    Expression and Structural Studies of Fasciclin I, an Insect Cell Adhesion Molecule

    Fasciclin I is a lipid-linked cell-surface glycoprotein that can act as a homophilic adhesion molecule in tissue culture cells. It is thought to be involved in growth cone guidance in the embryonic insect nervous system. To facilitate structure-function studies, we have generated Chinese hamster ovary (CHO) cell lines expressing high levels of cell surface grasshopper and Drosophila fasciclin I. Grasshopper fasciclin I released by phospholipase C cleavage was purified on an immunoaffinity column and single crystals were obtained that diffracted to approximately 5-A resolution. We also generated CHO and Drosophila S2 cell lines that produce a secreted form of fasciclin I. Fasciclin I expressed in S2 cells contains significantly less carbohydrate than the protein expressed in CHO cells, and may therefore be more suitable for crystallization. Biochemical characterization of purified fasciclin I indicates that the extracellular portion exists as a monomer in solution. Circular dichroism studies suggest that fasciclin I is primarily alpha-helical. Its structure is therefore different from other known cell adhesion molecules, which are predicted to be elongated beta-sheet structures. This suggests that fasciclin I may define a new structural motif used to mediate adhesive interactions between cell surfaces

    THE EFFECTS OF MIDSOLE CONSTRUCTION ON BOWLING DELIVERY

    The purpose of this study was to examine the effects of midsole construction on the mechanics of bowling delivery. Twelve participants performed five candle bowling ball deliveries in three footwear conditions (barefoot, traditional and modified with E-TPU). Both kinematic and kinetic analyses were conducted. The results of the barefoot condition showed hip, knee and ankle joints at 92.3 ± 9.9°, 133.0 ± 14.5°, and 111.6 ± 11.2°, respectively, which were not significantly different from the modified condition of 93.9 ± 9.9° (p =.289) 132.1 ± 14.5° (p =.758), and 117.7 ± 9.4° (p =.123). Also, no significant difference was found in peak vertical force to body weight ratio. These findings provide a preliminary understanding on the effects of footwear on bowling delivery. Future studies are warranted to evaluate 3D motion analysis with experienced bowlers at the bowling alley

    KINETIC ANALYSES OF E-TPU MATERIAL IN BOWLING FOOTWEAR

    The purpose of this study was to examine the vertical ground reaction force between the traditional bowling shoes made of rubber midsole and the modified bowling shoes made of E-TPU midsole. All shoes underwent both static and dynamic performance testing on a force platform. The preliminary results indicated that the bowling footwear with the E-TPU material provided lower amount of vertical ground reaction force in both static and dynamic testing, which may potentially be beneficial to bowlers to minimize lower extremity injury. Future studies are warranted to evaluate the internal joint forces of bowling delivery mechanics with the E-TPU material footwear

    Kinematic Comparisons between Forward and Backward Skating in Ice Hockey

    The purpose of this study was to examine the kinematics of lower body and trunk between forward and backward skating. Seven male collegiate ice hockey players skated both forward and backward (C-cut) four times each. A standard two-dimensional kinematic analysis was conducted to examine the lower body extremity and trunk at the instants of weight acceptance and propulsion. No significant differences in the joint angles were found at the weight acceptance. At the propulsion, significant differences were found in the joint angles of hip, knee, ankle and trunk between forward and backward skating. Hence, these findings indicate the importance of strengthening lower body joints and trunk, particularly for forward skating. Future studies are warranted to examine the mechanics of forward and backward skating using a 3D analysis

    Willingness of Private Physicians to Be Involved in Smallpox Preparedness and Response Activities

