    GRACE Measurements of Mass Variability in the Earth System

    Monthly gravity field estimates made by the twin Gravity Recovery and Climate Experiment (GRACE) satellites have a geoid height accuracy of 2 to 3 millimeters at a spatial resolution as small as 400 kilometers. The annual cycle in the geoid variations, up to 10 millimeters in some regions, peaked predominantly in the spring and fall seasons. Geoid variations observed over South America that can be largely attributed to surface water and groundwater changes show a clear separation between the large Amazon watershed and the smaller watersheds to the north. Such observations will help hydrologists to connect processes at traditional length scales (tens of kilometers or less) to those at regional and global scales

    Measuring and Correcting Wind-Induced Pointing Errors of the Green Bank Telescope Using an Optical Quadrant Detector

    Wind-induced pointing errors are a serious concern for large-aperture high-frequency radio telescopes. In this paper, we describe the implementation of an optical quadrant detector instrument that can detect and provide a correction signal for wind-induced pointing errors on the 100m diameter Green Bank Telescope (GBT). The instrument was calibrated using a combination of astronomical measurements and metrology. We find that the main wind-induced pointing errors on time scales of minutes are caused by the feedarm being blown along the direction of the wind vector. We also find that wind-induced structural excitation is virtually non-existent. We have implemented offline software to apply pointing corrections to the data from imaging instruments such as the MUSTANG 3.3 mm bolometer array, which can recover ~70% of sensitivity lost due to wind-induced pointing errors. We have also performed preliminary tests that show great promise for correcting these pointing errors in real-time using the telescope's subreflector servo system in combination with the quadrant detector signal.Comment: 17 pages, 11 figures; accepted for publication in PAS

    Holographic Measurement and Improvement of the Green Bank Telescope Surface

    We describe the successful design, implementation, and operation of a 12 GHz holography system installed on the Robert C. Byrd Green Bank Telescope (GBT). We have used a geostationary satellite beacon to construct high-resolution holographic images of the telescope mirror surface irregularities. These images have allowed us to infer and apply improved position offsets for the 2209 actuators which control the active surface of the primary mirror, thereby achieving a dramatic reduction in the total surface error (from 390 microns to ~240 microns, rms). We have also performed manual adjustments of the corner offsets for a few panels. The expected improvement in the radiometric aperture efficiency has been rigorously modeled and confirmed at 43 GHz and 90 GHz. The improvement in the telescope beam pattern has also been measured at 11.7 GHz with greater than 60 dB of dynamic range. Symmetric features in the beam pattern have emerged which are consistent with a repetitive pattern in the aperture due to systematic panel distortions. By computing average images for each tier of panels from the holography images, we confirm that the magnitude and direction of the panel distortions, in response to the combination of gravity and thermal gradients, are in general agreement with finite-element model predictions. The holography system is now fully integrated into the GBT control system, and by enabling the telescope staff to monitor the health of the individual actuators, it continues to be an essential tool to support high-frequency observations.Comment: Accepted for publication in PASP.     Low field vortex dynamics over seven time decades in a Bi_2Sr_2CaCu_2O_{8+\delta} single crystal for temperatures 13 K < T < 83 K

    Using a custom made dc-SQUID magnetometer, we have measured the time relaxation of the remanent magnetization M_rem of a Bi_2Sr_2CaCu_2O_{8+\delta} single crystal from the fully critical state for temperatures 13 K < T < 83 K. The measurements cover a time window of seven decades 10^{-2} s < t < 10^5 s, so that the current density j can be studied from values very close to j_c down to values considerably smaller than j_c. From the data we have obtained: (i) the flux creep activation barriers U as a function of current density j, (ii) the current-voltage characteristics E(j) in a typical range of 10^{-7} V/cm to 10^{-15} V/cm, and (iii) the critical current density j_c(0) at T = 0. Three different regimes of vortex dynamics are observed: For temperatures T < 20 K the activation barrier U(j) is logarithmic, no unique functional dependence U(j) could be found for the intermediate temperature interval 20 K < T < 40 K, and finally for T > 40 K the activation barrier U(j) follows a power-law behavior with an exponent mu = 0.6. From the analysis of the data within the weak collective pinning theory for strongly layered superconductors, it is argued that for temperatures T < 20 K pancake-vortices are pinned individually, while for temperatures T > 40 K pinning involves large collectively pinned vortex bundles. A description of the vortex dynamics in the intermediate temperature interval 20 K < T < 40 K is given on the basis of a qualitative low field phase diagram of the vortex state in Bi_2Sr_2CaCu_2O_{8+\delta}. Within this description a second peak in the magnetization loop should occur for temperatures between 20 K and 40 K, as it has been observed in several magnetization measurements in the literature.Comment: 12 pages, 10 figure

