D-lactic acid biosynthesis from biomass-derived sugars via Lactobacillus delbrueckii fermentation
Poly-lactic acid (PLA) derived from renewable resources is considered to be a good substitute for petroleum-based plastics. The number of poly L-lactic acid applications is increased by the introduction of a stereocomplex PLA, which consists of both poly-L and D-lactic acid and has a higher melting temperature. To date, several studies have explored the production of L-lactic acid, but information on biosynthesis of D-lactic acid is limited. Pulp and corn stover are abundant, renewable lignocellulosic materials that can be hydrolyzed to sugars and used in biosynthesis of D-lactic acid. In our study, saccharification of pulp and corn stover was done by cellulase CTec2 and sugars generated from hydrolysis were converted to D-lactic acid by a homofermentative strain, L. delbrueckii, through a sequential hydrolysis and fermentation process (SHF) and a simultaneous saccharification and fermentation process (SSF). 36.3 g Lˉ¹ of D-lactic acid with 99.8 % optical purity was obtained in the batch fermentation of pulp and attained highest yield and productivity of 0.83 g gˉ¹ and 1.01 g Lˉ¹ hˉ¹, respectively. Luedeking–Piret model described the mixed growth-associated production of D-lactic acid with a maximum specific growth rate 0.2 hˉ¹ and product formation rate 0.026 hˉ¹, obtained for this strain. The efficient synthesis of D-lactic acid having high optical purity and melting point will lead to unique stereocomplex PLA with innovative applications in polymer industry
Cellulolytic Enzymes Production via Solid-State Fermentation: Effect of Pretreatment Methods on Physicochemical Characteristics of Substrate
We investigated the effect of pretreatment on the physicochemical characteristics—crystallinity, bed porosity, and volumetric specific surface of soybean hulls and production of cellulolytic enzymes in solid-state fermentation of Trichoderma reesei and Aspergillus oryzae cultures. Mild acid and alkali and steam pretreatments significantly increased crystallinity and bed porosity without significant change inholocellulosic composition of substrate. Crystalline and porous steam-pretreated soybean hulls inoculated with T. reesei culture had 4 filter paper units (FPU)/g-ds, 0.6 IU/g-ds β-glucosidase, and 45 IU/g-ds endocellulase, whereas untreated hulls had 0.75 FPU/g-ds, 0.06 IU/g-ds β-glucosidase, and 7.29 IU/g-ds endocellulase enzyme activities. In A. oryzae steam-pretreated soybean hulls had 47.10 IU/g-ds endocellulase compared to 30.82 IU/g-ds in untreated soybean hulls. Generalized linear statistical model fitted to enzyme activity data showed that effects of physicochemical characteristics on enzymes production were both culture and enzyme specific. The paper shows a correlation between substrate physicochemical properties and enzyme production
Production of methyl ethyl ketone from biomass using a hybrid biochemical/catalytic approach
The recent demand for sustainable routes to fuels and chemicals has led to an increased amount of research in conversion of natural resources. A potential approach for conversion of biomass to fuels and chemicals is to combine biochemical and chemical processes. This research used microbial fermentation to produce 2,3-butanediol, which was then converted to methyl ethyl ketone by dehydration over a solid acid catalyst. The fermentation process was performed using the bacteria Klebsiella oxytoca (K.O). 2,3-butanediol then dehydrated to form methyl ethyl ketone on a solid acid catalyst, the proton form of ZSM-5, and heat. The goal was to determine the reaction kinetics of 2,3-butanediol dehydration over ZSM-5, and to demonstrate the hybrid biochemical/thermochemical approach for synthesizing chemicals from biomass. It was found that ZSM-5 produced methyl ethyl ketone with high selectivity (greater than 90%), and could convert fermentative 2,3-butanediol to methyl ethyl ketone. The reaction order of 2,3-butanediol dehydration was found to be slightly large than one, and an activation energy of 32.3 kJ/mol was measured
The Investigation of Virginiamycin-Added Fungal Fermentation on the Size and Immunoreactivity of Heat-Sensitive Soy Protein

