    N-terminal pro-B-type natriuretic peptide and the prediction of primary cardiovascular events: results from 15-year follow-up of WOSCOPS

    <b>Aims:</b>To test whether N-terminal pro-B-type natriuretic peptide (NT-proBNP) was independently associated with, and improved the prediction of, cardiovascular disease (CVD) in a primary prevention cohort. <b>Methods and results:</b> In the West of Scotland Coronary Prevention Study (WOSCOPS), a cohort of middle-aged men with hypercholesterolaemia at a moderate risk of CVD, we related the baseline NT-proBNP (geometric mean 28 pg/mL) in 4801 men to the risk of CVD over 15 years during which 1690 experienced CVD events. Taking into account the competing risk of non-CVD death, NT-proBNP was associated with an increased risk of all CVD [HR: 1.17 (95% CI: 1.11–1.23) per standard deviation increase in log NT-proBNP] after adjustment for classical and clinical cardiovascular risk factors plus C-reactive protein. N-terminal pro-B-type natriuretic peptide was more strongly related to the risk of fatal [HR: 1.34 (95% CI: 1.19–1.52)] than non-fatal CVD [HR: 1.17 (95% CI: 1.10–1.24)] (P= 0.022). The addition of NT-proBNP to traditional risk factors improved the C-index (+0.013; P < 0.001). The continuous net reclassification index improved with the addition of NT-proBNP by 19.8% (95% CI: 13.6–25.9%) compared with 9.8% (95% CI: 4.2–15.6%) with the addition of C-reactive protein. N-terminal pro-B-type natriuretic peptide correctly reclassified 14.7% of events, whereas C-reactive protein correctly reclassified 3.4% of events. Results were similar in the 4128 men without evidence of angina, nitrate prescription, minor ECG abnormalities, or prior cerebrovascular disease. <b>Conclusion:</b> N-terminal pro-B-type natriuretic peptide predicts CVD events in men without clinical evidence of CHD, angina, or history of stroke, and appears related more strongly to the risk for fatal events. N-terminal pro-B-type natriuretic peptide also provides moderate risk discrimination, in excess of that provided by the measurement of C-reactive protein

    Effects of 1α,25 dihydroxyvitamin D3 and testosterone on miRNA and mRNA expression in LNCaP cells

    <p>Abstract</p> <p>Background</p> <p>There is evidence from epidemiological and <it>in vitro </it>studies that the biological effects of testosterone (T) on cell cycle and survival are modulated by 1,25-dihydroxyvitamin D<sub>3 </sub>(1,25(OH)<sub>2</sub>D<sub>3</sub>) in prostate cancer. To investigate the cross talk between androgen- and vitamin D-mediated intracellular signaling pathways, the individual and combined effects of T and 1,25(OH)<sub>2</sub>D<sub>3 </sub>on global gene expression in LNCaP prostate cancer cells were assessed.</p> <p>Results</p> <p>Stringent statistical analysis identifies a cohort of genes that lack one or both androgen response elements (AREs) or vitamin D response elements (VDREs) in their promoters, which are nevertheless differentially regulated by both steroids (either additively or synergistically). This suggests that mechanisms in addition to VDR- and AR-mediated transcription are responsible for the modulation of gene expression. Microarray analysis shows that fifteen miRNAs are also differentially regulated by 1,25(OH)<sub>2</sub>D<sub>3 </sub>and T. Among these miR-22, miR-29ab, miR-134, miR-1207-5p and miR-371-5p are up regulated, while miR-17 and miR-20a, members of the miR-17/92 cluster are down regulated. A number of genes implicated in cell cycle progression, lipid synthesis and accumulation and calcium homeostasis are among the mRNA targets of these miRNAs. Thus, in addition to their well characterized effects on transcription, mediated by either or both cognate nuclear receptors, 1,25(OH)<sub>2</sub>D<sub>3 </sub>and T regulate the steady state mRNA levels by modulating miRNA-mediated mRNA degradation, generating attenuation feedback loops that result in global changes in mRNA and protein levels. Changes in genes involved in calcium homeostasis may have specific clinical importance since the second messenger Ca<sup>2+ </sup>is known to modulate various cellular processes, including cell proliferation, cell death and cell motility, which affects prostate cancer tumor progression and responsiveness to therapy.</p> <p>Conclusions</p> <p>These data indicate that these two hormones combine to drive a differentiated phenotype, and reinforce the idea that the age dependent decline in both hormones results in the de-differentiation of prostate tumor cells, which results in increased proliferation, motility and invasion common to aggressive tumors. These studies also reinforce the potential importance of miRNAs in prostate cancer progression and therapeutic outcomes.</p

