    Assessing the accuracy of δ<sup>18<\sup>O<sub>sw<?sub> estimates from corals: lessons from simple Monte Carlo simulations

    EGU2008-A-04391 Paired measurements of δ18O and Sr/Ca in coral aragonite are routinely used for deriving estimates of δ18Osw and, by extension, sea surface salinity variations over the past centuries. However, in practice, the accuracy (or the error) of these estimates is often difficult to assess. Here, we use simulated proxy data and Monte-Carlo simulations to investigate the accuracy of δ18Osw estimates from paired coral δ18O and Sr/Ca measurements. First, we estimate expected values of coral Sr/Ca and δ18O from instrumental or reanalysis data of sea surface temperature (SST) and sea surface salinity (SSS). We then add the typical analytical errors onto the expected Sr/Ca (δ18O) data as random numbers and compute δ18Osw+error from the noisy proxy data for a 1000 sample Monte Carlo. From this simple Monte Carlo simulation, the range of correlation coefficients between δ18Osw+error and expected δ18Osw is estimated. As expected, we find that this range mainly depends on the magnitude of the actual SSS variations at a given site, as well as on the slope of the δ18Osw-SSS relationship. A comparison with real coral-based δ18Osw reconstructions from multiple sites indicates that correlations between reconstructed δ18Osw and instrumental SSS fall within the range of correlation coefficients predicted based on our Monte-Carlo simulation. Thus, our simple simulation exercise may help to assess the feasibility of δ18Osw and salinity reconstructions from corals in different climatic settings, provided that (i) some instrumental data of δ18Osw and/or SSS is available, and (ii) the slope of the δ18Osw-SSS relationship is known

    AbstractNew results on three-dimensional micro-structuring of tungsten carbide hard metal and steel using femtosecond laser pulses will be presented. For the investigations, a largely automated high-precision fs-laser micromachining station was used. The fs-laser beam is focused onto the sample surface using different objectives. The investigations of the ablation behaviour of the various materials in dependence of the laser processing parameters will be presented. In the second part, complex 3D microstructures with a variety of geometries and resolutions down to a few micrometers will be presented. On of the Goal of these investigations was to create defined microstructures in tooling equipments such as cutting inserts

    Mutational analysis of the internal membrane proximal domain of the HIV glycoprotein C-terminus

    AbstractThis study focuses on the long stretch of highly conserved amino acids in the membrane proximal part of the HIV-1 cytoplasmic tail (Env amino acids (aa) 706–718) upstream of the overlap with the tat and rev second coding exons. Changes in Env aa 713 and 715, although they did not affect Env function, abrogated replicative spread. Other amino acid substitutions, i.e., 706–712, 714 and 716, despite their conservation, did not result in defective replicative phenotypes even in primary peripheral blood lymphocytes. Our results point to their involvement in presently unrecognized essential Env functions pertinent only in in vivo. Interestingly, changes in the codons for residues 717–718 as well as some mutations in residues 714–716 abrogated Gag expression but still allowed expression of functional Env in a rev-independent manner. This could be due to the inactivation of a rev-regulated negative element within the respective nucleotide sequence (8354–8368)

    Sr/Ca and δ<sup>18</sup>O in a fast-growing Diploria strigosa coral - evaluation of a new climate archive for the tropical Atlantic

    This study provides the first monthly resolved, 41-year record of geochemical variations (δ18O and Sr/Ca) in a fast-growing Diploria strigosa brain coral from Guadeloupe, Caribbean Sea. Linear regression yields a significant correlation of coral Sr/Ca (δ18O) with instrumental sea surface temperature (SST) on both monthly and mean annual scales (e.g., r = −0.59 for correlation between Simple Ocean Data Assimilation (SODA) SST and Sr/Ca, and r = −0.66 for δ18O; mean annual scale, p < 0.0001). The generated coral Sr/Ca (δ18O)-SST calibration equations are consistent with each other and with published equations using other coral species from different regions. Moreover, a high correlation of coral Sr/Ca and δ18O with local air temperature on a mean annual scale (r = −0.78 for Sr/Ca; r = −0.73 for δ18O; p < 0.0001) demonstrates the applicability of geochemical proxies measured from Diploria strigosa corals as reliable recorders for interannual temperature variability. Both coral proxies are highly correlated with annual and seasonal mean time series of major SST indices in the northern tropical Atlantic (e.g., r = −0.71 for correlation between the index of North Tropical Atlantic SST anomaly and Sr/Ca, and r = −0.70 for δ18O; mean annual scale, p < 0.001). Furthermore, the coral proxies capture the impact of the El Niño–Southern Oscillation on the northern tropical Atlantic during boreal spring. Thus fast-growing Diploria strigosa corals are a promising new archive for the Atlantic Ocean

    Digital twin based design and experimental validation of a continuous peptide polishing step

    Optimizing or debottlenecking existing production plants is a challenging task. In this case study, an existing reversed phased chromatography polishing step for peptide purification was optimized with the help of a digital twin. The existing batch chromatography was depicted digitally with the general rate model. Model parameter determination and model validation was done with dedicated experiments. The digital twin was then used to identify optimized process variants, especially continuous chromatography steps. MCSGP was found to achieve high purities and yield but at the cost of productivity due to column synchronization. An alternative Continuous Twin Column chromatography process (CTCC) was established that eliminates unnecessary waiting times. Ensuring the same or higher purity compared to the batch process, the continuous process achieved a yield increase of 31% and productivity increase of 27.6%. Experimental long runs confirmed these results

    Multivariate parameter determination of multi-component isotherms for chromatography digital twins

    Many fundamental decisions in the process design of a separation task are conducted in an early stage where, unfortunately, process simulation does not have the highest priority. Subsequently, during the setup of the digital twin, dedicated experiments are carried out in the design space that was established earlier. These experiments are most often too complicated to conduct directly. This paper addresses the idea of a combined approach. The early-stage buffer screening and optimization experiments were planned with the Design of Experiments, carried out and then analyzed statistically to extract not only the best buffer composition but also the crucial model parameters, in this case the isotherm dependency on the buffer composition. This allowed the digital twin to predict the best buffer composition, and if the model-predicted control was applied to keep the process at the optimal productivity at a predetermined purity. The methodology was tested with an industrial peptide purification step
