    Vasorelaxant effect of total flavones from Dendranthema morifolium on rat thoracic aorta

    To investigate the vasorelaxant effect of total flavones from Dendranthema morifolium (Ramat.) Tzvel. cv. Hangju (FDM), tension was recorded from rat thoracic aortic rings. FDM completely relaxed, in a dose-dependent manner, the contractions induced by either phenylephrine (PE) or a high concentration of KCl (60 mM) in rings with intact endothelium. Mechanical removal of the endothelium did not significantly modify the vasorelaxant effect of FDM. In endothelium-denuded aortic rings depolarized by 60 mM KCl, FDM inhibited Ca/sup 2+/-induced contraction. FDM also reduced the transient contraction elicited by PE in Ca/sup 2+/-free medium, but had no effect on active phorbol ester-induced contraction. Pretreatment of endothelium-denuded aorta with propranolol, a beta-adrenoceptor antagonist, significantly attenuated the relaxant effect of FDM. These results indicate that FDM induces an endothelium-independent relaxation in rat aortic rings. The mechanisms may include the activation of beta-adrenergic receptors, reduction in Ca/sup 2+/ influx through the voltage-dependent and receptor-operated channels, and inhibition of intracellular Ca release in vascular smooth muscle cells.published_or_final_versio

    Extraction of Total Alkaloids from Corydalis saxicola Bunting by the Macroporous Resin

    The total alkaloids were extracted from Corydalis saxicola Bunting by macroporous resin.Six different macroporous resins were used for extraction of the total alkaloids and dehydrocavidine by using static and dynamic adsorption-desorption experiments.The specific adsorption capacity and eluting of D101 macroporous resin were comparatively high.The optimum parameters of the purification process were established as the sample concentration of 0.2 g/ml at the absorption ? ow rate of 2 BV/h and with 3 BV of 60% ethanol as the desorption solvent.The contents of total alkaloids in the purifi ed extract were over 60%. 利用大孔吸附樹脂分離巖黃連中的總生物堿,采用靜態和動態吸附-解吸附方法,以總生物堿和脫氫卡維丁含量為指標,考察了6種大孔吸附樹脂對巖黃連總生物堿的吸附能力,其中D101型大孔吸附樹脂的比吸附量和比洗脫量均較高。優化后的提取工藝為:上樣藥液原藥材濃度0.2g/ml,吸附流速為2BV/h,解吸附溶劑為60%乙醇(3BV),所得巖黃連提取物中總生物堿含量大于60%

    Establishing production service system and information collaboration platform for mold and die products

    This paper investigates how the new concept of product service systems can be used and extended to transform, elevate, and revitalize traditional equipment manufacturing industry such as the Mold and Die (MD) sector. A mold and die production service systems (MPSS) framework is established based on recent developments within our industrial collaborators. Within the MPSS framework, MD manufacturers become more specialized in producing MD products and components while sharing and outsourcing manufacturing-oriented services (MOS) from a service provider. Typical services include collaborative order pooling and release, collaborative project progress status tracking, contractor-managed collaborative outsourcing, collaborative product design, collaborative production planning and scheduling, and after-sales technical supports. MOSs are designed, developed, and deployed as SaaS (software as application services) following the service-oriented architecture. Collectively, they form iMPSS-an Information and Collaboration Platform that enables MPSS.     Cardio-Protection of Salvianolic Acid B through Inhibition of Apoptosis Network

    Targeting cellular function as a system rather than on the level of the single target significantly increases therapeutic potency. In the present study, we detect the target pathway of salvianolic acid B (SalB) in vivo. Acute myocardial infarction (AMI) was induced in rats followed by the treatment with 10 mg/kg SalB. Hemodynamic detection and pathological stain, 2-dimensional electrophoresis, MALDI-TOF MS/MS, Western blot, pathway identification, apoptosis assay and transmission electron microscope were used to elucidate the effects and mechanism of SalB on cardioprotection. Higher SalB concentration was found in ischemic area compared to no-ischemic area of heart, correlating with improved heart function and histological structure. Thirty-three proteins regulated by SalB in AMI rats were identified by biochemical analysis and were classified as the components of metabolism and apoptosis networks. SalB protected cardiomyocytes from apoptosis, inhibited poly (ADP-ribose) polymerase-1 pathway, and improved the integrity of mitochondrial and nucleus of heart tissue during AMI. Furthermore, the protective effects of SalB against apoptosis were verified in H9c2 cells. Our results provide evidence that SalB regulates multi-targets involved in the apoptosis pathway during AMI and therefore may be a candidate for novel therapeutics of heart diseases

