    Events and Controversies: Influences of a Shocking News Event on Information Seeking

    It has been suggested that online search and retrieval contributes to the intellectual isolation of users within their preexisting ideologies, where people's prior views are strengthened and alternative viewpoints are infrequently encountered. This so-called "filter bubble" phenomenon has been called out as especially detrimental when it comes to dialog among people on controversial, emotionally charged topics, such as the labeling of genetically modified food, the right to bear arms, the death penalty, and online privacy. We seek to identify and study information-seeking behavior and access to alternative versus reinforcing viewpoints following shocking, emotional, and large-scale news events. We choose for a case study to analyze search and browsing on gun control/rights, a strongly polarizing topic for both citizens and leaders of the United States. We study the period of time preceding and following a mass shooting to understand how its occurrence, follow-on discussions, and debate may have been linked to changes in the patterns of searching and browsing. We employ information-theoretic measures to quantify the diversity of Web domains of interest to users and understand the browsing patterns of users. We use these measures to characterize the influence of news events on these web search and browsing patterns

    Comparative studies of vulva development between Caenorhabditis elegans and other nematode species have provided some insight into the evolution of patterning networks. However, molecular genetic details are available only in C. elegans and Pristionchus pacificus. To extend our knowledge on the evolution of patterning networks, we studied the C. elegans male hook competence group (HCG), an equivalence group that has similar developmental origins to the vulval precursor cells (VPCs), which generate the vulva in the hermaphrodite. Similar to VPC fate specification, each HCG cell adopts one of three fates (1°, 2°, 3°), and 2° HCG fate specification is mediated by LIN-12/Notch. We show that 2° HCG specification depends on the presence of a cell with the 1° fate. We also provide evidence that Wnt signaling via the Frizzled-like Wnt receptor LIN-17 acts to specify the 1° and 2° HCG fate. A requirement for EGF signaling during 1° fate specification is seen only when LIN-17 activity is compromised. In addition, activation of the EGF pathway decreases dependence on LIN-17 and causes ectopic hook development. Our results suggest that WNT plays a more significant role than EGF signaling in specifying HCG fates, whereas in VPC specification EGF signaling is the major inductive signal. Nonetheless, the overall logic is similar in the VPCs and the HCG: EGF and/or WNT induce a 1° lineage, and LIN-12/NOTCH induces a 2° lineage. Wnt signaling is also required for execution of the 1° and 2° HCG lineages. lin-17 and bar-1/β-catenin are preferentially expressed in the presumptive 1° cell P11.p.     Search and breast cancer: On disruptive shifts of attention over life histories of an illness

    ABSTRACT We seek to understand the evolving needs of people who are faced with a life-changing medical diagnosis based on analyses of queries extracted from an anonymized search query log. Focusing on breast cancer, we manually tag a set of Web searchers as showing disruptive shifts in focus of attention and long-term patterns of search behavior consistent with the diagnosis and treatment of breast cancer. We build and apply probabilistic classifiers to detect these searchers from multiple sessions and to detect the timing of diagnosis, using a variety of temporal and statistical features. We explore the changes in information-seeking over time before and after an inferred diagnosis of breast cancer by aligning multiple searchers by the likely time of diagnosis. We automatically identify 1700 candidate searchers with an estimated 90% precision, and we predict the day of diagnosis within 15 days with an 88% accuracy. We show that the geographic and demographic attributes of searchers identified with high probability are strongly correlated with ground truth of reported incidence rates. We then analyze the content of queries over time from searchers for whom diagnosis was predicted, using a detailed ontology of cancerrelated search terms. Our analysis reveals the rich temporal structure of the evolving queries of people likely diagnosed with breast cancer. Finally, we focus on subtypes of illness based on inferred stages of cancer and show clinically relevant dynamics of information seeking based on dominant stage expressed by searchers

    The LIN-15A and LIN-56 Transcriptional Regulators Interact to Negatively Regulate EGF/Ras Signaling in Caenorhabditis elegans Vulval Cell-Fate Determination

    The restricted expression of epidermal growth factor (EGF) family ligands is important for proper development and for preventing cancerous growth in mammals. In Caenorhabditis elegans, the class A and B synthetic multivulva (synMuv) genes redundantly repress expression of lin-3 EGF to negatively regulate Ras-mediated vulval development. The class B synMuv genes encode proteins homologous to components of the NuRD and Myb-MuvB/dREAM transcriptional repressor complexes, indicating that they likely silence lin-3 EGF through chromatin remodeling. The two class A synMuv genes cloned thus far, lin-8 and lin-15A, both encode novel proteins. The LIN-8 protein is nuclear. We have characterized the class A synMuv gene lin-56 and found it to encode a novel protein that shares a THAP-like C_2CH motif with LIN-15A. Both the LIN-56 and LIN-15A proteins localize to nuclei. Wild-type levels of LIN-56 require LIN-15A, and wild-type levels and/or localization of LIN-15A requires LIN-56. Furthermore, LIN-56 and LIN-15A interact in the yeast two-hybrid system. We propose that LIN-56 and LIN-15A associate in a nuclear complex that inhibits vulval specification by repressing lin-3 EGF expression
