Determination of Tetracycline Residues in Broilers Reared in Makurdi Metropolis
Tetracycline residues in broiler tissues were assayed using a microbiological assay method. A total of four hundred samples representing one hundred samples of liver, kidney, lungs and breast muscles each, were collected from five different farms within Makurdi Metropolis. The analysis showed a total of 70 % residue incidence with the liver having 60 %, Kidney 31 %, lungs 14 % and muscles 5 % of the total. The highest concentration, of 6 µg/kg, was seen in the liver from farm 1, which had an incidence of 50 %, with the liver contributing 90 % of the incidence. Farm 2 had an incidence of 75 %, in which the liver contributed with 50 % of the cases. Farm 3 had a 100 % incidence, with the liver contributing with 75 % of the cases. Farm 4 had 75 % incidence, with the liver involved in all cases. Farm 5 had a 50 % incidence, all from the liver and kidney. Despite the higher incidence of 70 %, all the values were significantly (p < 0.01), lower than the recommended Maximum Residue Limits (MRL) or Tolerance of 600, 300 and 200 µg/kg for the liver, kidney and muscle respectively. The values were also in accordance within the Acceptable Daily Intake (ADI) of 0-30 µg/kg, recommended by the Codex Alimentarius. This may be an indication of the rational use of antibiotics in good agricultural management in the selected farms in Makurdi Metropolis. Significantly, it shows that this method can be used mathematically to quantify drug residues in lower income areas.

Effect of intramuscular administration of oxytetracycline on serum kinetics of diminazene aceturate in healthy male Sahel goats
Serum kinetics of diminazene aceturate following intramuscular (IM) administration of diminazene aceturate alone at 3.5 mg/kg and its combination with oxytetracycline at 20 mg/kg were evaluated in 6 healthy male Sahel goats to ascertain the effect of oxytetracycline on serum concentration and pharmacokinetic parameters of diminazene aceturate. Two groups (A and B) of three goats each were used, and oxytetracycline was administered 30 minutes prior to diminazene aceturate administration. Blood samples were collected at various intervals (0.17 h – 72 h) post-drug administration, and diminazene serum concentrations were measured using the method of Klatt and Hadju. Kinetic determinants were calculated employing a two-compartment open model. Mean serum concentrations of diminazene aceturate of 2.43 ± 0.95 μg/ml and 1.73 ± 0.44 μg/ml at 0.17 h were measured in groups A and B, respectively. These serum concentrations were found to increase until a peak concentration of 6.91 ± 0.59 μg/ml and 7.55 ± 1.20 μg/ml were reached at 2.0 h in groups A and B, respectively. The peak concentrations subsequently decreased at 72 h post diminazene aceturate administration with serum concentrations of 0.00 ± 0.00 and 0.32 ± 0.28 μg/ml in groups A and B, respectively. Pharmacokinetics parameters like the volume of distribution (Vd), elimination half-life (T½β), concentration maximum (Cmax), absorption rate constant (α), and area under the curve from 0 to 72 h (AUC0 – 72) were significantly higher in goats treated with diminazene aceturate and oxytetracycline combination while total body clearance (Cl), and elimination rate constant (β), were significantly higher in goats treated with diminazene aceturate alone. The mean residence time (MRT) of diminazene aceturate increased from 19.70 ± 2.53 h in diminazene aceturate treatment to 25.11 ± 1.81 h in diminazene aceturate and oxytetracycline treatment. Działanie biochemiczne wodnego wyciągu z liści Abrus precatorius na myszy szczepu Swiss
The leaf of Abrus precatorius has been used in Nigeria for the treatment of myriad of diseases
including malaria, typhoid, cough, respiratory tract infections and hepatitis. Since
the plant, especially the seeds, has been widely known to be toxic, there is a need to
investigate biochemical and histopathological effect of aqueous extract of Abrus leaf in
swiss albino mice. Biochemical analysis revealed significantly (p<0.05) increased aspartate
aminotransferase, alanine aminotransferase, alkaline phosphatase, creatinine, urea, and
chloride ion. There were also noted hypophosphataemia, hypokalaemia and hypoglycaemia
(p<0.05). Histopathology revealed mononuclear cellular infiltrations of heart, lung,
liver, intestine, spleen and kidney cells. Zenker’s necrosis of hepatocytes, thickening of
interalveolar septae of pneumocytes and degeneration of splenic and kidney cells were
observed as well. Hence aqueous extract of Abrus precatorius leaf is toxic at dose levels of
12.5, 25, 50, 100 and 200 mg/kg body weight when administered for a period of 21 days.Liście Abrus precatorius wykorzystywane są w Nigerii w leczeniu wielu chorób, m. in. malarii,
duru brzusznego, kaszlu, infekcji dróg oddechowych oraz zapalenia wątroby. Ponieważ
roślinę uznano za toksyczną, w szczególności jej nasiona, dlatego przeprowadzono badania
mające na celu ocenę biochemicznego oraz histopatologicznego działania wodnego
wyciągu z liści Abrus z wykorzystaniem myszy szczepu Swiss. W wyniku biochemicznych
analiz stwierdzono statystycznie znamienny (p<0,05) wzrost aktywności aminotransferazy
asparaginowej oraz alaninowej, fosfatazy alkalicznej, stężenia kreatyniny, mocznika i jonów
chlorku. Odnotowano także hypofosfatemię, hypokaliemię oraz hypoglikemię (p<0,05).
