    Design and Expression of Specific Hybrid Lantibiotics Active Against Pathogenic Clostridium spp.

    Clostridium difficile has been reported as the most common cause of nosocomial diarrhea (antibiotic-associated diarrhea), resulting in significant morbidity and mortality in hospitalized patients. The resistance of the clostridia' spores to antibiotics and their side effects on the gut microbiota are two factors related to the emergence of infection and its relapses. Lantibiotics provide an innovative alternative for cell growth inhibition due to their dual mechanism of action (membrane pore-forming and cell wall synthesis inhibition) and low resistance rate. Based on the fact that bacteriocins are usually active against bacteria closely related to the producer strains, a new dual approach combining genome mining and synthetic biology was performed, by designing new lantibiotics with high activity and specificity toward Clostridium. We first attempted the heterologous expression of putative lantibiotics identified following Clostridium genome mining. Subsequently, we designed new hybrid lantibiotics combining the start or end of the putative clostridial peptides and the start or end parts of nisin. The designed peptides were cloned and expressed using the nisin biosynthetic machinery in Lactococcus lactis. From the 20 initial peptides, only 1 fulfilled the requirements established in this work to be considered as a good candidate: high heterologous production level and high specificity/activity against clostridial species. The high specificity and activity observed for the peptide AMV10 makes it an interesting candidate as an alternative to traditional antibiotics in the treatment of C. difficile infections, avoiding side effects and protecting the normal gut microbiota

    Field induced magnetic transition and metastability in Co substituted Mn2SbMn_{2}Sb

    A detailed investigation of first order ferrimagnetic (FRI) to antiferromagnetic (AFM) transition in Co (15%) doped Mn2SbMn_2Sb is carried out. These measurements demonstrate anomalous thermomagnetic irreversibility and glass-like frozen FRI phase at low temperatures. The irreversibility arising between the supercooling and superheating spinodals is distinguised in an ingenious way from the irreversibility arising due to kinetic arrest. Field annealing measurements shows reentrant FRI-AFM-FRI transition with increasing temperature. These measurements also show that kinetic arrest band and supercooling band are anitcorrelated i.e regions which are kinetically arrested at higher temperature have lower supercooling temperature and vice versa.Comment: 10 pages, 8 figure

    Interleukin-7, a New Cytokine Targeting the Mouse Hypothalamic Arcuate Nucleus: Role in Body Weight and Food Intake Regulation

    Body weight is controlled through peripheral (white adipose tissue) and central (mainly hypothalamus) mechanisms. We have recently obtained evidence that overexpression of interleukin (IL)-7, a critical cytokine involved in lymphopoiesis, can protect against the development of diet-induced obesity in mice. Here we assessed whether IL-7 mediated its effects by modulating hypothalamic function. Acute subcutaneous injection of IL-7 prevented monosodium glutamate-induced obesity, this being correlated with partial protection against cell death in the hypothalamic arcuate nucleus (ARC). Moreover, we showed that IL-7 activated hypothalamic areas involved in regulation of feeding behavior, as indicated by induction of the activation marker c-Fos in neural cells located in the ventromedial part of the ARC and by inhibition of food intake after fasting. Both chains of the IL-7 receptor (IL-7Rα and γc) were expressed in the ARC and IL-7 injection induced STAT-3 phosphorylation in this area. Finally, we established that IL-7 modulated the expression of neuropeptides that tune food intake, with a stimulatory effect on the expression of pro-opiomelanocortin and an inhibitory effect on agouti-related peptide expression in accordance with IL-7 promoting anorectic effects. These results suggest that the immunomodulatory cytokine IL-7 plays an important and unappreciated role in hypothalamic body weight regulation

