    Gravity-driven draining of a thin rivulet with constant width down a slowly varying substrate

    The locally unidirectional gravity-driven draining of a thin rivulet with constant width but slowly varying contact angle down a slowly varying substrate is considered. Specifically, the flow of a rivulet in the azimuthal direction from the top to the bottom of a large horizontal cylinder is investigated. In particular, it is shown that, despite behaving the same locally, this flow has qualitatively different global behaviour from that of a rivulet with constant contact angle but slowly varying width. For example, whereas in the case of constant contact angle there is always a rivulet that runs all the way from the top to the bottom of the cylinder, in the case of constant width this is possible only for sufficiently narrow rivulets. Wider rivulets with constant width are possible only between the top of the cylinder and a critical azimuthal angle on the lower half of the cylinder. Assuming that the contact lines de-pin at this critical angle (where the contact angle is zero) the rivulet runs from the critical angle to the bottom of the cylinder with zero contact angle, monotonically decreasing width and monotonically increasing maximum thickness. The total mass of fluid on the cylinder is found to be a monotonically increasing function of the value of the constant width

    Turbofan forced mixer lobe flow modeling. 1: Experimental and analytical assessment

    A joint analytical and experimental investigation of three-dimensional flowfield development within the lobe region of turbofan forced mixer nozzles is described. The objective was to develop a method for predicting the lobe exit flowfield. In the analytical approach, a linearized inviscid aerodynamical theory was used for representing the axial and secondary flows within the three-dimensional convoluted mixer lobes and three-dimensional boundary layer analysis was applied thereafter to account for viscous effects. The experimental phase of the program employed three planar mixer lobe models having different waveform shapes and lobe heights for which detailed measurements were made of the three-dimensional velocity field and total pressure field at the lobe exit plane. Velocity data was obtained using Laser Doppler Velocimetry (LDV) and total pressure probing and hot wire anemometry were employed to define exit plane total pressure and boundary layer development. Comparison of data and analysis was performed to assess analytical model prediction accuracy. As a result of this study a planar mixed geometry analysis was developed. A principal conclusion is that the global mixer lobe flowfield is inviscid and can be predicted from an inviscid analysis and Kutta condition

    A thin rivulet or ridge subject to a uniform transverse shear stress at its free surface due to an external airflow

    We use the lubrication approximation to analyse three closely related problems involving a thin rivulet or ridge (i.e. a two-dimensional droplet) of fluid subject to a prescribed uniform transverse shear stress at its free surface due to an external airflow, namely a rivulet draining under gravity down a vertical substrate, a rivulet driven by a longitudinal shear stress at its free surface, and a ridge on a horizontal substrate, and find qualitatively similar behaviour for all three problems. We show that, in agreement with previous numerical studies, the free surface profile of an equilibrium rivulet/ridge with pinned contact lines is skewed as the shear stress is increased from zero, and that there is a maximum value of the shear stress beyond which no solution with prescribed semi-width is possible. In practice, one or both of the contact lines will de-pin before this maximum value of the shear stress is reached, and so we consider situations in which the rivulet/ridge de-pins at one or both contact lines. In the case of de-pinning only at the advancing contact line, the rivulet/ridge is flattened and widened as the shear stress is increased from its critical value, and there is a second maximum value of the shear stress beyond which no solution with a prescribed advancing contact angle is possible. In contrast, in the case of de-pinning only at the receding contact line, the rivulet/ridge is thickened and narrowed as the shear stress is increased from its critical value, and there is a solution with a prescribed receding contact angle for all values of the shear stress. In general, in the case of de-pinning at both contact lines there is a critical “yield” value of the shear stress beyond which no equilibrium solution is possible and the rivulet/ridge will evolve unsteadily. In an Appendix we show that an equilibrium rivulet/ridge with prescribed flux/area is quasi-statically stable to two-dimensional perturbations

    GABA-B receptor function in healthy volunteers, a pharmacokinetic and pharmacodynamic study of two doses of baclofen compared to placebo