W wyniku przeprowadzonych badań histopatologicznych komórek jednojądrzastych
wykryto nacieki w sercu, płucach, wątrobie, jelitach, śledzionie oraz nerkach. Ponadto zaobserwowano
martwicę Zenkera hepatocytów, zgrubienie przegród między pęcherzykami
płucnymi oraz degenerację komórek śledziony i nerek. Wykazano toksyczne działanie
wodnego wyciągu z liści Abrus precatorius w dawkach 12,5, 25, 50, 100 oraz 200mg/kg
masy ciała po podawaniu go przez 21 dni
Badanie działania wyciągu z kory Annona muricata przeciw śpiączce afrykańskiej in vitro i in vivo
The control of trypanosomosis in animals and humans based on chemotherapy is limited and not ideal, since the agents used are associated with severe side effects, and emergence of relapse and drug resistant parasites. The need for the development of new, cheap and safe compounds stimulated this study. Three concentrations (211, 21.1 and 2.11 mg per ml) of chloroform stem bark extract of Annona muricata were screened for trypanocidal activity against Trypanosoma brucei brucei in vitro. Also, two doses (200 mg per kg and 100 mg per kg) of the extract were evaluated for trypanocidal activity in rats infected with the parasite. Haematological parameters were determined on day 1 post infection and on days 1, 6 and 30-post extract treatment. The extracts inhibited parasite motility and totally eliminated the organisms at the concentrations used in vitro. The extract also showed promising in vivo trypanocidal activity. The observed in vitro and in vivo trypanocidal activities may be due to the presence of bioactive compounds present in the extracts as seen in this study. The extract also improved the observed decreases in haematological parameters of the treated rats, which may be due to their ability to decrease parasite load. The observed oral LD50 of 1,725.05 mg per kg of the chloroform A. muricata extract using up and down method is an indication of very low toxicity, implying that the extract could be administered with some degree of safety.Leczenie chemioterapeutykami śpiączki afrykańskiej, tak u ludzi jak i u zwierząt, jest
trudne ze względu na powstawanie wielu poważnych objawów niepożądanych oraz wywoływanie
zjawiska oporności podczas ich stosowania, co powoduje brak skuteczności
i pojawianie się nawrotów tej choroby. Stąd potrzeba znalezienia nowego skutecznego
i taniego leku była powodem podjęcia badań przedstawianych w tej pracy. Celem badań
było określenie aktywności wyciągu chloroformowego z kory Annona muricata stosowanego
w warunkach in vitro dla trzech stężeń (211, 21,1 i 2,11 mg/ml) przeciwko aktywności
Trypanosoma brucei brucei. Ponadto w badaniach in vivo określano aktywność tego ekstraktu
w dwóch dawkach (100 i 200 mg/kg) u szczurów zarażonych pasożytem. Do oceny stanu
zwierząt wykorzystano pomiar odpowiednich parametrów hematologicznych po 1 dniu od ich zainfekowania oraz po 1, 6 i 30 dniu podawania badanego wyciągu. W badaniach in
vitro we wszystkich zastosowanych stężeniach wyciąg hamował ruchliwość omawianego
pasożyta. Ponadto w badaniach in vivo ekstrakt całkowicie eliminował obecność Trypanosoma
brucei brucei u zainfekowanych zwierząt, wykazując w ten sposób obiecującą aktywność
przeciwko trypanosomatozie. Zaobserwowane działanie, tak in vitro jak i in vivo,
było być może związane z obecnością związków czynnych charakteryzujących stosowany
wyciąg. Analizowany ekstrakt, prawdopodobnie ze względu na obniżenie liczby pasożytów
po jego podawaniu, miał także zdolność poprawiania parametrów hematologicznych
u zainfekowanych zwierząt. Ze względu na otrzymaną wartość LD50 dla zastosowanego
wyciągu chloroformowego z kory Annona muricata, wynoszącą 1725,05 mg/kg po podaniu
dożołądkowym szczurom, można sądzić, że stosowanie badanego wyciągu jest względnie
bezpieczne
Age-sex dimorphisms in the estimation of median lethal dose (LD50) of lead diacetate in rabbits using up-and-down procedure (Arithmetic method)
