    Prepartum and Postpartum Rumen Fluid Microbiomes: Characterization and Correlation with Production Traits in Dairy Cows

    Microbes present in the rumen of dairy cows are essential for degradation of cellulosic and nonstructural carbohydrates of plant origin. The prepartum and postpartum diets of high-producing dairy cows are substantially different, but in what ways the rumen microbiome changes in response and how those changes may influence production traits are not well elucidated. Here, we sequenced the 16S and 18S rRNA genes using the MiSeq platform to characterize the prepartum and postpartum rumen fluid microbiomes in 115 high-producing dairy cows, including both primiparous and multiparous animals. Discriminant analysis identified differences between the microbiomes of prepartum and postpartum samples and between primiparous and multiparous cows. 18S rRNA sequencing revealed an overwhelming dominance of the protozoan class Litostomatea, with over 90% of the eukaryotic microbial population belonging to that group. Additionally, fungi were relatively more prevalent and Litostomatea relatively less prevalent in prepartum samples than in postpartum ones. The core rumen microbiome (common to all samples) consisted of 64 bacterial taxa, of which members of the genus Prevotella were the most prevalent. The Chao1 richness index was greater for prepartum multiparous cows than for postpartum multiparous cows. Multivariable models identified bacterial taxa associated with increased or reduced milk production, and general linear models revealed that a metagenomically based prediction of productivity is highly associated with production of actual milk and milk components. In conclusion, the structure of the rumen fluid microbiome shifts between the prepartum and first-week postpartum periods, and its profile within the context of this study could be used to accurately predict production traits

    Influence of fluid therapy for stabilization of dogs in shock caused with different etiology

    The aim of this work is the portrayal of shock in a small practice and its timely and proper treatment. This study included a total of 8 dogs that are numbered from 1 to 8. In all animals was performed complete general clinical index, the venous route was applyed, and are accompanied by clinical parameters during treatment. Research showing that all animals were received in shock. In all our study patients, fluid therapy was occurred to update and maintain lost volume, exception makes patient number one witch with respect to the diagnosis and classification of shock succumb on other therapy. All patients received fluid therapy in the form of one or multiple bolus colloid fluids Hydroxyethyl Starch 6% (HAES-a) dosage of 3-15 ml/kg with a physiological solution in the dosage of 10-50 ml/kg. Our study showed a positive response after applications of bolus HAES and physiological solutions. Dogs 1, 2, 5 and 6 after therapy was stabilize body temperature (TT), CRT, and the frequency. The most relevant example is the dog number 2, who received value of TT 40.9, CRT 4 seconds, and the frequency was 222, which the value for 18 hours changed to TT 38.4, CRT 3 seconds and the frequency was 180th. Dogs (5, 6) gave a positive response to the protocol to stabilize shock. Dogs 3, 4, 7 and 8 have an initial positive reactions in some parameters, but etiological situation has led to deterioration and mortalities. We have concluded that the time of applications and attempt have significant role in the successible treatment to stabilize a patients. Significant results are possible only if treatment is timely and adequately (a combination of colloids and crystalloid). The basis of shock therapy is the correction hipovolemia appropriate liquid, intravenous applied in the fastest possible time

    Laparoscopy-promising tool for improvement of reproductive efficiency of small ruminants

    Assisted reproductive technologies are used to accelerate genetic gain and improve reproductive performances in farm animals, including small ruminants. This technologies include estrous synchronization, artifi cial insemination (AI) using fresh, frozen or sexed semen, embryo transfer (ET) using in vivo or in vitro produced embryos, and more advanced - cloning and production of transgenic animals. Diagnostic procedures, such as ultrasonography and laparoscopy, have been used as additional tools for monitoring the ovarian response to superovulatory treatment in donor animals as well as for AI and collection and transfer of embryos. The use of laparoscopy for assisted reproduction techniques in Macedonia commenced in the early 90ā€™s, with the acquisition of a set of ,,Karl Storzā€ equipment. After the adoption of the required routine, our group has completed several scientifi c projects where laparoscopy was used for intrauterine inseminations as well as for recovery and transfer of embryos in both sheep andgoats. In the following period our group endeavored into introduction of laparoscopic insemination in the routine farm practice. Ovine intrauterine/intracornual insemination by frozen-thawed semen resulted with pregnancy rates of 45% and 60%, when AI was performed out of season and during the breeding season, respectively. In goats, this percentage occasionally peaked at 85%.     Immunohistochemical detection of estrogen receptors in canine mammary tumors

    Mammary tumors are among the most common neoplasms in intact female dogs.They have a complex morphology, usually affecting middle age and older bitches. Almost 50% of the mammary tumors in dogs are malignant neoplasms. Prognosis is based on several factors: stage, age, tumor size, metastasis, histopathology, ovariectomy status and hormone-receptor activity. Immunohistochemical (IHC) measurement has become increasingly an important diagnostic and prognostic parameter, with the development of monoclonal antibodies against nuclear estrogen and progestin receptors. The aim of this study was to detect the presence of ER receptors in malignant canine mammary tumors and to identify their association with the clinical course of the tumor. Mammary tumor samples have been obtained by mastectomy from dogs presented at our clinic. Detailed clinical examination, CBC and basic serum biochemical profile were performed in all patients. Surgery was the only treatment. Histopathological examination and immunohistochemical detection of estrogen Ī± receptors (ERĪ±) was performed on 8 formalin-fixed, paraffin-embedded tissue samples, using the PT LINK immunoperoxidase technique. Histopathological examination of the mammary tumor samples (n=11) revealed tubular adenocarcinoma (n=6,54.5%) and ductal adenocarcinoma (n=3, 27.3%), one patient with benign adenoma and one with mastitis. Patients with positive ER tumors are alive, without remission, while 3 of the patients that were ER negative died due to lung metastases. According to our results, it can be concluded that the appearance and development of canine mammary tumors is highly connected with ovarian steroid hormones and that immunostaining of the tumors may be used as a good prognostic parameter in these patients