    AIMS AND HYPOTHESIS To assess the subjective and objective effects of baclofen on brain function in healthy volunteers. BACKGROUND Recent evidence suggests baclofen, a γ-aminobutyric acid type B (GABA-B) receptor agonist, reduces alcohol consumption and craving and promotes abstinence in alcoholics. However, characterisation of the GABA-B receptor system in clinical addiction is limited, and it is unclear why some patients require, or tolerate, higher doses to treat alcoholism. This study assesses the effects of baclofen on brain function in healthy volunteers to inform future studies investigating the sensitivity of GABA-B receptors in alcohol addiction. METHODS Eight healthy male volunteers completed a double blind randomised 3-way cross over study, receiving oral placebo (vitamin C 100mg), 10mg and 60mg baclofen. Subjective and objective measurements were taken at baseline (before medication) and at +30mins, 1, 2, 3, 4 and 6 hours after dosing. Objective measures included blood plasma samples, heart rate and blood pressure. Subjective measures included; the Subjective High Assessment Questionnaire (SHAS), visual analogue scales for sleepy, relaxed, tense and alert and a motor coordination task (zig-zag task). Pharmacokinetic data was obtained using liquid chromatography mass-spectrometry (LC-MS) to measure plasma baclofen concentrations. RESULTS 60mg Baclofen showed changes in subjective measures peaking at 2 hours post dosing compared with placebo, including a significant increase (p<0.05) in total SHAS scores with individual items, including feeling ‘drunk or intoxicated’, effects of alcohol and ‘muddled or confused’ particular affected.. Systolic blood pressure was significantly increased (p<0.05) at the 2 hours post 60mg dose. For both 10mg and 60mg baclofen, peak plasma concentration was achieved 60 minutes post dose. Pharmacokinetic data will be presented. There were no significant changes in these measures between 10mg Baclofen and placebo. CONCLUSIONS The objective and subjective measures used in this study are able to differentiate between placebo and 60mg baclofen. These findings will inform further research investigating the sensitivity of GABA-B receptors in alcohol addiction

    Validated method for the quantification of buprenorphine in postmortem blood using solid-phase extraction and two-dimensional gas chromatography–mass spectrometry

    A highly sensitive and fully validated method was developed for the quantification of buprenorphine in postmortem blood. After a two-step protein precipitation process using acetonitrile, buprenorphine was purified using mixed-mode (C8/cation exchange) solid-phase extraction cartridges. Endogenous water-soluble compounds and lipids were removed from the cartridges before the samples were eluted, concentrated and derivatized using N-methyl-N-trimethylsilyltrifluoroacetamide. The samples were analyzed using two-dimensional gas chromatography–mass spectrometry (2D GC–MS) in selective ion-monitoring mode. A low polarity RxiÂź-5MS (30 m × 0.25 mm I.D. × 0.25 ”m) was used as the primary column and the secondary column was a mid-polarity RxiÂź -17Sil MS (15 m × 0.32 mm I.D. × 0.25 ”m). The assay was linear from 1.0 to 50.0 ng/mL (r2 > 0.99; n = 6). Intraday (n = 6) and interday (n = 9) imprecisions (percentage relative standard deviation, % RSD) were <5% and the average recovery was 60%. The limit of detection (LOD) of the method was 0.5 ng/mL and limit of quantification was 1.0 ng/mL. 2D GC–MS improved the LOD of buprenorphine by 20-fold compared with analysis on a conventional GC–MS. The method was highly selective with no interference from endogenous compounds or from 62 commonly encountered drugs.     Zearalenone production and growth in drinking water inoculated with Fusarium graminearum

    Production of the mycotoxin zearalenone (ZEN) was examined in drinking water inoculated with Fusarium graminearum. The strain employed was isolated from a US water distribution system. ZEN was purified with an immunoaffinity column and quantified by high-performance liquid chromatography (HPLC) with fluorescence detection. The extracellular yield of ZEN was 15.0 ng l−1. Visual growth was observed. Ergosterol was also indicative of growth and an average of 6.2 ÎŒg l−1 was obtained. Other compounds were also detected although remain unidentified. There is no equivalent information available. More work is required on metabolite expression in water as mycotoxins have consequences for human and animal health. The levels detected in this study were low. Water needs to be accepted as a potential source as it attracts high quality demands in terms of purity.Fundação para a CiĂȘncia e a Tecnologia (FCT