The investigation of fungi fermentation effect on soy protein has been interrupted by substrate sterilization. Virginiamycin at 0.05% was added together with Aspergillus oryzae for solid state fermentation (SSF) in unsterilized soymeal (SM). When compared to A. oryzae SSF alone, virginiamycin did not cause the interference of fungal fermentation but elucidated the protein degradation. SDS-PAGE results showed that both alpha and alpha' subunits of beta-conglycinin were degraded significantly. In addition, western blot results showed that the immunoreactive signals of soy protein were considerably reduced in virginiamycin-added fermentation with unsterilized SM. Furthermore, fungal fermentation increased total protein and essential amino acid contents, suggesting the value enhancement of SM products. Taken together, this study demonstrated for the first time that virginiamycin could help investigate fermentation effect on heat-sensitive soy protein. Fermented SM has several potential applications in feed industry
Increased growing temperature reduces content of polyunsaturated fatty acids in four oilseed crops
Environmental temperature directly influences the lipid profile produced by oilseeds. If growing temperatures increase, as is predicted by current models, the precise profile of lipids produced are likely to change. This paper develops models to predict lipid profiles as a function of growing temperature. Data relating to lipid profiles of soybean (Glycine max), spring canola (Brassica napus), spring camelina (Camelina sativa), and sunflower (Helianthus annuus) were gathered from the literature and evaluated to examine the influence of temperature on relative production of oleic, linoleic, and linolenic acid. For each crop, a set of linear regressions was used to correlate temperature during the grain fill, defined as 30 days before harvest, with the molar percentages of oleic, linoleic, and linolenic acid present. An increase in temperature from 10 to 40°C resulted in an increase in the production of oleic acid and a decrease in the production of linoleic and linolenic acid in soybeans, canola, and sunflowers. Over the range of data available, the lipid profile of camelina was temperature insensitive. To test the validity of the correlations, the four crops were grown in a field study in Manhattan, Kansas simultaneously, in the same environment, in 2011. The correlations accurately predicted the field data for soybean, canola, and camelina but not for sunflower. The correlation for sunflower under-predicted the molar amount of oleic acid and over-predicted the molar amount of linoleic acid. This study indicates increasing growing temperatures from 10 to 40°C will result in more monounsaturated oils and less polyunsaturated oils in soybean, canola, and sunflower
A diploid wheat TILLING resource for wheat functional genomics

https://doi.org/10.1186/1471-2229-12-205Background: Triticum monococcum L., an A genome diploid einkorn wheat, was the first domesticated crop. As a diploid, it is attractive genetic model for the study of gene structure and function of wheat-specific traits. Diploid wheat is currently not amenable to reverse genetics approaches such as insertion mutagenesis and post-transcriptional gene silencing strategies. However, TILLING offers a powerful functional genetics approach for wheat gene analysis.
Results: We developed a TILLING population of 1,532 M[subscript 2] families using EMS as a mutagen. A total of 67 mutants were obtained for the four genes studied. Waxy gene mutation frequencies are known to be 1/17.6 - 34.4 kb DNA in polyploid wheat TILLING populations. The T. monococcum diploid wheat TILLING population had a mutation frequency of 1/90 kb for the same gene. Lignin biosynthesis pathway genes- COMT1, HCT2, and 4CL1 had mutation frequencies of 1/86 kb, 1/92 kb and 1/100 kb, respectively. The overall mutation frequency of the diploid wheat TILLING population was 1/92 kb.
Conclusion: The mutation frequency of a diploid wheat TILLING population was found to be higher than that reported for other diploid grasses. The rate, however, is lower than tetraploid and hexaploid wheat TILLING populations because of the higher tolerance of polyploids to mutations. Unlike polyploid wheat, most mutants in diploid wheat have a phenotype amenable to forward and reverse genetic analysis and establish diploid wheat as an attractive model to study gene function in wheat. We estimate that a TILLING population of 5, 520 will be needed to get a non-sense mutation for every wheat gene of interest with 95% probability