    FIBONACCI SUPERLATTICES OF NARROW-GAP III-V SEMICONDUCTORS

    We report theoretical electronic structure of Fibonacci superlattices of narrow-gap III-V semiconductors. Electron dynamics is accurately described within the envelope-function approximation in a two-band model. Quasiperiodicity is introduced by considering two different III-V semiconductor layers and arranging them according to the Fibonacci series along the growth direction. The resulting energy spectrum is then found by solving exactly the corresponding effective-mass (Dirac-like) wave equation using tranfer-matrix techniques. We find that a self-similar electronic spectrum can be seen in the band structure. Electronic transport properties of samples are also studied and related to the degree of spatial localization of electronic envelope-functions via Landauer resistance and Lyapunov coefficient. As a working example, we consider type II InAs/GaSb superlattices and discuss in detail our results in this system.Comment: REVTeX 3.0, 16 pages, 8 figures available upon request.     The demand for agricultural products has been increasing in all countries in the last decades. Together with the decrease in natural resources and the possible impacts to agriculture caused by climatic changes, this aspect has aroused concerns in terms of the development of new global agricultural frontiers. In this context, some regions in Africa have been drawing attention from the world due to their edaphoclimatic conditions, which are favorable for agricultural development. The vision of a modern and dynamic agriculture must be based on the capacity of applying appropriate knowledge and technologies to the environment, as well as to the countryÂŽs economic and social situation. Such condition demands the installment of an efficient agricultural innovation system composed by a technical and operational structure which is able to support the agricultural industry along all its stages. Thus, this article aims at presenting some contributions to the formatting of a geographic database of natural resources for the Nacala Corridor, Mozambique. Geotechnology instruments were used for planning, generating and maintaining the accomplished database's data and information. This database will subsidize the integrated management of the region's geographic space from spatial information about landscape, soil, climate, land use, and land cover, thus subsidizing the management of the natural resources and of Mozambique's agricultural sustainability as well

    On the Prevalence, Impact, and Evolution of SQL code smells in Data-Intensive Systems

    ABSTRACT: Code smells indicate software design problems that harm software quality. Data-intensive systems that frequently access databases often suffer from SQL code smells besides the traditional smells. While there have been extensive studies on traditional code smells, recently, there has been a growing interest in SQL code smells. In this paper, we conduct an empirical study to investigate the prevalence and evolution of SQL code smells in open-source, data-intensive systems. We collected 150 projects and examined both traditional and SQL code smells in these projects. Our investigation delivers several important findings. First, SQL code smells are indeed prevalent in data-intensive software systems. Second, SQL code smells have a weak co-occurrence with traditional code smells. Third, SQL code smells have a weaker association with bugs than that of traditional code smells. Fourth, SQL code smells are more likely to be introduced at the beginning of the project lifetime and likely to be left in the code without a fix, compared to traditional code smells. Overall, our results show that SQL code smells are indeed prevalent and persistent in the studied data-intensive software systems. Developers should be aware of these smells and consider detecting and refactoring SQL code smells and traditional code smells separately, using dedicated tools

    A global Carleman estimate in a transmission wave equation and application to a one-measurement inverse problem

    We consider a transmission wave equation in two embedded domains in R2R^2, where the speed is a1>0a1 > 0 in the inner domain and a2>0a2 > 0 in the outer domain. We prove a global Carleman inequality for this problem under the hypothesis that the inner domain is strictly convex and a1>a2a1 > a2 . As a consequence of this inequality, uniqueness and Lip- schitz stability are obtained for the inverse problem of retrieving a stationary potential for the wave equation with Dirichlet data and discontinuous principal coefficient from a single time-dependent Neumann boundary measurement

    Multiplexed immunosensors for point-of-care diagnostic applications

    Accurate, reliable, and cost-effective immunosensors are clinically important for the early diagnosis and monitoring of progressive diseases, and multiplexed sensing is a promising strategy for the next generation of diagnostics. This strategy allows for the simultaneous detection and quantification of multiple biomarkers with significantly enhanced reproducibility and reliability, whilst requiring smaller sample volumes, fewer materials, and shorter average analysis time for individual biomarkers than individual tests. In this opinionated review, we compare different techniques for the development of multiplexed immunosensors. We review the state-of-the-art approaches in the field of multiplexed immunosensors using electrical, electrochemical, and optical methods. The barriers that prevent translating this sensing strategy into clinics are outlined together with the potential solutions. We also share our vision on how multiplexed immunosensors will continue their evolution in the coming years