    On the Reliability of Cross Correlation Function Lag Determinations in Active Galactic Nuclei

    Many AGN exhibit a highly variable luminosity. Some AGN also show a pronounced time delay between variations seen in their optical continuum and in their emission lines. In effect, the emission lines are light echoes of the continuum. This light travel-time delay provides a characteristic radius of the region producing the emission lines. The cross correlation function (CCF) is the standard tool used to measure the time lag between the continuum and line variations. For the few well-sampled AGN, the lag ranges from 1-100 days, depending upon which line is used and the luminosity of the AGN. In the best sampled AGN, NGC 5548, the H_beta lag shows year-to-year changes, ranging from about 8.7 days to about 22.9 days over a span of 8 years. In this paper it is demonstrated that, in the context of AGN variability studies, the lag estimate using the CCF is biased too low and subject to a large variance. Thus the year-to-year changes of the measured lag in NGC 5548 do not necessarily imply changes in the AGN structure. The bias and large variance are consequences of finite duration sampling and the dominance of long timescale trends in the light curves, not due to noise or irregular sampling. Lag estimates can be substantially improved by removing low frequency power from the light curves prior to computing the CCF.Comment: To appear in the PASP, vol 111, 1999 Nov; 37 pages; 10 figure

    Discovery of Reflection Nebulosity Around Five Vega-like Stars

    Coronagraphic optical observations of six Vega-like stars reveal reflection nebulosities, five of which were previously unknown. The nebulosities illuminated by HD 4881, HD 23362, HD 23680, HD 26676, and HD 49662 resemble that of the Pleiades, indicating an interstellar origin for dust grains. The reflection nebulosity around HD 123160 has a double-arm morphology, but no disk-like feature is seen as close as 2.5 arcsec from the star in K-band adaptive optics data. We demonstrate that uniform density dust clouds surrounding HD 23362, HD 23680 and HD 123160 can account for the observed 12-100 micron spectral energy distributions. For HD 4881, HD 26676, and HD 49662 an additional emission source, such as from a circumstellar disk or non-equilibrium grain heating, is required to fit the 12-25 micron data. These results indicate that in some cases, particularly for Vega-like stars located beyond the Local Bubble (>100 pc), the dust responsible for excess thermal emission may originate from the interstellar medium rather than from a planetary debris system.Comment: The Astrophysical Journal, in press for March, 2002 (32 pages, 13 figures

    Local height probabilities in a composite Andrews-Baxter-Forrester model

    We study the local height probabilities in a composite height model, derived from the restricted solid-on-solid model introduced by Andrews, Baxter and Forrester, and their connection with conformal field theory characters. The obtained conformal field theories also describe the critical behavior of the model at two different critical points. In addition, at criticality, the model is equivalent to a one-dimensional chain of anyons, subject to competing two- and three-body interactions. The anyonic-chain interpretation provided the original motivation to introduce the composite height model, and by obtaining the critical behaviour of the composite height model, the critical behaviour of the anyonic chains is established as well. Depending on the overall sign of the hamiltonian, this critical behaviour is described by a diagonal coset-model, generalizing the minimal models for one sign, and by Fateev-Zamolodchikov parafermions for the other.Comment: 34 pages, 5 figures; v2: expanded introduction, references added and other minor change

