    A novel breeding programme for improved growth in barramundi Lates calcarifer (Bloch) using foundation stock from progeny-tested parents

    Rapid genetic gains for growth in barramundi ( Lates calcarifer) appear achievable by starting a breeding programme using foundation stock from progeny tested broodstock. The potential gains of this novel breeding design were investigated using biologically feasible scenarios tested with computer simulation models. The design involves the production of a large number of full-sib families using artificial mating which are compared in common growout conditions. The estimated breeding values of their paternal parents are calculated using a binomial probit analysis to assess their suitability as foundation broodstock. The programme can theoretically yield faster rates of genetic gain compared to other breeding programmes for aquaculture species. Assuming a heritability of 0.25 for growth, foundation broodstock evaluated in two years had breeding values for faster growth ranging from 21% to 51% depending on the genetic diversity of stock under evaluation. As a comparison it will take between nine and twenty-two years to identify broodstock with similar breeding values in a contemporary barramundi breeding programme

    Nutritional status of a nereidid polychaete cultured in sand filters of mariculture wastewater

    This study examined the nutritional composition of the intertidal marine polychaete Perinereis helleri (Nereididae)when artificially cultured in sand filters treating mariculture wastewater. Moisture levels in harvested P. helleri ranged from 758 to 855 g kg1, and ash, from 23 to 61 g kg1 wet matter (WM). Stocking density and graded size after harvest significantly affected their composition. Higher total lipid contents were found in large (>0.6 g) P. helleri(16–19 g kg1 WM) and those grown at the lowest density(1000 m2: 18 g kg 1 WM) than in small (≤0.6 g) ones (14 g kg1 WM) and those grown at the highest densities (4000–6000 m2: 13–16 g kg1 WM). Several fatty acids within a very broad profile (some 30 identified) reflected this pattern, yet their ARA/EPA/DHA ratios were relatively unaffected. Feeding the polychaete-assisted sand filters (PASF) with fish meal to increase worm biomass productivity significantly increased their DHA content. Other components (e.g. protein, phospholipids, cholesterol, carbohydrate, amino acids, nitrogen, minerals and bromophenols) and nutritional factors (e.g. maturity, feeding seaweed and endemic shrimp viral content) were also investigated. Results suggest that PASF-produced P. helleri have a well-balanced nutritional profile for penaeid shrimp and fish broodstock

    Empirical analyses of the length, weight, and condition of adult Atlantic salmon on return to the Scottish coast between 1963 and 2006

    Sea age, size, and condition of adult Atlantic salmon (Salmo salar) are prime determinants of individual, and hence population, productivity. To elucidate potential mechanisms, 151 000 records of salmon returning to six Scottish coastal sites over 44 years were analysedfor length, weight, and condition, by site, sex, sea age, and river age. After correcting for capture effort biases, all sites showed seasonal increases in length and weight for both 1 sea winter (1SW) and 2SW fish. However, whereas condition increased slightly with season for 2SW, it decreased notably for 1SW. Sites showed common decadal trends in length, weight, and condition. Within years, length and weight residuals from trends were coherent across sites, but residuals from condition trends were not. Rates of seasonal condition change also showed decadal trends, dramatically different between sea ages, but common across sites within sea-age groups. Longer salmon were disproportionately heavy in all seasons. 1SW condition was markedly lower in 2006. Detrended correlations withoceanic environmental variables were generally not significant, and always weak. A published correlation between the condition of 1SW salmon caught at a single site and sea surface temperatures in the Northeast Atlantic could not be substantiated for any of the six fisheries over the wider time-scales examined

    Proliferation and differentiation of progenitor cells throughout the intact adult rat spinal cord

    The existence of multipotent progenitor populations in the adult forebrain has been widely studied. To extend this knowledge to the adult spinal cord we have examined the proliferation, distribution, and phenotypic fate of dividing cells in the adult rat spinal cord. Bromodeoxyuridine (BrdU) was used to label dividing cells in 13- to 14-week-old, intact Fischer rats. Single daily injections of BrdU were administered over a 12 d period. Animals were killed either 1 d or 4 weeks after the last injection of BrdU. We observed frequent cell division throughout the adult rodent spinal cord, particularly in white matter tracts (5-7% of all nuclei). The majority of BrdU-labeled cells colocalized with markers of immature glial cells. At 4 weeks, 10% of dividing cells expressed mature astrocyte and oligodendroglial markers. These data predict that 0.75% of all astrocytes and 0.82% of all oligodendrocytes are derived from a dividing population over a 4 week period. To determine the migratory nature of dividing cells, a single BrdU injection was given to animals that were killed 1 hr after the injection. In these tissues, the distribution and incidence of BrdU labeling matched those of the 4 week post injection (pi) groups, suggesting that proliferating cells divide in situ rather than migrate from the ependymal zone. These data suggest a higher level of cellular plasticity for the intact spinal cord than has previously been observed and that glial progenitors exist in the outer circumference of the spinal cord that can give rise to both astrocytes and oligodendrocytes

