    Innovative interstellar explorer

    An interstellar "precursor" mission has been under discussion in the scientific community for at least 30 years. Fundamental scientific questions about the interaction of the Sun with the interstellar medium can only be answered with in situ measurements that such a mission can provide. The Innovative Interstellar Explorer (IIE) and its use of Radioisotope Electric Propulsion (REP) is being studied under a NASA "Vision Mission" grant. Speed is provided by a combination of a high-energy launch, using current launch vehicle technology, a Jupiter gravity assist, and long-term, low-thrust, continuous acceleration provided by an ion thruster running off electricity provided by advanced radioisotope electric generators. A payload of ten instruments with an aggregate mass of ~35 kg and requiring ~30 W has been carefully chosen to address the compelling science questions. The nominal 20-day launch window opens on 22 October 2014 followed by a Jupiter gravity assist on 5 February 2016. The REP system accelerates the spacecraft to a "burnout" speed of 7.8 AU per year at 104 AU on 13 October 2032 (Voyager 1's current speed is ~3.6 AU/yr). The spacecraft will return at least 500 bits per second from at least 200 AU ~30 years after launch. Additional (backup) launch opportunities occur every 13 months to early 2018. In addition to addressing basic heliospheric science, the mission will ensure continued information on the far-heliospheric galactic cosmic ray population after the Voyagers have fallen silent and as the era of human Mars exploration begins

    Neuromuscular recovery from botulism involves multiple forms of compensatory plasticity

    IntroductionBotulinum neurotoxin (BoNT) causes neuroparalytic disease and death by blocking neuromuscular transmission. There are no specific therapies for clinical botulism and the only treatment option is supportive care until neuromuscular function spontaneously recovers, which can take weeks or months after exposure. The highly specialized neuromuscular junction (NMJ) between phrenic motor neurons and diaphragm muscle fibers is the main clinical target of BoNT. Due to the difficulty in eliciting respiratory paralysis without a high mortality rate, few studies have characterized the neurophysiological mechanisms involved in diaphragm recovery from intoxication. Here, we develop a mouse model of botulism that involves partial paralysis of respiratory muscles with low mortality rates, allowing for longitudinal analysis of recovery.Methods and resultsMice challenged by systemic administration of 0.7 LD50 BoNT/A developed physiological signs of botulism, such as respiratory depression and reduced voluntary running activity, that persisted for an average of 8–12 d. Studies in isolated hemidiaphragm preparations from intoxicated mice revealed profound reductions in nerve-elicited, tetanic and twitch muscle contraction strengths that recovered to baseline 21 d after intoxication. Despite apparent functional recovery, neurophysiological parameters remained depressed for 28 d, including end plate potential (EPP) amplitude, EPP success rate, quantal content (QC), and miniature EPP (mEPP) frequency. However, QC recovered more quickly than mEPP frequency, which could explain the discrepancy between muscle function studies and neurophysiological recordings. Hypothesizing that differential modulation of voltage-gated calcium channels (VGCC) contributed to the uncoupling of QC from mEPP frequency, pharmacological inhibition studies were used to study the contributions of different VGCCs to neurophysiological function. We found that N-type VGCC and P/Q-type VGCC partially restored QC but not mEPP frequency during recovery from paralysis, potentially explaining the accelerated recovery of evoked release versus spontaneous release. We identified additional changes that presumably compensate for reduced acetylcholine release during recovery, including increased depolarization of muscle fiber resting membrane potential and increased quantal size.DiscussionIn addition to identifying multiple forms of compensatory plasticity that occur in response to reduced NMJ function, it is expected that insights into the molecular mechanisms involved in recovery from neuromuscular paralysis will support new host-targeted treatments for multiple neuromuscular diseases

    Correlations between SO2 flux, seismicity, and outgassing activity at the open vent of Villarrica volcano, Chile

    The characteristics of the open vent activity of Villarrica volcano, Chile, were studied in detail by integrating visual observations of the lava lake, analysis of the seismic tremor, and measurements of SO2 flux. The outgassing activity comprises a persistent gas plume emission from the bottom of the crater as well as frequent explosive events. Three main styles of bubble bursting were identified at the surface of the active lava lake: seething magma, small short-lived lava fountains, and Strombolian explosions. Seething magma consists of continual burst of relatively small bubbles (a few meters in diameter) with varying strength over the entire surface of the lava lake. Small lava fountains, seen as a vigorous extension of seething magma, commonly have durations of 20–120 s and reach 10–40 m high above the lava lake. Correlations between seismicity and visual observations indicate that the seismic tremor is mostly caused by the explosive outgassing activity. Furthermore, for different periods between 2000 and 2006, during which the activity remained comparable, the real-time seismic amplitude measurement system (RSAM) and SO2 emission rates show a very good correlation. Higher SO2 emissions appeared to be related to higher levels of the lava lake, stronger bubble bursting activity, and changes in the morphology and texture of the crater floor. Background (low) levels of activity correspond to a lava lake located >80 m below the crater rim, small and/or blocky morphology of the roof, seismic amplitude (RSAM) lower than 25 units, few volcano-tectonic earthquakes, and daily averages of SO2 emissions lower than 600 Mg/d
