtrA2 plays an important role in the types of renal injuries that are induced by neonatal postasphyxial serum. Human renal proximal tubular cell line (HK-2) cells were used as targets. A 20% serum taken from neonates one day after asphyxia was applied to target cells as an attacking factor. We initially included control and postasphyxial serum-attacked groups and later included a ucf-101 group in the study. In the postasphyxial serum-treated group, cytosolic Omi/HtrA2 and caspase-3 expression in HK-2 cells was significantly higher than in the control group. Moreover, the concentration of cytosolic caspase-3 was found to be markedly decreased in HK-2 cells in the ucf-101 group. Our results suggest both that postasphyxial serum has a potent apoptosis-inducing effect on HK-2 cells and that this effect can be partially blocked by ucf-101. Taken together, our results demonstrate for the first time that postasphyxial serum from neonates results in Omi/HtrA2 translocation from the mitochondria to the cytosol, where it promotes HK-2 cell apoptosis via a protease activity-dependent, caspase-mediated pathway

    SPOC is characterized by improving teaching effectiveness. Currently open teaching mode is the popular trend, which is mainly related to several aspects: how to carry out teaching practice by using MOOC proprietary, high-quality online teaching resources in open education, that is, deep integration of curriculum resources and teaching design. On the basis of SPOC development, combined with open education analysis of SPOC with instructional design theory and philosophy from flipped classroom as a guide, the present research try to propose instructional design model based on flipped classroom, including design of teaching content system, design of personalized learning strategies, design of teaching activities and teaching evaluation system. Four designs above contribute to the effective implementation of the open teaching activities based on the blended model of flipped classroom and SPOC teaching, so as to enhance the quality of education.     BACKGROUND Human papillomavirus type 16 (HPV 16) E2 protein is a multifunctional DNA-binding protein. HPV 16 E2 regulates many biological responses, including DNA replication, gene expression, and apoptosis. The purpose of this study was to investigate the relationship among the receptor for globular heads of the human C1q (gC1qR) gene expression, HPV 16 E2 transfection and apoptosis regulation in human cervical squamous carcinoma cells (C33a and SiHa). METHODS gC1qR expression was examined in C33a and SiHa cells using real-time PCR and Western blot analysis. Apoptosis of C33a and SiHa cells was assessed by flow cytometry. C33a and SiHa cell viability, migration and proliferation were detected using the water-soluble tetrazolium salt (WST-1) assay, a transwell assay and 3H-thymidine incorporation into DNA (3H-TdR), respectively. RESULTS C33a and SiHa cells that were transfected with a vector encoding HPV 16 E2 displayed significantly increased gC1qR gene expression and p38 mitogen-activated protein kinase (p38 MAPK)/c-jun N-terminal kinase (JNK) activation as well as up-regulation of cellular apoptosis, which was abrogated by the addition of gC1qR small interfering RNA (siRNA). Furthermore, the changes in C33a and SiHa cell viability, migration and proliferation that were observed upon HPV 16 E2 transfection were abrogated by SB203580 (a p38 MAPK inhibitor) or SP600125 (a JNK inhibitor) treatment. CONCLUSION These data support a mechanism whereby HPV 16 E2 induces apoptosis by silencing the gC1qR gene or inhibiting p38 MAPK/JNK signalling in cervical squamous cell carcinoma.This study was supported by grants from the National Natural Science Foundation of China (No. 81000251) and the Nanjing Medical Science and Technique Development Foundation

    Somatomotor-Visual Resting State Functional Connectivity Increases After Two Years in the UK Biobank Longitudinal Cohort

    Functional magnetic resonance imaging (fMRI) and functional connectivity (FC) have been used to follow aging in both children and older adults. Robust changes have been observed in children, where high connectivity among all brain regions changes to a more modular structure with maturation. In older adults, prior work has identified changes in connectivity associated with the default mode network (DMN); other work has used brain age to predict pre-clinical Alzheimer's disease. In this work, we find an increasing connectivity between the Somatomotor (SMT) and Visual (VIS) Networks using the Power264 atlas in a longitudinal cohort of the UK Biobank (UKB). This cohort consists of 2,722 subjects, with scans being taken an average of two years apart. The average connectivity increase between SMT-VIS is 6.8% compared to the younger scan baseline (from ρ=0.39\rho=0.39 to ρ=0.42\rho=0.42), and occurs in male, female, older subject (>65>65 years old), and younger subject (<55<55 years old) groups. Among all inter-network connections, this average SMT-VIS connectivity is the best predictor of relative scan age, accurately predicting which scan is older 57% of the time. Using the full FC and a training set of 2,000 subjects, one is able to predict which scan is older 82.5% of the time when using the difference of FC between the two scans as input to a classifier. This previously under-reported relationship may shed light on normal changes in aging brain FC, identifies a potential confound for longitudinal studies, and proposes a new area for investigation, specifically the SMT-VIS connectivity.Comment: 12 pages, 10 figures, 3 table
