    Cytokine gene polymorphisms in preterm infants with necrotising enterocolitis: genetic association study

    BACKGROUND The inflammatory cytokine cascade is implicated in the pathogenesis of necrotising enterocolitis (NEC). Genetic association studies of cytokine polymorphisms may help to detect molecular mechanisms that are causally related to the disease process. AIM To examine associations between the common genetic variants in candidate inflammatory cytokine genes and NEC in preterm infants. METHODS Multi-centre case-control and genetic association study. DNA samples were collected from 50 preterm infants with NEC and 50 controls matched for gestational age and ethnic group recruited to a multi-centre case-control study. Ten candidate single-nucleotide polymorphisms in cytokines previously associated with infectious or inflammatory diseases were genotyped. The findings were included in random-effects meta-analyses with data from previous genetic association studies. RESULTS All allele distributions were in Hardy-Weinberg equilibrium. None of the studied cytokine polymorphisms was significantly associated with NEC. Four previous genetic association studies of cytokine polymorphisms and NEC in preterm infants were found. Meta-analyses were possible for several single-nucleotide polymorphisms. These increased the precision of the estimates of effect size but did not reveal any significant associations. CONCLUSIONS The available data are not consistent with more than modest associations between these candidate cytokine variant alleles and NEC in preterm infants. Data from future association studies of these polymorphisms may be added to the meta-analyses to obtain more precise estimates of effects sizes.The study was funded by Tenovus (Scotland)

    Ab initio molecular dynamics study of manganese porphine hydration and interaction with nitric oxide

    The authors use ab initio molecular dynamics and the density functional theory+U (DFT+U) method to compute the hydration environment of the manganese ion in manganese (II) and manganese (III) porphines (MnP) dispersed in liquid water. These are intended as simple models for more complex water soluble porphyrins, which have important physiological and electrochemical applications. The manganese ion in Mn(II)P exhibits significant out-of-porphine plane displacement and binds strongly to a single H2O molecule in liquid water. The Mn in Mn(III)P is on average coplanar with the porphine plane and forms a stable complex with two H2O molecules. The residence times of these water molecules exceed 15 ps. The DFT+U method correctly predicts that water displaces NO from Mn(III)P-NO, but yields an ambiguous spin state for the MnP(II)-NO complex.Comment: 10 pages, 6 figure

    Anti-money laundering in the United Kingdom: new directions for a more effective regime

    Purpose: This paper aims to provide a more nuanced understanding of the effectiveness of the anti-money laundering (AML) regime in the UK and explore opportunities to improve policy and performance. Design/methodology/approach: Qualitative research design using semi-structured interviews and a focus group with practitioners from both public and private sectors. Findings: This paper identifies preventive measures are underfunded by the public sector; there is a disconnect between the regulatory requirement and the regulators’ supervisory approach leading to the ineffective application of the risk-based approach; and authorities have limited ability to stop low-utility reports. Increased collaboration across institutions and sectors, better utilisation of innovative technologies and a sustainable funding plan are needed to drive a collective response to money laundering. Research limitations/implications: Few practitioners in the industry have the knowledge and expertise to discuss the topic at a strategic level and participants were limited (n = 8). Practical implications: This paper adds to the growing corpus of research showing that the AML regime in the UK is ineffective and needs reform. Social implications: This paper encourages practitioners to improve the AML regime, this research contributes to the reform of the existing measures against financial crime. Originality/value: This paper presents new data from AML practitioners to provide a better understanding of the limitations of the AML regime in the UK

    Can we evaluate population screening strategies in UK general practice? A pilot randomised controlled trial comparing postal and opportunistic screening for genital chlamydial infection

    STUDY OBJECTIVE: To assess whether opportunistic and postal screening strategies for Chlamydia trachomatis can be compared with usual care in a randomised trial in general practice DESIGN: Feasibility study for a randomised controlled trial. SETTING: Three West of Scotland general medical practices: one rural, one urban/deprived and one urban/affluent. PARTICIPANTS: 600 women aged 16-30 years, 200 from each of three participating practices selected at random from a sample of West of Scotland practices that had expressed interest in the study. The women could opt out of the study. Those who did not were randomly assigned to one of three groups: postal screening, opportunistic screening or usual care. MAIN RESULTS: 38% (85/221) of the approached practices expressed interest in the study. Data were collected successfully from the 3 participating practices, although intensive support was required. There were considerable workload implications for staff, both in relation to implementing the screening strategies and managing the research process. 124 of the 600 women opted out of the study. During the four-month study period, 55% (81/146) of the control group attended their practice but none was offered screening. 59% (80/136) women in the opportunistic group attended their practice of whom 55% (44/80) were offered screening. Of those, 64% (28/44) accepted, representing 21% of the opportunistic group. 48% (59/124) of the postal group returned samples. CONCLUSION: A randomised controlled trial comparing postal and opportunistic screening for chlamydial infection in general practice is feasible, though resource intensive. There may be problems with generalising from screening trials in which patients may opt out from the offer of screening

    Magnetic Flux Braiding: Force-Free Equilibria and Current Sheets

    We use a numerical nonlinear multigrid magnetic relaxation technique to investigate the generation of current sheets in three-dimensional magnetic flux braiding experiments. We are able to catalogue the relaxed nonlinear force-free equilibria resulting from the application of deformations to an initially undisturbed region of plasma containing a uniform, vertical magnetic field. The deformations are manifested by imposing motions on the bounding planes to which the magnetic field is anchored. Once imposed the new distribution of magnetic footpoints are then taken to be fixed, so that the rest of the plasma must then relax to a new equilibrium configuration. For the class of footpoint motions we have examined, we find that singular and nonsingular equilibria can be generated. By singular we mean that within the limits imposed by numerical resolution we find that there is no convergence to a well-defined equilibrium as the number of grid points in the numerical domain is increased. These singular equilibria contain current "sheets" of ever-increasing current intensity and decreasing width; they occur when the footpoint motions exceed a certain threshold, and must include both twist and shear to be effective. On the basis of these results we contend that flux braiding will indeed result in significant current generation. We discuss the implications of our results for coronal heating.Comment: 13 pages, 12 figure

    Polar phonons and intrinsic dielectric response of the ferromagnetic insulating spinel CdCr2_2S4_4 from first principles

    We have studied the dielectric properties of the ferromagnetic spinel CdCr2_2S4_4 from first principles. Zone-center phonons and Born effective charges were calculated by frozen-phonon and Berry phase techniques within LSDA+U. We find that all infrared-active phonons are quite stable within the cubic space group. The calculated static dielectric constant agrees well with previous measurements. These results suggest that the recently observed anomalous dielectric behavior in CdCr2_2S4_4 is not due to the softening of a polar mode. We suggest further experiments to clarify this point

    Direct Urca neutrino rate in colour superconducting quark matter

    If deconfined quark matter exists inside compact stars, the primary cooling mechanism is neutrino radiation via the direct Urca processes d->u+e+antinu_e and u+e->d+nu_e. Below a critical temperature, T_c, quark matter forms a colour superconductor, one possible manifestation of which is a condensate of quark Cooper pairs in an electric-charge neutralising background of electrons. We compute the neutrino emission rate from such a phase, including charged pair-breaking and recombination effects, and find that on a material temperature domain below T_c the pairing-induced suppression of the neutrino emission rate is not uniformly exponential. If gapless modes are present in the condensed phase, the emissivity at low temperatures is moderately enhanced above that of completely unpaired matter. The importance of charged current pair-breaking processes for neutrino emission both in the fully gapped and partially gapped regimes is emphasised.Comment: 5 pages, 2 figures; to appear in Phys.