    Implementation of population screening for colorectal cancer by repeated fecal occult blood test in the Netherlands

    <p>Abstract</p> <p>Background</p> <p>Colorectal cancer (CRC) is the third most prevalent type of cancer in the world. Its prognosis is closely related to the disease stage at the time of diagnosis. Early detection of symptomless CRC or precursor lesions through population screening could reduce CRC mortality. However, screening programs are only effective if enough people are willing to participate. This study aims to asses the uptake of a second round of fecal occult blood test (FOBt) based screening and to explore factors that could potentially increase this uptake.</p> <p>Methods and design</p> <p>Two years after the first screening round, 10.000 average risk persons, aged 50 to 75, will again receive an invitation to participate in immunohistochemical FOBt (iFOBt) based screening. Eligible persons will be recruited through a city population database. Invitees will be randomized to receive either an iFOBt with a faeces collection paper or an iFOBt without a collection paper. The iFOBts will be analyzed in a specialized laboratory at the Academic Medical Centre. Positive iFOBts will be followed by a consultation at our outpatient clinic and, in the absence of contra-indications and after informed consent, by a colonoscopy. The primary outcome measure is the participation rate. Secondary outcome measures are the effect of the addition of a collection paper on the participation rate, reasons for participation and non-participation, measures of informed choice and psychological consequences of screening and measures of psychological and physical burden associated with the iFOBt and the colonoscopy. Another secondary outcome measure is the diagnostic yield of the program.</p> <p>Discussion</p> <p>In order to implement population screening for colorectal cancer in the Netherlands, information is needed on the uptake of repeated rounds of FOBt-based screening and on factors that could potentially increase this uptake in the future since effectiveness of such a program depends on the willingness of persons to participate. This study will provide information on the actual uptake and perception of a second round of iFOBt-based screening. The results of this study will contribute to the future implementation of a national colorectal screening program in the Netherlands.</p> <p>Trial registration</p> <p>Dutch Trial register: NTR1327</p

    PAX2 Regulates ADAM10 Expression and Mediates Anchorage-Independent Cell Growth of Melanoma Cells

    PAX transcription factors play an important role during development and carcinogenesis. In this study, we investigated PAX2 protein levels in melanocytes and melanoma cells by Western Blot and immunofluorescence analysis and characterized the role of PAX2 in the pathogenesis of melanoma. In vitro we found weak PAX2 protein expression in keratinocytes and melanocytes. Compared to melanocytes increased PAX2 protein levels were detectable in melanoma cell lines. Interestingly, in tissue sections of melanoma patients nuclear PAX2 expression strongly correlated with nuclear atypia and the degree of prominent nucleoli, indicating an association of PAX2 with a more atypical cellular phenotype. In addition, with chromatin immunoprecipitation assay, PAX2 overexpression and PAX2 siRNA we present compelling evidence that PAX2 can regulate ADAM10 expression, a metalloproteinase known to play important roles in melanoma metastasis. In human tissue samples we found co-expression of PAX2 and ADAM10 in melanocytes of benign nevi and in melanoma cells of patients with malignant melanoma. Importantly, the downregulation of PAX2 by specific siRNA inhibited the anchorage independent cell growth and decreased the migratory and invasive capacity of melanoma cells. Furthermore, the downregulation of PAX2 abrogated the chemoresistance of melanoma cells against cisplatin, indicating that PAX2 expression mediates cell survival and plays important roles during melanoma progression
