    94/Międzybłoniak opłucnej – diagnostyka i leczenie w materiale Centrum

    Celem pracyjest prezentacja wyników leczenia chorych na międzybłoniaka opłucnej [MO] leczonych w latach 1965–2001 oraz ocena roli radioterapii w zapobieganiu niepowodzeniom miejscowym.Materiał i metodyW latach 1965–2001 w Centrum Onkologii w Krakowie leczono 75 chorych z powodu [MO]. Analizą objęto grupę 34 chorych leczonych od 1991 roku, u których postępowanie diagnostyczne i terapeutyczne było jednolite. U wszystkich chorych zastosowano leczenie operacyjne, które u 7 chorych miało charakter radykalny (pleuro-pericardio-diaphra-gmo-pneumonectomia). Pooperacyjna radioterapia zastosowana była u 23 chorych, z czego 14 chorych (61%) napromienianych było wiązką mieszaną fotonowo-elektronową. Zakres dawki podanej na teren jamy opłucnej wynosił od 45 do 55 Gy. W oszacowaniu odsetków przeżyć 1, 2 i 3-letnich zastosowano metodę Kaplana-Meiera. Wyniki uzyskane w grupie leczonej od 1991 roku porównano z wynikami leczenia grupy chorych leczonych w latach 1965–1990.Wyniki i wnioskiW analizowej grupie uzyskano nastęujące odsetki przeżyć: 1-rocznych 52.9%, 2-letnich 26.5%, 3-letnich 6.6%. Tolerancja leczenia była dobra. Stwierdzono, że wyniki te są lepsze w stosunku do grupy chorych leczonych do 1991 r. Różnice te dotyczą okresu 2 lat po zakończeniu leczenia. Zastosowanie u chorych na [MO] mieszanej wiązki fotonowoelektronowej pozwala na uzyskanie stosunkowo dobrych wyników leczenia przy dobrej tolerancji leczenia przez tych chorych

    Arctic, especially western coast of Spitsbergen has been a field of taking research by scientist from UMCS and AR in Lublin for more than 15 years. Researches have been taken there are connected with earth sciences like: meteorology and climatology, geomorphology, hydrography, geology and agriculture and botany. Meteorological observations were steady point of researches taking on every UMCS?s trips in Spitsbergen. At the beginning, at the 1986-1988 seasons were very wide, and measurements were taken every 3 hours. Between 1989 and 1999 measurements were taken 4 times a day a.e. every 6 hours. Since 1999 measurements have been doing by automatic stations. Appliance this equipment let us taking measurements with 10 minutes time step. The aim of this paper is to present course of meteorological conditions during the scientific trip to Spitsbergen in 2001 at the background of period 1986-2000. There were made comparisons of course mean daily four meteorological elements as: air temperature measured at 200 cm height [°C], wind speed [m/s], cloudiness, and precipitation [mm] from 2001 season with data collected during 15-years UMCS geographical trips on Spitsbergen. We can make statement that in thermic season 2001 was a bit cooler (0.1°C) than the period 1986-2000. At that season variability of mean daily temperatures was lower than in period. The lowest mean daily temperature was 2.5°C, and all days in period 2001 were lukewarm. The rate of mean daily cloudiness in period 1986-2000 is compensated. 2001 season was the same as other seasons during period. The air masses are strongly modified by circulation and orography. Season of 2001 was featured by lower mean daily wind speed (3.2 m/s) than in the period ? 4 m/s, also highest mean daily wind speed in 2001 was lower than in period 1986-2000. The greatest variability of four analysed elements on that period has precipitation. The variable of this element exceed 71.7 mm, which we can express as a ratio 1 to 20. 2001 season was dryer than mean of period about 11 mm

    Kallikrein-Related Peptidase 14 Activates Zymogens of Membrane Type Matrix Metalloproteinases (MT-MMPs)—A CleavEx Based Analysis

    Kallikrein-related peptidases (KLKs) and matrix metalloproteinases (MMPs) are secretory proteinases known to proteolytically process components of the extracellular matrix, modulating the pericellular environment in physiology and in pathologies. The interconnection between these families remains elusive. To assess the cross-activation of these families, we developed a peptide, fusion protein-based exposition system (Cleavage of exposed amino acid sequences, CleavEx) aiming at investigating the potential of KLK14 to recognize and hydrolyze proMMP sequences. Initial assessment identified ten MMP activation domain sequences which were validated by Edman degradation. The analysis revealed that membrane-type MMPs (MT-MMPs) are targeted by KLK14 for activation. Correspondingly, proMMP14-17 were investigated in vitro and found to be effectively processed by KLK14. Again, the expected neo-N-termini of the activated MT-MMPs was confirmed by Edman degradation. The effectiveness of proMMP activation was analyzed by gelatin zymography, confirming the release of fully active, mature MT-MMPs upon KLK14 treatment. Lastly, MMP14 was shown to be processed on the cell surface by KLK14 using murine fibroblasts overexpressing human MMP14. Herein, we propose KLK14-mediated selective activation of cell-membrane located MT-MMPs as an additional layer of their regulation.
