    Histological investigations on the thyroid glands of marine mammals (Phoca vitulina, Phocoena phocoena) and the possible implications of marine pollution

    In 1988 and 1989, thousands of harbor seals (Phoca vitulina) died in the North Sea from phocine distemper infection. The morphology of thyroid glands from 40 harbor seals found dead on the North Sea coastlines of Schleswig-Holstein, Federal Republic of Germany, during an epizootic of phocine distemper, was compared with the morphology of thyroid glands from five healthy harbor seals collected in Iceland. Thyroid glands from seven harbor porpoises (Phocoena phocoena) found dead in 1990 on the North Sea coastlines also were evaluated. Colloid depletion and fibrosis were found in the thyroid glands of harbor seals which died during the epizootic, but not in animals from Iceland. Thyroid glands of the porpoises showed similar lesions, but to a lesser degree, than those observed in the North Sea seals

    En este trabajo se presentan los resultados de la implementación de una etapa de una Secuencia de Enseñanza y Aprendizaje diseñada para abordar la Teoría Especial de la Relatividad en la escuela secundaria superior, en Argentina. Se adopta el modelo de Enseñanza para la Comprensión dentro de un marco teórico más amplio que promueve un aprendizaje significativo de la Teoría Especial de la Relatividad.El objetivo de esta investigación es analizar si la utilización de los diagramas de Minkowski, para establecer la simultaneidad de eventos en la Teoría Especial de la Relatividad, es una herramienta apropiada para la escuela secundaria.La metodología empleada en la investigación es cualitativa de tipo descriptiva. La implementación se realizó en dos cursos de secundaria superior de una escuela pública dependiente de la Universidad Nacional del Centro, conformados por 65 alumnos. Los resultados obtenidos evidencian rasgos de aprendizaje significativo de la Teoría Especial de la Relatividad, por parte de los alumnos.In this paper the results of the implementation of a step in a Teaching - Learning sequence designed to approach the Special Relativity Theory in high school in Argentina are presented. The purpose of this research is to analyze whether the use of Minkowski diagrams to establish the simultaneity of events in the Special Relativity Theory is an appropriate tool for high school. The methodology used in this research is qualitative and descriptive. The implementation was done in two courses of a public high school that is dependent of the Universidad Nacional del Centro (Argentina), with 65 students.     Embryonic Stem Cell-Derived Neurons Grown on Multi-Electrode Arrays as a Novel In vitro Bioassay for the Detection of Clostridium botulinum Neurotoxins

    Clostridium botulinum neurotoxins (BoNTs) are the most poisonous naturally occurring protein toxins known to mankind and are the causative agents of the severe and potentially life-threatening disease botulism. They are also known for their application as cosmetics and as unique bio-pharmaceuticals to treat an increasing number of neurological and non-neurological disorders. Currently, the potency of biologically active BoNT for therapeutic use is mainly monitored by the murine LD50-assay, an ethically disputable test causing suffering and death of a considerable number of mice. The aim of this study was to establish an in-vitro assay as an alternative to the widely used in-vivo mouse bioassay. We report a novel BoNT detection assay using mouse embryonic stem cell-derived neurons (mESN) cultured on multi-electrode arrays. After 21 days in culture, the mESN formed a neuronal network showing spontaneous bursting activity based on functional synapses and express the necessary target proteins for BoNTs. Treating cultures for 6 h with 16.6 pM of BoNT serotype A and incubation with 1.66 pM BoNT/A or 33 Units/ml of Botox® for 24 h lead to a significant reduction of both spontaneous network bursts and average spike rate. This data suggests that mESN cultured on multi-electrode arrays pose a novel, biologically relevant model that can be used to detect and quantify functional BoNT effects, thus accelerating BoNT research while decreasing animal use

    Orbitally Driven Spin Pairing in the 3D Non-Magnetic Mott Insulator BaVS3: Evidence from Single Crystal Studies

    Static electrical and magnetic properties of single crystal BaVS_3 were measured over the structural (T_S=240K), metal-insulator (T_MI=69K), and suspected orbital ordering (T_X=30K) transitions. The resistivity is almost isotropic both in the metallic and insulating states. An anomaly in the magnetic anisotropy at T_X signals a phase transition to an ordered low-T state. The results are interpreted in terms of orbital ordering and spin pairing within the lowest crystal field quasi-doublet. The disordered insulator at T_X<T<T_MI is described as a classical liquid of non-magnetic pairs.Comment: 4 pages, 5 figures, revtex, epsf, and multicol style. Problem with figures fixed.     Production of a Fermi gas of atoms in an optical lattice

    We prepare a degenerate Fermi gas of potassium atoms by sympathetic cooling with rubidium atoms in a one-dimensional optical lattice. In a tight lattice we observe a change of the density of states of the system, which is a signature of quasi two dimensional confinement.     Structural basis for c-di-AMP–dependent regulation of the bacterial stringent response by receptor protein DarB

    The bacterial second messenger c-di-AMP controls essential cellular processes, including potassium and osmolyte homeostasis. This makes synthesizing enzymes and components involved in c-di-AMP signal transduction intriguing as potential targets for drug development. The c-di-AMP receptor protein DarB of Bacillus subtilis binds the Rel protein and triggers the Rel-dependent stringent response to stress conditions; however, the structural basis for this trigger is unclear. Here, we report crystal structures of DarB in the ligand-free state and of DarB complexed with c-di-AMP, 3′3′-cGAMP, and AMP. We show that DarB forms a homodimer with a parallel, head-to-head assembly of the monomers. We also confirm the DarB dimer binds two cyclic dinucleotide molecules or two AMP molecules; only one adenine of bound c-di-AMP is specifically recognized by DarB, while the second protrudes out of the donut-shaped protein. This enables DarB to bind also 3′3′-cGAMP, as only the adenine fits in the active site. In absence of c-di-AMP, DarB binds to Rel and stimulates (p)ppGpp synthesis, whereas the presence of c-di-AMP abolishes this interaction. Furthermore, the DarB crystal structures reveal no conformational changes upon c-di-AMP binding, leading us to conclude the regulatory function of DarB on Rel must be controlled directly by the bound c-di-AMP. We thus derived a structural model of the DarB–Rel complex via in silico docking, which was validated with mass spectrometric analysis of the chemically crosslinked DarB–Rel complex and mutagenesis studies. We suggest, based on the predicted complex structure, a mechanism of stringent response regulation by c-di-AMP

    Investigation of the thermal stability of Mg/Co periodic multilayers for EUV applications

    We present the results of the characterization of Mg/Co periodic multilayers and their thermal stability for the EUV range. The annealing study is performed up to a temperature of 400\degree C. Images obtained by scanning transmission electron microscopy and electron energy loss spectroscopy clearly show the good quality of the multilayer structure. The measurements of the EUV reflectivity around 25 nm (~49 eV) indicate that the reflectivity decreases when the annealing temperature increases above 300\degreeC. X-ray emission spectroscopy is performed to determine the chemical state of the Mg atoms within the Mg/Co multilayer. Nuclear magnetic resonance used to determine the chemical state of the Co atoms and scanning electron microscopy images of cross sections of the Mg/Co multilayers reveal changes in the morphology of the stack from an annealing temperature of 305\degreee;C.