    Abstract: The Middle East is a growing and lucrative marketplace. This exploratory study examines retail service in Kuwait and Lebanon, regions with long histories of trade. Retail service, however, has not been well documented in this region. To our knowledge, this is the first study that examines customer and salespeople perceptions of service encounters in these countries, in light of their culture, religion, and nationalities. As retailers expand into new markets worldwide, such information is vital to their success. Future research directions are discussed

    On the segmentation of astronomical images via level-set methods

    Astronomical images are of crucial importance for astronomers since they contain a lot of information about celestial bodies that can not be directly accessible. Most of the information available for the analysis of these objects starts with sky explorations via telescopes and satellites. Unfortunately, the quality of astronomical images is usually very low with respect to other real images and this is due to technical and physical features related to their acquisition process. This increases the percentage of noise and makes more difficult to use directly standard segmentation methods on the original image. In this work we will describe how to process astronomical images in two steps: in the first step we improve the image quality by a rescaling of light intensity whereas in the second step we apply level-set methods to identify the objects. Several experiments will show the effectiveness of this procedure and the results obtained via various discretization techniques for level-set equations.Comment: 24 pages, 59 figures, paper submitte

    Connectivity and Nitrate Uptake Potential of Intermittent Streams in the Northeast USA

    Non-perennial streams dominate the extent of stream networks worldwide. Intermittent streams can provide ecosystem services to the entire network—including nitrate uptake to alleviate eutrophication of coastal waters—and are threatened by lack of legal protection. We examined 12 intermittent streams in the temperate, humid climate of the Northeast USA. Over 3 years of monitoring, continuous flow was observed a median of 277 d yr−1, with no-flow conditions from early summer into fall. Estimated median discharge was 2.9 L s−1 or 0.36mm d−1. All intermittent streams originated from source wetlands (median area: 0.27 ha) and the median length of the intermittent stream from the source wetland to the downstream perennial stream was 344m. Through regional geospatial analysis with high resolution orthophotography, we estimated that widely available, “high resolution” (1:24,000) hydrography databases (e.g., NHDPlus HR) only displayed 43% of the total number of intermittent streams. Whole-stream gross nitrate-N uptake rates were estimated at six intermittent streams during continuous flow conditions using pulse additions of nitrate and a conservative tracer. These rates displayed high temporal variability (range: no detect to over 6,000mg N m−1 d−1); hot moments were noted in nine of the 65 pulse additions. Whole-stream gross nitrate-N uptake rates were significantly inversely related to discharge, with no measurable rates above 7 L s−1. Temperature was significantly positively correlated with whole-stream gross nitrate-N uptake rates, with more hot moments in the spring. Microbial assays demonstrated that nitrate cycling in intermittent streams are consistent with results from low order, perennial forested streams and highlighted the importance of debris dams and pools—potential locations for transient storage. Our assessment suggests that intermittent streams in our region may annually contribute 24–47% of the flow to perennial streams and potentially remove 4.1 to 80.4 kg nitrate-N km−2 annually. If development in these areas continues, perennial streams are in danger of losing a portion of their headwaters and potential nitrate uptake areas may become nitrate sources to downstream areas. These results argue to manage fluvial systems with a holistic approach that couples intermittent and perennial components

    Application of Volcano Plots in Analyses of mRNA Differential Expressions with Microarrays

    Volcano plot displays unstandardized signal (e.g. log-fold-change) against noise-adjusted/standardized signal (e.g. t-statistic or -log10(p-value) from the t test). We review the basic and an interactive use of the volcano plot, and its crucial role in understanding the regularized t-statistic. The joint filtering gene selection criterion based on regularized statistics has a curved discriminant line in the volcano plot, as compared to the two perpendicular lines for the "double filtering" criterion. This review attempts to provide an unifying framework for discussions on alternative measures of differential expression, improved methods for estimating variance, and visual display of a microarray analysis result. We also discuss the possibility to apply volcano plots to other fields beyond microarray.Comment: 8 figure