    Lipid-dependent gating of a voltage-gated potassium channel

    Recent studies hypothesized that phospholipids stabilize two voltage-sensing arginine residues of certain voltage-gated potassium channels in activated conformations. It remains unclear how lipids directly affect these channels. Here, by examining the conformations of the KvAP in different lipids, we showed that without voltage change, the voltage-sensor domains switched from the activated to the resting state when their surrounding lipids were changed from phospholipids to nonphospholipids. Such lipid-determined conformational change was coupled to the ion-conducting pore, suggesting that parallel to voltage gating, the channel is gated by its annular lipids. Our measurements recognized that the energetic cost of lipid-dependent gating approaches that of voltage gating, but kinetically it appears much slower. Our data support that a channel and its surrounding lipids together constitute a functional unit, and natural nonphospholipids such as cholesterol should exert strong effects on voltage-gated channels. Our first observation of lipid-dependent gating may have general implications to other membrane proteins

    A Triad of Lys12, Lys41, Arg78 Spatial Domain, a Novel Identified Heparin Binding Site on Tat Protein, Facilitates Tat-Driven Cell Adhesion

    Tat protein, released by HIV-infected cells, has a battery of important biological effects leading to distinct AIDS-associated pathologies. Cell surface heparan sulfate protoglycans (HSPGs) have been accepted as endogenous Tat receptors, and the Tat basic domain has been identified as the heparin binding site. However, findings that deletion or substitution of the basic domain inhibits but does not completely eliminate Tat–heparin interactions suggest that the basic domain is not the sole Tat heparin binding site. In the current study, an approach integrating computational modeling, mutagenesis, biophysical and cell-based assays was used to elucidate a novel, high affinity heparin-binding site: a Lys12, Lys41, Arg78 (KKR) spatial domain. This domain was also found to facilitate Tat-driven β1 integrin activation, producing subsequent SLK cell adhesion in an HSPG-dependent manner, but was not involved in Tat internalization. The identification of this new heparin binding site may foster further insight into the nature of Tat-heparin interactions and subsequent biological functions, facilitating the rational design of new therapeutics against Tat-mediated pathological events

    Differential Proteomic Analysis of Platelets Suggested Possible Signal Cascades Network in Platelets Treated with Salvianolic Acid B

    Salvianolic acid B (SB) is an active component isolated from Danshen, a traditional Chinese medicine widely used for the treatment of cardiovascular disorders. Previous study suggested that SB might inhibit adhesion as well as aggregation of platelets by a mechanism involving the integrin α2β1. But, the signal cascades in platelets after SB binding are still not clear.In the present study, a differential proteomic analysis (two-dimensional electrophoresis) was conducted to check the protein expression profiles of rat platelets with or without treatment of SB. Proteins altered in level after SB exposure were identified by MALDI-TOF MS/MS. Treatment of SB caused regulation of 20 proteins such as heat shock-related 70 kDa protein 2 (hsp70), LIM domain protein CLP-36, copine I, peroxiredoxin-2, coronin-1 B and cytoplasmic dynein intermediate chain 2C. The regulation of SB on protein levels was confirmed by Western blotting. The signal cascades network induced by SB after its binding with integrin α2β1 was predicted. To certify the predicted network, binding affinity of SB to integrin α2β1 was checked in vitro and ex vivo in platelets. Furthermore, the effects of SB on protein levels of hsp70, coronin-1B and intracellular levels of Ca²+ and reactive oxygen species (ROS) were checked with or without pre-treatment of platelets using antibody against integrin α2β1. Electron microscopy study confirmed that SB affected cytoskeleton structure of platelets.Integrin α2β1 might be one of the direct target proteins of SB in platelets. The signal cascades network of SB after binding with integrin α2β1 might include regulation of intracellular Ca²+ level, cytoskeleton-related proteins such as coronin-1B and cytoskeleton structure of platelets

    Self-Cleaning Glass of Photocatalytic Anatase TiO2@Carbon Nanotubes Thin Film by Polymer-Assisted Approach

    Due to the good photocatalytic activity, the TiO2@CNTs thin film is highly desirable to apply to the self-cleaning glass for green intelligent building. Here, the TiO2@CNTs thin film has been successfully achieved by polymer-assisted approach of an aqueous chemical solution method. The polymer, polyethylenimine, aims to combine the Ti4+ with CNTs for film formation of TiO2@CNTs. The resultant thin film was uniform, highly transparent, and super-hydrophilic. Owing to fast electron transport and effectively hindering electron-hole recombination, the TiO2@CNTs thin film has nearly twofold photocatalytic performance than pure TiO2. The TiO2@CNTs thin films show a good application for self-cleaning glasses
