    Online Gaming Traffic Generator for Reproducing Gamer Behavior

    International audienceIn this paper, we proposed an online gaming traffic generator reflecting user behavior patterns. We analyzed the packet size and inter departure time distributions of a popular FPS game (Left4Dead) and MMORPG (World of Warcraft) for regenerating gaming traffic. The proposed traffic generator generates an inter departure time and gaming packetbased on analytical model of the gamer behaviors, then transmits the packet according to the inter departure time. Packet generation results show that generated packets of World of Warcraft is much different with analytical model, unlike Left4Dead. It is caused by Nagle algorithm and Delayed Acknowledgments of TCP. Thus, we disabled the Nagle algorithm in the proposed traffic generator. The generation results show that the revised proposed traffic generator guarantees goodness of fit in the generated traffic distribution

    A Wideband Digital RF Receiver Front-end Employing a New Discrete-time Filter for m-WiMAX

    A wideband digital RF receiver front-end employing a discrete-time (DT) filter is presented for application to m-WiMAX (WiBro). By employing a sampling mixer and a DT filter, the receiver operates in the charge domain. In addition to the flexibility of the DT filter, the new non-decimation finite impulse response (NDF) filter can be cascaded to a conventional finite impulse response (FIR) filter without the decimation effect. Thus, we can easily increase the order of the sinc(n) function-type filtering response and the signal processing bandwidth. The FIR filter is also modified to reduce the noise and the number of required clock signal. Because of the new filter configuration, clock signals can be shared by the FIR filter and NDF filter and the clock generator circuit can be simplified. The designed receiver front-end is implemented using an IBM 0.13-mu m RF CMOS process. The fabricated chip satisfies the m-WiMAX specification of an 8.75 MHz channel bandwidth and the total system current dissipation is 26.63 mA from a 1.5-V supply voltage.X116sciescopu

    Optimization of primer sets and detection protocols for SARS-CoV-2 of coronavirus disease 2019 (COVID-19) using PCR and real-time PCR

    © 2020, The Author(s). SARS-CoV-2 is very contagious and has rapidly spread globally. Due to various symptomatic and asymptomatic cases and the possibility of asymptomatic transmission, there is a pressing need for a fast and sensitive detection protocol to diagnose asymptomatic people. Various SARS-CoV-2 diagnostic kits are already available from many companies and national health agencies. However, publicly available information on these diagnostic kits is lacking. In response to the growing need and the lack of information, we developed and made available a low-cost, easy-access, real-time PCR-based protocol for the early detection of the virus in a previous study. During the development of the detection protocol, we found that unoptimized primer sets could inadvertently show false-positive results, raising the possibility that commercially available diagnostic kits might also contain primer sets that produce false-positive results. Here, we provide three-step guidelines for the design and optimization of specific primer sets. The three steps include (1) the selection of primer sets for target genes (RdRP,N,E, andS) in the genome of interest (SARS-CoV-2), (2) the in silico validation of primer and amplicon sequences, and (3) the optimization of PCR conditions (i.e., primer concentrations and annealing temperatures) for specific hybridization between the primers and target genes, and the elimination of spurious primer dimers. Furthermore, we have expanded the previously developed real-time PCR-based protocol to more conventional PCR-based protocols and applied a multiplex PCR-based protocol that allows the simultaneous testing of primer sets forRdRP,N,E, andSall in one reaction. Our newly optimized protocol should be helpful for the large-scale, high-fidelity screening of asymptomatic people, even without any high-specification equipment, for the further prevention of transmission, and to achieve early intervention and treatment for the rapidly propagating virus.     Inhibitors of synaptic vesicle exocytosis reduce surface expression of postsynaptic glutamate receptors

    Bafilomycin A1, a vacuolar H+-ATPase inhibitor, and botulinum toxin B and tetanus toxin, both vesicle fusion inhibitors, are widely known exocytosis blockers that have been used to inhibit the presynaptic release of neurotransmitters. However, protein trafficking mechanisms, such as the insertion of postsynaptic receptors and astrocytic glutamate-releasing channels into the plasma membrane, also require exocytosis. In our previous study, exocytosis inhibitors reduced the surface expression of astrocytic glutamate-releasing channels. Here, we further investigated whether exocytosis inhibitors influence the surface expression of postsynaptic receptors. Using pH-sensitive superecliptic pHluorin (SEP)-tagged postsynaptic glutamate receptors, including GluA1, GluA2, GluN1, and GluN2A, we found that bafilomycin A1, botulinum toxin B, and/or tetanus toxin reduce the SEP fluorescence of SEP-GluA1, SEP-GluA2, SEP-GluN1, and SEP-GluN2A. These findings indicate that presynaptic vesicle exocytosis inhibitors also affect the postsynaptic trafficking machinery for surface expression. Finally, this study provides profound insights assembling presynaptic, postsynaptic and astrocytic viewpoints into the interpretation of the data obtained using these synaptic vesicle exocytosis inhibitors11Nsciescopuskc
