    Antitumor and Immunopotentiating Activity of Polysaccharide PST001 Isolated from the Seed Kernel of Tamarindus indica: An InVivo Study inMice

    Antitumor activity of polysaccharide PST001 isolated from the seed kernel of Tamarindus indica was evaluated using different cancer cell lines. Human cancer cell lines A549, KB, and MCF-7 and murine cancer cell lines DLA and EAC were treated with PST001 and cell growth inhibition was assessed by MTT assay. In vivo studies were carried out for toxicity, tumor reduction and immunomodulation. The respective IC50 of PST001 in A549, KB, and DLA was at 80.72, 190.99, and 91.14 μg/mL. Significant tumor reduction was obtained in both DLA and EAC tumors on treatment with PST001 which was more prominent when PST001 was administered with CTX/5-fluorouracil. Increase in total WBC, CD4+ T-cell population, and bonemarrow cellularity suggested strong immunomodulatory activity for this compound. No significant abnormality was observed in toxicity studies. Thus the results of the present study suggest that PST001 has immunomodulatory and tumor inhibitory activities and has the potential to be developed as an anticancer agent and immunomodulator either as a sole agent or as an adjuvant to other chemotherapeutic drugs

    DEVELOPMENT AND VALIDATION OF HIGH-PERFORMANCE LIQUID CHROMATOGRAPHIC METHOD FOR DETERMINATION OF DAPAGLIFLOZIN AND ITS IMPURITIES IN TABLET DOSAGE FORM

    Objective: The aim of the present work is the development of new, sensitive, specific, and accurate high-performance liquid chromatographic method for the separation and determination of dapagliflozin and its impurities in tablet dosage form. Methods: The chromatographic separation of drug and its impurities was achieved using Hypersil BDS C18 column (250 mm × 4.6 mm, 5 μ) with mobile phase consisted of mobile phase-A (Buffer pH 6.5) and mobile phase-B (acetonitrile:water 90:10) by gradient program at a flow rate of 1 mL/min with ultraviolet detection at 245 nm. Results: Dapagliflozin and its impurities A, B, C, D, E, and impurity-F were successfully eluted at the retention time of 16.95, 2.72, 7.82, 10.58, 21.11, 30.37, and 34.36 min, respectively, with good resolution. The method was validated according to the international conference on harmonization guidelines. The validation results showed good precision, accuracy, linearity, specificity, sensitivity, and robustness. Conclusion: Successful separation and determination of dapagliflozin and its six impurities were achieved by the proposed method. The developed method can be applied for the routine analysis of dapagliflozin and its impurities in pharmaceutical formulations

    ANALYTICAL METHOD DEVELOPMENT FOR SIMULTANEOUS DETERMINATION OF UBIDECARENONE AND VITAMIN E ACETATE IN CAPSULE DOSAGE FORM BY HPLC

    Objective: To develop and validate a new simple, accurate, precise and sensitive high performance liquid chromatographic method (HPLC) method for simultaneous estimation of ubidecarenone and vitamin E acetate in capsule dosage form as per international conference on harmonization (ICH) guidelines. Methods: The chromatographic separation of drugs were achieved using hypersil C8 column (250 mm x 4.6 mm, 5µ) in isocratic elution mode with a mobile phase of methanol: ethanol: n-hexane (80:10:10 v/v/v) at a flow rate of 1 ml/min with ultra-violet (UV) detection at 210 nm. Results: The optimized method produced sharp peaks with good resolution, minimum tailing factor and satisfactory retention time were found to be 5.745 min and 12.565 min for vitamin E acetate and ubidecarenone respectively. The method was linear in the range of 60-180 µg/ml for ubidecarenone and 20-60 µg/ml for vitamin E acetate with a correlation coefficient of 0.999 and 0.9993 respectively. Mean recoveries observed for ubidecarenone and vitamin E acetate were 99.85% and 99.73% respectively. The percentage relative standard deviation (% RSD) of peak area for system precision, method precision, and intermediate precision were found to be less than 0.37%. The lower degree of % RSD obtained has proved that the method was precise and robust. Conclusion: A new simple HPLC method was developed and validated as per ICH guidelines for the simultaneous estimation of ubidecarenone and vitamin E acetate and the method can be effectively applied for the routine analysis of active pharmaceutical ingredient (API) and formulations

    Measurements of carbon dioxide and heat fluxes during monsoon-2011 season over rural site of India by eddy covariance technique

    An increase in carbon dioxide (CO2) concentrations in the atmosphere due to anthropogenic activities is responsible for global warming and hence in recent years, CO2 measurement network has expanded globally. In the monsoon season (July-September) of year 2011, we carried out measurements of CO2 and water vapour (H2O) concentrations along with wind and air temperature over a tropical site in southeast India having rural topography. To collect these observations, the instrumentations used were the sonic anemometer for wind and temperature, and the open path H2O/CO2 infrared gas analyzer for CO2 and H2O concentrations. Using these observations, we explored the diurnal variability of CO2 flux along with sensible and latent heat. The CO2 flux was positive during night-time and negative during daytime and in phase with convective instability. The CO2 flux relationships with the meteorological parameters such as wind speed, temperature and heat fluxes have been analysed. The seasonal (monsoon) half hour mean of CO2 flux which was -3.55 μmol m-2 s-1 indicated the experimental site as a CO2 sink region (net seasonal uptake). An increase in CO2 concentrations during weekends was not observed due to unavailability of heavy vehicular traffic

    A comparative study of management of Colles fracture by closed reduction with cast versus closed reduction with internal fixation (K-wires/4 mm CC screws)

    Background: Distal radius fracture is extremely common and represents 16% of fractures treated by orthopaedic surgeons. Near anatomical reduction with restoration of radial length, radial tilt and ulnar variances are important for good functional results. A variety of treatment options have been proposed for distal radius fracture closed reduction and immbolization in cast has been the main stay of treatment, but because it invariably results in malunion, poor functional outcome and cosmetic outcome, other modality of treatment were brought into practice like percutaneous intrafocal pinning, transulnar percutaneous pinning, external fixation, plating etc. This comparative study was to compare the clinical outcomes of closed reduction with cast and closed reduction with Percutaneous Kirschner wiring (PKW)/canullated cancellous screws (CC screws 4 mm)for the management of Colles fractures in patients between 20 and 70 years old. In this study we did an prospective study in 40 patients with extra articular distal radius fracture treated by both closed reduction with cast and closed reduction internal fixation (PKW/CC screws).Methods: There were 20 patients with Colles fractures treated by closed reduction with cast and 20 patients treated with closed reduction internal fixation (PKW/CC screws). We compared both the managements together. All patients were followed up in the orthopaedic department prospectively for at least 12 months between June 2015-June 2016 The functional outcomes and radiological results were compared between the two groups.Results: The study included 40 patients, aged between 20 to 69 yrs. Average follow up was 12 months. Using the demerit scoring system of Gartland and Werley we had excellent to good results of 60% in closed reduction internal fixation compared to closed reduction with casting excellent to good was 10%.Conclusions: The percutaneous pinning/cc screws and immobilization in neutral position for 3 weeks followed by physiotherapy proved to be better and simple procedure for extra articular non comminuted distal radius fractures.