Lead had been known to be toxic since pre-antiquity. It causes neurodevelopmental, cardiovascular and renal pathologies. Other toxicological studies of lead (Pb) salts are studied in laboratory animals such as rat, mice, hamsters, rabbits and monkeys; however, there is paucity of information on the median lethal dose (LD50) of Pb salts in rabbits especially when used as a model for molecular studies of Pb toxicity. In view of this, oral LD50 of lead (Pb) diacetate [Pb(CH3COO)2] was determined in both young (6 weeks old, weighing 0.40 ± 0.03 kg) and adult (34 weeks old, weighing 1.63 ± 0.10 kg), male and female New Zealand White (NZW) rabbits (Oryctolagus cuniculus) using the revised arithmetic method of Up-and-Down Procedure (UDP). The estimated LD50 of Pb diacetate in young male and female NZW rabbits was 1214.20 ± 275.80 and 1214.20 ± 275.80 mg/kg body weight respectively. Also, the LD50 of the adult male and female rabbits was 1503.30 ± 342.90 and 1792.50 ± 354.40 mg/kg body weight respectively. Similarities in the estimated LD50 of the young rabbits could be attributed to poorly developed xenobiotic metabolic processes. Sex-dimorphism in the toxicity of Pb diacetate was observed in the adult animals, where the male animals were found to be more sensitive to the toxicant than the female. Hence, Pb diacetate is moderately toxic in NZW Rabbits according to “Gosselin, Smith and Hodge scale” of toxicity rating.Keywords: Arithmetic mean, Dimorphism, Dose, Lead, Rabbits, Sex, Toxicit
Antidiarrheal activity of fractions from aqueous extract of Detarium senegalense
The aim of this study was to evaluate the effect of different fractions of the aqueous crude extract of Detarium senegalense stem bark on castor oil-induced diarrhea. Castor oilinduced diarrhea, gastrointestinal motility and castor oil-induced enteropooling methods were used to evaluate the antidiarrheal effects of the fractions. Castor oil was used to induce diarrhea and the effect of all the fractions (chloroform, ethyl acetate, n-butanol, methanol and residual aqueous) were evaluated at the doses of 200 and 400 mg/kg body weight. The results show that all fractions significantly (p<0.05) decreased the frequency of defecation in rats following the induction of diarrhea with castor oil. Ethyl acetate which produced the highest antidiarrheal activity was further subjected to gastrointestinal motility and castor oil-induced enteropooling tests. In the gastrointestinal motility, two test doses of the extract (200 and 400 mg/kg) were administered orally to two groups of rats (n=5), while the third group of rats (control), were treated with normal saline, and the fourth group were treated with diphenoxylate, a conventional anti-diarrheal drug. In the castor oil-induced enteropooling experiment, normal saline was used for the control animals, while 200 and 400 mg/kg of the extract was administered to groups two and three, respectively and atropine, a standard drug, was administered to rats in group four. The ethyl acetate fraction significantly (p<0.05) decreased the gastro-intestinal motility, as shown by the extent of movement of the charcoal meal in the treated rats. It also significantly inhibited the fluid accumulation within the intestine. These findings suggest that the ethyl acetate fraction possess antidiarrheal effect, which may be due to the presence of some phytochemical constituents (alkaloids, flavonoids and tannins) in the plant, which may either be working alone or in combination with each other. This study has demonstrated that D. senegalense fractions, especially the ethyl acetate fraction, possess antidiarrheal activity thus supporting the use of the plant in the treatment of diarrheal diseases.Celem niniejszej pracy było określenie wpływu różnych frakcji surowego wyciągu wodnego
z kory pnia Detarium senegalense na hamowanie biegunki indukowanej olejem rycynowym
u szczurów. Działanie przeciwbiegunkowe tych frakcji, po wywołaniu biegunki
olejem rycynowym, określono na podstawie obserwacji ruchów robaczkowych jelit oraz gromadzenia płynu w świetle jelita. Badano działanie przeciwbiegunkowe różnych frakcji
(chloroformowej, n-butanolowej, metanolowej oraz otrzymanej za pomocą octanu etylu,
jak i pozostałości wodnej) w dawkach 200 i 400 mg/kg masy ciała szczura. Badania wykazały,
że wszystkie frakcje istotnie (p<0.05) obniżyły częstość defekacji u szczurów po wywołaniu
biegunki w stosowanym modelu. Frakcja otrzymana przez działanie octanem etylu,
wykazująca najsilniejsze działanie przeciwbiegunkowe, została następnie użyta w ocenie
ruchów robaczkowych jelita i gromadzenia płynów w jego świetle. W teście oceny ruchów

drzewnego szczurom. Frakcja ta również istotnie hamowała gromadzenie płynu w jelicie.
