    Rational Design of α-Helix-Stabilized Exendin-4 Analogues

    Exendin-4 (Ex4) is a potent glucagon-like peptide-1 receptor agonist, a drug regulating the plasma glucose level of patients suffering from type 2 diabetes. The molecule’s poor solubility and its readiness to form aggregates increase the likelihood of unwanted side effects. Therefore, we designed Ex4 analogues with improved structural characteristics and better water solubility. Rational design was started from the parent 20-amino acid, well-folded Trp cage (TC) miniprotein and involved the step-by-step N-terminal elongation of the TC head, resulting in the 39-amino acid Ex4 analogue, E19. Helical propensity coupled to tertiary structure compactness was monitored and quantitatively analyzed by electronic circular dichroism and nuclear magnetic resonance (NMR) spectroscopy for the 14 peptides of different lengths. Both 15N relaxation- and diffusion-ordered NMR measurements were established to investigate the inherent mobility and self-association propensity of Ex4 and E19. Our designed E19 molecule has the same tertiary structure as Ex4 but is more helical than Ex4 under all studied conditions; it is less prone to oligomerization and has preserved biological activity. These conditions make E19 a perfect lead compound for further drug discovery. We believe that this structural study improves our understanding of the relationship between local molecular features and global physicochemical properties such as water solubility and could help in the development of more potent Ex4 analogues with improved pharmacokinetic properties

    Testing of Microsatellite Markers for Individual Identification of Fallow Deer

    The fallow deer (Dama dama) of Hungary has excellent value to our country due to its game meat and antler trophies. As an attempt to aid law enforcement against illegal activities, such as poaching, illegal trading, and in other cases like traffic accidents, we aimed to develop a genetic marker set suitable for individual identification. During our research, 28 microsatellite markers on 15 fallow deer samples from two different populations were tested. Four microsatellites were found to be polymorphic, each with two or three alleles. Based on our current results, Hungarian fallow deer populations show low genetic diversity. This is in agreement with previous studies conducted on the species and is probably a direct result of the species’ past extinction from the most of Europe during the Pleistocene and later its human-mediated reintroduction to most of its current range. The low number of polymorphic markers presents the need to include additional markers

    Engineering SERS Properties of Silicon Nanotrees at the Nanoscale

    Large specific surface area nanostructures are desirable in a wide range of sensing applications due to their longer light-trapping path and increased absorption. Engineering of the specific nanotree structure which possesses a high branch density turned out to be challenging from the experimental point of view, and certainly not adequately explored. This paper shows how to design substrates with a silicon nanotree structure for surface-enhanced Raman spectroscopy (SERS) applications. Silicon nanotrees were synthesized by a Ag-Au nanocluster-catalyzed low- pressure chemical vapor deposition method (LPCVD). By the presented approaches, it is possible to manipulate branches’ number, length and thickness. The synthesized nanostructures are flexible after immersion in water which improves SERS performance. The amount of sputtered metal played a key role in preserving the flexibility of the nanotree structure. The obtained substrates with highly fractal nanostructure were tested on 4- mercaptophenylboronic acid (MPBA) to match the optimal SERS parameters. The silicon nanotrees fabrication, and particularly obtained SERS substrates plated with Ag and Au nanoparticles, demonstrated good features and a promising approach for further sensor development

Цель работы–расчет и выбор аппарата для нагрева вакуумного дистиллята легким газойлем. В процессе исследования проведены тепловые, конструктивные и механические расчеты оборудования. Основные конструктивные, технологические и технико-эксплуатационные характеристики теплообменника: диаметр кожуха 1400 мм, длина труб 6 м, диаметр труб 25х2,5 мм, с линзовым компенсатором, четырехходовой, горизонтальный.The object of the study is a shell-and-tube heat exchanger with a lens compensator for heating a vacuum distillate. Vizsgáltuk a terc-amino effektus 2. típusának kiterjesztését a reakcióban résztvevő vinil és terc-amino csoportot két külön gyűrűn tartalmazó (kondenzált) bi- és triaril - bifenil/fenilpiridazin, trifenil/difenilpiridazin, naftalin illetve fenilnaftalin - modellvegyületekre; a gyűrűzárási reakciók révén új azepin-, azocin-, azonin- és azecin-anellált gyűrűrendszereket állítottunk elő. E munkánk révén a terc-amino effektus az eddigi ismeretek szerinti alkalmazását lényegesen bővítettük. 2. Antimaláriás hatásirányban nemzetközi együttműködésben vizsgáltuk azocinszármazékok, illetve indolokinolin alkaloidok bi- és triciklusos analógjainak aktivitását. Ez utóbbiak közül két származék is jelentős in vitro aktivitást mutatott. 3. SSAO hatásirányban vizsgáltuk a terc-amino effektussal nyert diciano-szubsztituált azaheterociklusokból előállított aminometil származékok, illetve további vegyületcsaládok (oxim típusú vegyületek, aminometil-piridazinok) aktivitását; együttműködésben kiválasztott SSAO-gátlók további in vivo vizsgálatára került sor gyulladásos modelleken. E vegyületek egyike (SZV-1287) mind in vitro, mind in vivo jelentős SSAO-gátló és gyulladáscsökkentő hatást fejt ki; fejlesztéséről a közeljövőben döntünk. | 1. Novel extensions of the type 2 tert-amino effect have been studied to compounds having the interacting vinyl and tert-amino moieties on two different rings. Namely, cyclizations of bi- and triaryl - biphenyl/phenylpyridazine, triphenyl/diphenylpyridazine, napthalene and phenylnapthalene - model systems were studied, affording novel azepine-, azocine-, azonine- and azecine-fused ring systems. Our studies facilitate novel applications of the tert-amino effect as compared to cyclizations described thus far. 2. In the frame of an international collaboration, the antimalarial activity of azocine derivatives, and bi- and tricyclic analogues of indoloquinoline alkaloids were studied. Two representatives of the latter group exhibited substantial in vitro activity. 3. SSAO activity of aminomethyl derivatives obtained from dicyano-substituted azaheterocycles synthesized via tert-amino effect cyclization and further classes of compounds (oxime derivatives, aminomethyl-pyridazines) were studied; in collaboration in vivo activity of selected SSAO inhibitors were studied in inflammatory models. On selected derivative (SZV-1287) exhibited substantial in vitro and in vivo SSAO inhibitor and anti-inflammatory activity; we are to decide on its further development

    An Investigation of Surface-Enhanced Raman Scattering of Different Analytes Adsorbed on Gold Nanoislands

    In this study, metallic nanoislands were prepared by thermal annealing of gold thin film produced by vacuum evaporation on a glass substrate to investigate the surface-enhanced Raman scattering (SERS) effect on them. The influence of the analyte on the enhancement factor of SERS was studied with riboflavin and rhodamine 6G dye. Two laser excitation sources at 532 and 633 nm wavelengths were used for SERS measurements. We found that the enhancement factors of the gold nanoisland SERS substrates were influenced by the analytes’ adsorption tendency onto their surfaces. The SERS amplification was also found to be dependent on the electronic structure of the molecules; higher enhancement factors were obtained for rhodamine 6G with 532 nm excitation, while for riboflavin the 633 nm source performed better
