    Transition from confined to bulk dynamics in symmetric star-linear polymer mixtures

    We report on the linear viscoelastic properties of mixtures comprising multiarm star (as model soft colloids) and long linear chain homopolymers in a good solvent. In contrast to earlier works, we investigated symmetric mixtures (with a size ratio of 1) and showed that the polymeric and colloidal responses can be decoupled. The adopted experimental protocol involved probing the linear chain dynamics in different star environments. To this end, we studied mixtures with different star mass fraction, which was kept constant while linear chains were added and their entanglement plateau modulus (GpG_p) and terminal relaxation time (Ï„d\tau_d) were measured as functions of their concentration. Two distinct scaling regimes were observed for both GpG_p and Ï„d\tau_d: at low linear polymer concentrations, a weak concentration dependence was observed, that became even weaker as the fraction of stars in the mixtures increased into the star glassy regime. On the other hand, at higher linear polymer concentrations, the classical entangled polymer scaling was recovered. Simple scaling arguments show that the threshold crossover concentration between the two regimes corresponds to the maximum osmotic star compression and signals the transition from confined to bulk dynamics. These results provide the needed ingredients to complete the state diagram of soft colloid-polymer mixtures and investigate their dynamics at large polymer-colloid size ratios. They also offer an alternative way to explore aspects of the colloidal glass transition and the polymer dynamics in confinement.     Cell Propagation of Cholera Toxin CTA ADP-Ribosylating Factor by Exosome Mediated Transfer

    In this study, we report how the cholera toxin (CT) A subunit (CTA), the enzyme moiety responsible for signaling alteration in host cells, enters the exosomal pathway, secretes extracellularly, transmits itself to a cell population. The first evidence for long-term transmission of CT's toxic effect via extracellular vesicles was obtained in Chinese hamster ovary (CHO) cells. To follow the CT intracellular route towards exosome secretion, we used a novel strategy for generating metabolically-labeled fluorescent exosomes that can be counted by flow cytometry assay (FACS) and characterized. Our results clearly show the association of CT with exosomes, together with the heat shock protein 90 (HSP90) and Protein Disulfide Isomerase (PDI) molecules, proteins required for translocation of CTA across the ER membrane into the cytoplasm. Confocal microscopy showed direct internalization of CT containing fluorescent exo into CHO cells coupled with morphological changes in the recipient cells that are characteristic of CT action. Moreover, Me665 cells treated with CT-containing exosomes showed an increase in Adenosine 3',5'-Cyclic Monophosphate (cAMP) level, reaching levels comparable to those seen in cells exposed directly to CT. Our results prompt the idea that CT can exploit an exosome-mediated cell communication pathway to extend its pathophysiological action beyond an initial host cell, into a multitude of cells. This finding could have implications for cholera disease pathogenesis and epidemiology

    Severe Kawasaki disease in a 3-month-old patient: a case report

    BACKGROUND: Kawasaki disease is a multi-system vasculitis which usually occurs in children under 5 years of age. In infants under three months of age, it is very rare and usually associated with a high incidence of incomplete or atypical forms, often unresponsive to treatment. This condition increases the risk of cardiovascular complications such as coronary artery aneurysms. CASE PRESENTATION: We describe a 3-month-old infant who developed early and severe aneurysms in three coronary arteries despite a timely administration of intravenous immunoglobulins, followed by three days of intravenous methylprednisolone. CONCLUSION: This case report underlines that the development of coronary artery aneurysm correlates with a delayed diagnosis and treatment, incomplete or atypical forms of the disease, and additionally the severity of clinical presentation, especially in cases of very young infants below 3 months of age. Our case is notable because of the very young age of the patient, the severity of clinical presentation with an early development of coronary artery aneurysms and the unresponsiveness to the therapy

    Effect of dietary supplementation of a mix of chestnut and quebracho tannins on intestinal morphology, bacterial load, Eimeria spp oocyst excretion and immune response after vaccination in rabbits

    Among plant products, tannins exhibited a strong antioxidant activity and their employment was studied as food or feed addictive with positive effects. Nevertheless, their health-enhancing properties remain to be completely clarified, particularly in rabbits. The aim of the present study was to investigate the influence of a mix of chestnut and quebracho tannins as supplementation in rabbit diet on intestinal morphology, as well as bacterial loads and Eimeria spp coccidial oocyst excretion and humoral immune response after vaccination. Rabbits were fed four different diets: basal diet (negative control, group CN), basal diet with anticoccidial supplementation (positive control, group CP) and basal diet with tannins mix 0.3% (group T0.3) or 0.6% (group T0.6). Samples of intestine, liver, spleen thymus, mesenteric lymph nodes, kidney and heart were submitted to histopathologic investigations. Morphometric analyses were performed on portion of duodenum, jejunum and ileum. Intestinal contents were analyzed by flotation test for Eimeria spp. oocysts excretion and by microbiological assays for Clostridium perfringens and Escherichia coli. Rabbits belonging to the four experimental groups were vaccinated by means of a live attenuated myxomatosis virus and sera were collected individually at three time points to determine antibody response.The obtained results demonstratedthe absence of toxic effects of the tannin mix on examined organs, particularly on liver and kidney and any negative influence on the growth performance. A relevant anti-inflammatory effect on the intestinal tract, a reduction of Eimeria spp. oocysts excretion and a positive immunomodulatory and antibacterial effects were also observed.
