Imaging with a small number of photons
Low-light-level imaging techniques have application in many diverse fields,
ranging from biological sciences to security. We demonstrate a single-photon
imaging system based on a time-gated inten- sified CCD (ICCD) camera in which
the image of an object can be inferred from very few detected photons. We show
that a ghost-imaging configuration, where the image is obtained from photons
that have never interacted with the object, is a useful approach for obtaining
images with high signal-to-noise ratios. The use of heralded single-photons
ensures that the background counts can be virtually eliminated from the
recorded images. By applying techniques of compressed sensing and associated
image reconstruction, we obtain high-quality images of the object from raw data
comprised of fewer than one detected photon per image pixel.Comment: 9 pages, 4 figure
C and N Abundances in Stars At the Base of the Red Giant Branch in M5
We present an analysis of a large sample of moderate resolution Keck LRIS
spectra of subgiant (V \sim 17.2) and fainter stars in the Galactic globular
cluster M5 (NGC 5904) with the goal of deriving C and N abundances.
Star-to-star stochastic variations with significant range in both [C/Fe] and
[N/Fe] are found at all luminosities extending to the bottom of the RGB at M_V
\sim +3. Similar variations in CH appear to be present in the main sequence
turnoff spectra. There is no sign of a change in the behavior of C and N with
evolutionary stage over the full range in luminosity of the RGB and SGB. The C
and N abundances appear strongly anti-correlated, as would be expected from the
CN-cycle processing of stellar material. Yet the present stars are considerably
fainter than the RGB bump, the point at which deep mixing is believed to set
in. On this basis, while the observed abundance pattern is consistent with
proton capture nucleosynthesis, we infer that the site of the reactions is
likely not within the present sample, but rather in a population of more
massive (2 -- 5 M(Sun)) now defunct stars. The range of variation of the N
abundances is very large and the sum of C+N increases as C decreases. To
reproduce this requires the incorporation not only of CN but also of
ON-processed material. Furthermore, the existence of this correlation is quite
difficult to reproduce with an external mechanism such as ``pollution'' with
material processed in a more massive AGB star, which mechanism is fundamentally
stochastic in nature. We therefore suggest that although the internal mixing
hypothesis has serious flaws,new theoretical insights are needed and it should

In Vivo Biotinylation of the Toxoplasma Parasitophorous Vacuole Reveals Novel Dense Granule Proteins Important for Parasite Growth and Pathogenesis.
UnlabelledToxoplasma gondii is an obligate intracellular parasite that invades host cells and replicates within a unique parasitophorous vacuole. To maintain this intracellular niche, the parasite secretes an array of dense granule proteins (GRAs) into the nascent parasitophorous vacuole. These GRAs are believed to play key roles in vacuolar remodeling, nutrient uptake, and immune evasion while the parasite is replicating within the host cell. Despite the central role of GRAs in the Toxoplasma life cycle, only a subset of these proteins have been identified, and many of their roles have not been fully elucidated. In this report, we utilize the promiscuous biotin ligase BirA* to biotinylate GRA proteins secreted into the vacuole and then identify those proteins by affinity purification and mass spectrometry. Using GRA-BirA* fusion proteins as bait, we have identified a large number of known and candidate GRAs and verified localization of 13 novel GRA proteins by endogenous gene tagging. We proceeded to functionally characterize three related GRAs from this group (GRA38, GRA39, and GRA40) by gene knockout. While Δgra38 and Δgra40 parasites showed no altered phenotype, disruption of GRA39 results in slow-growing parasites that contain striking lipid deposits in the parasitophorous vacuole, suggesting a role in lipid regulation that is important for parasite growth. In addition, parasites lacking GRA39 showed dramatically reduced virulence and a lower tissue cyst burden in vivo Together, the findings from this work reveal a partial vacuolar proteome of T. gondii and identify a novel GRA that plays a key role in parasite replication and pathogenesis.ImportanceMost intracellular pathogens reside inside a membrane-bound vacuole within their host cell that is extensively modified by the pathogen to optimize intracellular growth and avoid host defenses. In Toxoplasma, this vacuole is modified by a host of secretory GRA proteins, many of which remain unidentified. Here we demonstrate that in vivo biotinylation of proximal and interacting proteins using the promiscuous biotin ligase BirA* is a powerful approach to rapidly identify vacuolar GRA proteins. We further demonstrate that one factor identified by this approach, GRA39, plays an important role in the ability of the parasite to replicate within its host cell and cause disease

Social Determinants of Health: Underreported Heterogeneity in Systematic Reviews of Caregiver Interventions.
BACKGROUND AND OBJECTIVES:Although most people have some experience as caregivers, the nature and context of care are highly variable. Caregiving, socioeconomic factors, and health are all interrelated. For these reasons, caregiver interventions must consider these factors. This review examines the degree to which caregiver intervention research has reported and considered social determinants of health. RESEARCH DESIGN AND METHODS:We examined published systematic reviews and meta-analyses of interventions for older adults with age-related chronic conditions using the PRISMA and AMSTAR 2 checklists. From 2,707 papers meeting search criteria, we identified 197 potentially relevant systematic reviews, and selected 33 for the final analysis. RESULTS:We found scant information on the inclusion of social determinants; the papers lacked specificity regarding race/ethnicity, gender, sexual identity, socioeconomic status, and geographic location. The majority of studies focused on dementia, with other conditions common in later life vastly underrepresented. DISCUSSION AND IMPLICATIONS:Significant gaps in evidence persist, particularly for interventions targeting diverse conditions and populations. To advance health equity and improve the effectiveness of interventions, research should address caregiver heterogeneity and improve assessment, support, and instruction for diverse populations. Research must identify aspects of heterogeneity that matter in intervention design, while recognizing opportunities for common elements and strategies
