    Exposure to organophosphate compounds, such as chlorpyrifos, has been linked to disturbances on cell signaling pathways. Mitogen activated protein kinases (MAPK) are a family of protein kinases involved in a range of cellular processes, including stress response, apoptosis and survival. Therefore, changes in the activation state of these kinases may characterize key mechanisms of toxicity elicited by xenobiotics. Here we report data on the phosphorylation of p38MAPK and JNK, members of the MAPK family, in Drosophila melanogaster exposed to chlorpyrifos, as characterized by western blotting assays

In this context, crude ethanolic extract (CEE), ethyl acetate fraction (EAF), and methanolic fraction (MF) from Anacardium microcarpum were tested alone or in combination with antibiotics (amikacin, gentamicin, ciprofloxacin, and imipenem) against Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. Methods: Antibiotic resistance-modifying activity was performed using the microdilution method by determining the minimal inhibitory concentration (MIC). In addition, phytochemical prospecting analyses of tested samples were carried out. Results: Our results indicated that all the extracts showed low antibacterial activity against multidrug-resistant strains (MIC =512 µg/mL). However, addition of CEE, EAF, and MF to the growth medium at the subinhibitory concentration (MIC/8=64 µg/mL) significantly modulated amikacin- and gentamicin-resistant E. coli 06. CEE and EAF also demonstrated a significant (P<0.001) synergism with imipenem against S. aureus. In contrast, MF antagonized the antibacterial effect of ciprofloxacin and gentamicin against P. aeruginosa 03 and S. aureus 10, respectively. Qualitative phytochemical analysis of the extracts revealed the presence of secondary metabolites including phenols, flavonoids, xanthones, chalcones, and tannin pyrogallates. Conclusion: Taken together, our results suggest that A. microcarpum is a natural resource with resistance-modifying antibacterial activity that needs to be further investigated to overcome the present resistant-infection problem.     Protective effects of organoselenium compounds against methylmercury-induced oxidative stress in mouse brain mitochondrial-enriched fractions

    We evaluated the potential neuroprotective effect of 1-100 µM of four organoselenium compounds: diphenyl diselenide, 3’3-ditri-fluoromethyldiphenyl diselenide, p-methoxy-diphenyl diselenide, and p-chloro-diphenyl diselenide, against methylmercury-induced mitochondrial dysfunction and oxidative stress in mitochondrial-enriched fractions from adult Swiss mouse brain. Methylmercury (10-100 µM) significantly decreased mitochondrial activity, assessed by MTT reduction assay, in a dose-dependent manner, which occurred in parallel with increased glutathione oxidation, hydroperoxide formation (xylenol orange assay) and lipid peroxidation end-products (thiobarbituric acid reactive substances, TBARS). The co-incubation with diphenyl diselenide (100 µM) completely prevented the disruption of mitochondrial activity as well as the increase in TBARS levels caused by methylmercury. The compound 3’3-ditrifluoromethyldiphenyl diselenide provided a partial but significant protection against methylmercury-induced mitochondrial dysfunction (45.4 ± 5.8% inhibition of the methylmercury effect). Diphenyl diselenide showed a higher thiol peroxidase activity compared to the other three compounds. Catalase blocked methylmercury-induced TBARS, pointing to hydrogen peroxide as a vector during methylmercury toxicity in this model. This result also suggests that thiol peroxidase activity of organoselenium compounds accounts for their protective actions against methylmercury-induced oxidative stress. Our results show that diphenyl diselenide and potentially other organoselenium compounds may represent important molecules in the search for an improved therapy against the deleterious effects of methylmercury as well as other mercury compounds

    MULTIPLICAÇÃO IN VITRO DE PORTA-ENXERTOS DO GÊNERO PRUNUS SOB DIFERENTES CONCENTRAÇÕES DE BAP EM DOIS MEIOS DE CULTURA IN VITRO MULTIPLICATION OF PRUNUS ROOTSTOCKS IN DIFFERENT BAP CONCENTRATIONS IN TWO CULTURE MEDIA

    Este trabalho teve como objetivo determinar a melhor concentração de BAP (6-benzilaminopurina) e meio de cultura para a multiplicação in vitro de porta-enxertos de Prunus sp., recentemente introduzidos no Brasil. Os porta-enxertos G x N22, GF 677, Mr.S 2/5, Marianna e Mirabolano foram testados em dois meios de cultura, meio MS e meio MS ¾ (reduzido em 25% dos sais do meio inteiro), combinados com quatro concentrações da citocinina BAP: 0,1; 0,3; 0,5 e 0,7 mg.L-1. Observou-se que os genótipos apresentaram comportamentos diferentes entre si em relação às concentrações de BAP e aos meios. Concluiu-se que, para os porta-enxertos G x N22 e Mr.S 2/5, o melhor meio de multiplicação é o MS ¾ com BAP na concentração de 0,7 mg.L-1; para o porta-enxerto Marianna, o meio MS com BAP na concentração de 0,7 mg.L-1; para o porta-enxerto Mirabolano, o meio MS ¾ com BAP na concentração de 0,5 mg.L-1, sendo que, para este porta-enxerto, o BAP exerce efeito negativo sobre o tamanho das brotações, independentemente do tipo de meio. Já para o porta-enxerto G x N22, este efeito se fez notar apenas no meio MS. O porta-enxerto GF 677 não apresentou bons resultados na propagação in vitro.<br>The purpose of this work was to determine the best BAP concentration and culture media for the in vitro multiplication of Prunus rootstocks starting from nodal segments. The rootstocks G x N22, GF 677, Mr.S 2/5, Marianna and Mirabolano were tested in two culture media (MS and MS ¾) combined with four BAP concentrations (0.1; 0.3; 0.5 and 0.7 mg.L-1). It was observed that the genotypes had different performance as the two media and BAP concentrations are concerned. BAP provided better results in the MS ¾ for the rootstocks G x N22 and Mr.S 2/5. For 'Marianna', the best results were found in the MS at 0.7 mg.L-1 BAP. 'Mirabolano', showed better results in the MS ¾ at 0.5 mg.L-1 BAP and in the two tested media, the shoot length decreased with the increase for BAP. The rootstock G x N22, had the same effect only in the MS medium. The rootstock GF 677 didn't present good results in micropropagation

    Evaluation of behavioral states among morning and evening active healthy individuals

    The Horne-Östberg questionnaire partly covers some factors that may be important determinants of peak time and characterize patterns of behavior. We conducted a study for the evaluation of self-reported behavioral states (hunger sensation, availability for study, physical exercise, solving daily problems, and time preferences) as expressions of underlying cyclic activity. Three hundred and eighteen community subjects without history of medical, psychiatric, or sleep disorders were evaluated in a cross-sectional design. A self-report about daily highest level of activity was used to categorize individuals into morning, evening, and indifferently active. Time-related behavioral states were evaluated with 23 visual analog questions. The responses to most analogic questions were significantly different between morning and evening active subjects. Logistic regression analysis identified a group of behaviors more strongly associated with the self-reported activity pattern (common wake up time, highest subjective fatigue, as well as wake up, bedtime, exercise and study preferences). These findings suggested that the patterns of activity presented by normal adults were related to specific common behavioral characteristics that may contribute to peak time