    Interpreting complex fluvial channel and barform architecture: Carboniferous Central Pennine Province, northern England

    The Bashkirian Lower Brimham Grit of North Yorkshire, England, is a fluvio-deltaic sandstone succession that crops out as a complex series of pinnacles, the three-dimensional arrangement of which allows high-resolution architectural analysis of genetically-related lithofacies assemblages. Combined analysis of sedimentary graphic log profiles, architectural panels and palaeocurrent data have enabled three-dimensional geometrical relationships to be established for a suite of architectural elements so as to develop a comprehensive depositional model. Small-scale observations of facies have been related to larger-scale architectural elements to facilitate interpretation of the palaeoenvironment of deposition to a level of detail that has rarely been attempted previously, thereby allowing interpretation of formative processes. Detailed architectural panels form the basis of a semi-quantitative technique for recording the variety and complexity of the sedimentary lithofacies present, their association within recognizable architectural elements and, thus, the inferred spatio-temporal relationship of neighbouring elements. Fluvial channel-fill elements bounded by erosional surfaces are characterized internally by a hierarchy of sets and cosets with subtly varying compositions, textures and structures. Simple, cross-bedded sets represent in-channel migration of isolated mesoforms (dunes); cosets of both trough and planar-tabular cross-bedded facies represent lateral-accreting and downstream-accreting macroforms (bars) characterized by highly variable, yet predictable, patterns of palaeocurrent indicators. Relationships between sandstone-dominated strata bounded by third-order and fifth-order surfaces, which represent in-channel bar deposits and incised channel bases respectively, chronicle the origin of the preserved succession in response to autocyclic barform development and abandonment, major episodes of incision probably influenced by episodic tectonic subsidence, differential tilting and fluvial incision associated with slip on the nearby North Craven Fault system. Overall, the succession represents the preserved product of an upper-delta plain system that was traversed by a migratory fluvial braid-belt system comprising a poorly-confined network of fluvial channels developed between major sandy barforms that evolved via combined lateral-accretion and downstream-accretion

    New insights into landslide processes around volcanic islands from Remotely Operated Vehicle (ROV) observations offshore Montserrat

    Submarine landslide deposits have been mapped around many volcanic islands, but interpretations of their structure, composition, and emplacement are hindered by the challenges of investigating deposits directly. Here we report on detailed observations of four landslide deposits around Montserrat collected by Remotely Operated Vehicles, integrating direct imagery and sampling with sediment core and geophysical data. These complementary approaches enable a more comprehensive view of large-scale mass-wasting processes around island-arc volcanoes than has been achievable previously. The most recent landslide occurred at 11.5–14 ka (Deposit 1; 1.7 km3) and formed a radially spreading hummocky deposit that is morphologically similar to many subaerial debris-avalanche deposits. Hummocks comprise angular lava and hydrothermally altered fragments, implying a deep-seated, central subaerial collapse, inferred to have removed a major proportion of lavas from an eruptive period that now has little representation in the subaerial volcanic record. A larger landslide (Deposit 2; 10 km3) occurred at ∼130 ka and transported intact fragments of the volcanic edifice, up to 900 m across and over 100 m high. These fragments were rafted within the landslide, and are best exposed near the margins of the deposit. The largest block preserves a primary stratigraphy of subaerial volcanic breccias, of which the lower parts are encased in hemipelagic mud eroded from the seafloor. Landslide deposits south of Montserrat (Deposits 3 and 5) indicate the wide variety of debris-avalanche source lithologies around volcanic islands. Deposit 5 originated on the shallow submerged shelf, rather than the terrestrial volcanic edifice, and is dominated by carbonate debris

    The TatC component of the twin-arginine protein translocase functions as an obligate oligomer

    The Tat protein export system translocates folded proteins across the bacterial cytoplasmic membrane and the plant thylakoid membrane. The Tat system in Escherichia coli is composed of TatA, TatB and TatC proteins. TatB and TatC form an oligomeric, multivalent receptor complex that binds Tat substrates, while multiple protomers of TatA assemble at substrate-bound TatBC receptors to facilitate substrate transport. We have addressed whether oligomerisation of TatC is an absolute requirement for operation of the Tat pathway by screening for dominant negative alleles of tatC that inactivate Tat function in the presence of wild-type tatC. Single substitutions that confer dominant negative TatC activity were localised to the periplasmic cap region. The variant TatC proteins retained the ability to interact with TatB and with a Tat substrate but were unable to support the in vivo assembly of TatA complexes. Blue-native PAGE analysis showed that the variant TatC proteins produced smaller TatBC complexes than the wild-type TatC protein. The substitutions did not alter disulphide crosslinking to neighbouring TatC molecules from positions in the periplasmic cap but abolished a substrate-induced disulphide crosslink in transmembrane helix 5 of TatC. Our findings show that TatC functions as an obligate oligomer.</p