    Background. The public health system continues its efforts to prepare for bioterrorist events, such as a smallpox outbreak, but may need to call on other health professionals to ensure sufficient capacity to implement preparedness plans. Objective. The goal was to understand the willingness of primary care physicians to participate in possible smallpox pre- or post-event activities. Methods. A 23-question mail survey was sent to a national random sample of 727 internists and 720 family physicians. After three mailings, a one-page version of the survey was sent to nonrespondents. Results. Response rates were 26% for questions common to both surveys and 22% for questions on the longer survey only. Respondents to the survey expressed moderate support for participating in certain smallpox pre- and post-event activities. Under a pre-event scenario, many providers would be willing to vaccinate first responders in their practice, and roughly one-third would be willing to vaccinate patients in their practice or to work in a public health clinic as a vaccinator. Most physicians, however, would be unwilling to be vaccinated themselves. Under post-event conditions, most providers would be willing to vaccinate their own patients, and many would vaccinate other community members in their practice. Conclusions. Despite the low response rate, information from this study on the smallpox preparedness activities in which physicians are most willing to participate can help to inform efforts by public health officials and private physicians to collaborate on bioterrorism preparedness efforts.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/63165/1/bsp.2005.3.16.pd

    Localization of Mycobacterium tuberculosis topoisomerase I C-terminal sequence motif required for inhibition by endogenous toxin MazF4

    Only about half the multi-drug resistant tuberculosis (MDR-TB) cases are successfully cured. Thus, there is an urgent need of new TB treatment against a novel target. Mycobacterium tuberculosis (Mtb) topoisomerase I (TopA) is the only type IA topoisomerase in this organism and has been validated as an essential target for TB drug discovery. Toxin-antitoxin (TA) systems participate as gene regulators within bacteria. The TA systems contribute to the long-term dormancy of Mtb within the host-cell environment. Mtb’s toxin MazF4 (Rv1495) that is part of the MazEF4 TA system has been shown to have dual activities as endoribonuclease and topoisomerase I inhibitor. We have developed a complementary assay using an Escherichia coli strain with temperature-sensitive topA mutation to provide new insights into the MazF4 action. The assay showed that E. coli is not sensitive to the endoribonuclease activity of Mtb MazF4 but became vulnerable to MazF4 growth inhibition when recombinant Mtb TopA relaxation activity is required for growth. Results from the complementation by Mtb TopA mutants with C-terminal deletions showed that the lysine-rich C-terminal tail is required for interaction with MazF4. Site-directed mutagenesis is utilized to identify two lysine residues within a conserved motif in this C-terminal tail that are critical for MazF4 inhibition. We performed molecular dynamics simulations to predict the Mtb TopA-MazF4 complex. Our simulation results show that the complex is stabilized by hydrogen bonds and electrostatic interactions established by residues in the TopA C-terminal tail including the two conserved lysines. The mechanism of Mtb TopA inhibition by MazF4 could be useful for the discovery of novel inhibitors against a new antibacterial target in pathogenic mycobacteria for treatment of both TB and diseases caused by the non-tuberculosis mycobacteria (NTM)

    A Protocol of Process Evaluations of Interventions for the Prevention of Type 2 Diabetes in Women With Gestational Diabetes Mellitus: A Systematic Review

    Background Process evaluations of randomised controlled trials (RCTs) can provide insight and inform us on the intervention implementation, the causal mechanisms and the contextual factors. This will inform about interventions’ success or failure due to their implementation or the interventions themselves. We aim to consolidate the methodology from previous process evaluations of complex interventions upon their findings on facilitators and barriers to address the prevention of type 2 diabetes mellitus among women with gestational diabetes mellitus (GDM). Methods Comprehensive search will be conducted on electronic databases and reference lists of recent reviews for RCTs of complex interventions which address process evaluations of diabetes prevention intervention (DPI) for women with GDM in healthcare settings. There is no restriction on the language of the papers and year of publication until December 2020. Data from each study will be extracted by two reviewers independently using standardised forms. Data extracted include descriptive items on the study design and the outcomes of process evaluations from the three dimensions: (1) implementation; (2) mechanism of impact and (3) context. The quality of the studies will be assessed using mixed methods appraisal tool which is designed for the appraisal of mixed studies in systematic reviews. A narrative and framework analysis of the findings will be presented to inform the contents of a new DPI for women with GDM. Discussion The findings from this process evaluation findings are valuable in determining whether a complex intervention should be scaled up or modified for other contexts in future plan. It will give deeper understanding of potential challenges and solutions to aid in the implementation of effective DPIs for GDM in Malaysia
