    O desenvolvimento de alimentos funcionais tem aumentado mundialmente devido a seus compostos bioativos que diminuem o risco de doenças e promovem a saúde. Baseado nisso, a indústria e a comunidade científica estão, cada vez mais, buscando ingredientes que possam conferir tais características aos alimentos. A soja, além de ser importante fonte protéica, possui fibras, isoflavonas, oligossacarídeos com potencial prebiótico (rafinose e estaquinose), vitaminas e minerais. As raízes tuberosas de yacon (Smallanthus sonchifolius) possuem grande quantidade de frutooligossacarídeos do tipo inulina, um componente não digerível pelo nosso organismo, com função de auxiliar na seleção de determinadas bactérias da microbiota intestinal, principalmente as bifidobactérias, possuindo, portanto, efeito prebiótico. O objetivo do presente estudo foi o de verificar, através da metodologia da superfície de resposta, as melhores condições para a utilização simultânea de extratos aquosos de soja e de yacon no desenvolvimento de um novo produto fermentado por um cultivo probiótico constituído por Enterococcus faecium CRL 183 e Lactobacillus helveticus ssp jugurti 416. Para tanto, as variáveis independentes foram representadas pelo teor protéico fornecido pelo extrato hidrossolúvel de soja (g/L) e pelo extrato aquoso de yacon (%). As variáveis dependentes (respostas) foram obtidas através do teste de aceitação sensorial (“sabor” e “impressão global”). A otimização conjunta das variáveis apontou a formulação 7 (1,74 g/L de teor protéico fornecido pelo extrato de soja e 25,86% de extrato de yacon) do planejamento experimental como sendo a mais adequada. No entanto, as formulações 9 e 10 (ambas contendo 1,74 g/L de teor protéico fornecido pelo extrato de soja e 40% de extrato de yacon) apresentaram valores de aceitação que não diferiram significativamente...World-wide the development of functional food has been increased, due to its bioactive components which decrease the risk of getting some illnesses and promote health. Based on that, the industry and the scientific population are more and more searching for ingredients that may confer such characteristics to food. The soy, besides being an important proteic source, has fibers and isoflavones, oligosaccharides with potential prebiotic (raffinose and stachyose), vitamins and minerals. The yacon tuber (Smallanthus sonchifolius) is a source of fructooligosaccharides, type inulin, undigestible components by our organism that helps in the selection and development of some bacteria of intestinal microbiota, mainly the bifidobacteria, which means that yacon has prebiotic effect. The aim of this study was to check, through Response Surface Methodology, the optimum conditions to use, simultaneously, the soy and yacon watery extracts in the development of a new fermented product with the probiotic cultive constituted by Enterococcus faecium CRL 183 and Lactobacillus helveticus ssp jugurti 416. Therefore, the independent variables were represented by the protein of soymilk (g/L) and the yacon extract (%). The dependent variables (responses) were obtained through the acceptance test (“taste” and “overall acceptability”). The joint optimization of the variables pointed out the 7th formulation (1.74 g/L of soymilk protein and 25.86% of yacon extract) of the factorial design as the most adequate. However, the 9th and 10th formulations (both containing 1.74 g/L of soymilk protein and 40% of yacon extract) presented acceptance scores significantly equal to the 7th treatment.     Effect of ingestion of soy yogurt on intestinal parameters of rats fed on a beef-based animal diet

    The aim of this study was to investigate whether the ingestion of soy yogurt fermented with Enterococcus faecium CRL 183 would modify the intestinal count of enterococci, fecal pH and ammonia content in rats fed on a diet containing red meat. The rats were placed in 4 groups: for 60 days, group I was given a standard casein-based rodent feed and groups II-IV, the beef-based feed. From day 30, groups III-IV also received the following products: III) soy yogurt; IV) suspension of E. faecium CRL 183. At the start and on days 30 and 60, feces were collected for the determination of pH, ammonia content, count of enterococci and identification of their species. on day 60, rats were sacrificed and their colons also removed for count of enterococci and identification of their species. Rats that ingested soy yogurt showed no significant change (P<0.05) in fecal counts of Enterococcus spp., but, this rat group showed a higher count of E. faecium than rats that ingested suspension of E. faecium CRL 183. The ingestion of soy yogurt and E. faecium culture caused a significant rise (P < 0.05) in fecal pH and ammonia content. Our results suggest that consumption of soy yogurt fermented with E. faecium CRL 183 and L. helveticus subsp. jugurti could change the species of Enterococcus spp. present in the feces and colon of rats fed on a beef-based diet. However, the fermented soy product and the pure culture of E. faecium CRL 183 also induced undesirable effects such as the increase of fecal pH and ammonia content in the feces of rats fed on a beef-based diet.Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP

    ISOFLAVONES SUPPLEMENTATION OF A PROBIOTIC FERMENTED SOY PRODUCT: EFFECTS ON QUALITY CHARACTERISTICS AND ISOFLAVONES PROFILE

    &lt;p align="justify"&gt; The aim of the present study was to investigate the effect of isofl avones supplementation of a fermented soy product on its sensory acceptance, physicochemical properties and probiotic cell viable count. Additionally we also investigated the ability of the mixed starter cultures (Enterococcus faecium CRL 183 and Lactobacillus helveticus 416) to modify the isofl avones profi le of soy product during the fermentation process. Three products were analysed: soy product fermented with E. faecium CRL 183 and L. helveticus 416, isofl avonessupplemented soy product (fermented with E. faecium CRL 183 and L. helveticus 416; 50 mg/100g, Isofl avin®, Galena, Brazil) and unfermented soy product. A panel of judges evaluated the acceptability of the samples on a nine point structured hedonic scale. The chemical composition namely fat, protein, ash and total carbohydrate contents, pH, enumeration of viable Lactobacillus spp. and Enterococcus spp. and quantifi cation of isofl avones using HPLC were investigated. All determinations were conducted after 7 days storage at 10°C. The sensorial acceptance was reduced in the isofl avones-supplemented soy product, but this effect was not signifi cant compared to the sample without isofl avones addition. Chemical composition did not differ (p&lt;0.05) among the samples. Cell viable counts were reduced and total fermentation time was longer in the isofl avonessupplemented soy product, suggesting that the isofl avone addition could inhibit the starter cultures. However, all the products may be considered probiotic since they exhibited lactic acid bacterial populations varying from 2.3 x 109 up to 1.22 x 1010 CFU/mL. Fermentation of soymilk did not change the isofl avones profi le. In conclusion, it was possible to obtain a fermented soy product containing a high isofl avones concentration, adequate sensory and chemical characteristics and lactic acid bacterial viability suffi ciently high to characterize the product as a probiotic.