Otrzymane rezultaty sugerują, że frakcja otrzymana za pomocą octanu etylu ma właściwości
przeciwbiegunkowe, prawdopodobnie ze względu na obecność w roślinie aktywnych
związków chemicznych (alkaloidów, flawonoidów i tanin), które mogą działać bądź pojedynczo
lub jako ich kombinacja. Podsumowując wykazano, że frakcje z D. senegalense,
a szczególnie frakcja otrzymana za pomocą octanu etylu, mają właściwości przeciwbiegunkowe,
co może wskazywać na możliwość zastosowania badanej rośliny do leczenia
chorób przebiegających z biegunką

Porównanie toksyczności dożołądkowego oraz dootrzewnowego podawania wodnych ekstraktów z liści modligroszka pospolitego (Abrus precatorious) w badaniach na myszach
Phytochemical analysis revealed the presence of alkaloids, flavonoids, saponins, tannins, reducing
sugar, total glycoside and saponin glycosides in high concentration. Proximate analysis
revealed the presence of moisture (6.18%), dry matter (93.90%), crude protein (12.06%),
crude fibre (15.33%), nitrogen free extract (34.12%), ash (12.28%) and oil (19.21%). Comparative
toxicological assessment of orally and intraperitoneally administered aqueous extracts
of Abrus precatorius leaf was carried out in Swiss albino mice. Median lethal doses (LD50)
of orally and intraperitoneally administered aqueous extracts of Abrus precatorius leaf were
estimated at dose levels of 2558.9 mg/kg and 638 mg/kg body weight, respectively. Intraperitoneal
10 mg/kg body weight of the extract caused increased packed cell volume, neutropenia
and decreased aspartate aminotransferase. However, 50 mg/kg oral dose caused
increased packed cell volume, neutropenia, decreased alkaline phosphatase and hypochloraemia,
whereas oral aqueous dose of 250 mg/kg of body weight caused body weight gain,
neutropenia, decreased asparatate aminotransferase and alanineaminotransferase. All the
test doses caused lymphocytosis and hypercreatinaemia, hence aqueous extract of Abrus
precatorius leaf is toxic at dose levels of 10, 50 and 250 mg/kg body weight.Analiza fitochemiczna wodnego wyciągu z liści Abrus precatorious wykazała obecność dużych
ilości alkaloidów, saponin, tanin, cukrów redukujących, sumy glikozydów oraz glikozydów
saponinowych. W dalszych analizach oznaczono: wilgotność (6,18%), suchą masę
(93,90%) oraz zawartość białka (12,06%), włókna (15,33%), ekstraktu wolnego od azotu
(34,12%), popiołu (12.28%), oleju (19,21%). Badanie toksyczności ekstraktu wodnego z liści
Abrus precatorious podawanego dożołądkowo oraz dootrzewnowo przeprowadzono na
myszach szczepu Swiss. LD50 dla dożołądkowego oraz dootrzewnowego podawania wodnego
wyciągu z liści modligroszku pospolitego wynosiła odpowiednio 2558,9 mg/kg oraz
638 mg/kg masy ciała myszy. Dootrzewnowe podanie ekstraktu w ilości 10 mg/kg masy
ciała myszy powodowało wzrost łącznej objętości elementów morfotycznych krwi (PCV),
neutropenię oraz spadek poziomu aminotransferazy asparaginianowej. W przypadku dożołądkowego
podania wyciągu w ilości 50 mg/kg m.c. myszy odnotowano spadek łącznej
objętości elementów morfotycznych krwi (PCV), neutropenię, wzrost poziomu fosfatazy
alkalicznej oraz hipochloremię. Natomiast dożołądkowa dawka wodnego wyciągu w ilości
250 mg/kg m.c. myszy spowodowała wzrost masy ciała, neutropenię, spadek poziomu
aminotrasferazy asparaginianowej oraz alaninoaminotransferazy. Wszystkie badane dawki
powodowały limfocytozę oraz hiperkreatynemię i z tego powodu należy uznać, że wodny
wyciąg z liści Abrus precatorious jest toksyczny w dawkach 10, 50 oraz 250 mg/kg masy
ciała myszy
