The SNARE protein FolVam7 mediates intracellular trafficking to regulate conidiogenesis and pathogenicity in Fusarium oxysporum f. sp. lycopersici.
Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) are conserved in fungi, plants and animals. The Vam7 gene encodes a v-SNARE protein that involved in vesicle trafficking in fungi. Here, we identified and characterized the function of FolVam7, a homologue of the yeast SNARE protein Vam7p in Fusarium oxysporum f. sp. lycopersici (Fol), a fungal pathogen of tomato. FolVam7 contains SNARE and PX (Phox homology) domains that are indispensable for normal localization and function of FolVam7. Targeted gene deletion showed that FolVam7-mediated vesicle trafficking is important for vegetative growth, asexual development, conidial morphology and plant infection. Further cytological examinations revealed that FolVam7 is localized to vesicles and vacuole membranes in the hyphae stage. Moreover, the ΔFolvam7 mutant is insensitive to salt and osmotic stresses and hypersensitive to cell wall stressors. Coupled Photonic Crystal Micro-Cavities With Ultra-Low Threshold Power For Stiumulated Raman Scattering
We propose coupled cavities to realize a strong enhancement of the Raman scattering. Five sub cavities are embedded in the photonic crystals. Simulations through finite-difference time-domain (FDTD) method demonstrate that one cavity, which is used to propagate the pump beam at the optical-communication wavelength, has a Q factor as high as 1.254 × 108 and modal volume as small as 0.03μm3 (0.3192(λ/n)3). These parameters result in ultra-small threshold lasing power ~17.7nW and 2.58nW for Stokes and anti-Stokes respectively. The cavities are designed to support the required Stokes and anti-Stokes modal spacing in silicon. The proposed structure has the potential for sensor devices, especially for biological and medical diagnoses
Study on wind resistance coefficient of double split wire arrangement
Shape coefficients of a single traverse and twin branch lines are examined through wind tunnel experiments and numerical simulations under different angle of attacks and wind speeds. It was found that the shape coefficient at angle of attack equaling to 0 is obviously smaller than the others. And the effects of Reynolds number are also large. The numerical results give difference between cases reaching 20Â %, however, the experimental ones only 1.17Â %. It is because that the initial condition of CFD is idealized but the wind tunnel experiment is closer to the reality

Vertical Federated Learning (VFL) is a federated learning setting where
multiple parties with different features about the same set of users jointly
train machine learning models without exposing their raw data or model
parameters. Motivated by the rapid growth in VFL research and real-world
applications, we provide a comprehensive review of the concept and algorithms
of VFL, as well as current advances and challenges in various aspects,
including effectiveness, efficiency, and privacy. We provide an exhaustive
categorization for VFL settings and privacy-preserving protocols and
comprehensively analyze the privacy attacks and defense strategies for each
protocol. In the end, we propose a unified framework, termed VFLow, which
considers the VFL problem under communication, computation, privacy, and
effectiveness constraints. Finally, we review the most recent advances in
industrial applications, highlighting open challenges and future directions for
VFL
Clinicopathological and prognostic value of epithelial cell adhesion molecule in solid tumours: a meta-analysis
BackgroundMalignant tumors, mainly solid tumors, are a significant obstacle to the improvement of life expectancy at present. Epithelial cell adhesion molecule (EpCAM), a cancer stem cell biomarker, showed widespread expression in most normal epithelial cells and most cancers. Although the clinical significance of EpCAM in various malignant solid tumors has been studied extensively, the latent relationships between EpCAM and pathological and clinical characteristics in solid tumors and differences in the roles of EpCAM among tumors have not been clearly determined. The destination point of this study was to analyze the value of EpCAM in solid tumors in clinicopathological and prognostic dimension using a meta-analysis approach.Method and materialsA comprehensive and systematic search of the researches published up to March 7th, 2022, in PubMed, EMBASE, Web of Science, Cochrane library and PMC databases was performed. The relationships between EpCAM overexpression, clinicopathological characteristics, and survival outcomes were analyzed. Pooled hazard ratios (HRs) with 95% confidence intervals (CIs) and odds ratios (ORs) were estimated as indicators of the degree of correlation. This research was registered on PROSPERO (International prospective register of systematic reviews), ID: CRD42022315070.ResultsIn total, 57 articles and 14184 cases were included in this study. High EpCAM expression had a significant coherence with a poorer overall survival (OS) (HR: 1.30, 95% CI: 1.08–1.58, P < 0.01) and a worse disease-free survival (DFS) (HR: 1.58, 95% CI: 1.28–1.95, P < 0.01), especially of gastrointestinal tumors’ OS (HR: 1.50, 95% CI: 1.15–1.95, P < 0.01), and DFS (HR: 1.84, 95% CI: 1.52–2.33, P < 0.01). The DFS of head and neck tumors (HR: 2.33, 95% CI: 1.51–3.61, P < 0.01) was also associated with the overexpression of EpCAM. There were no positive relationships between the overexpression of EpCAM and sex (RR: 1.03, 95% CI: 0.99–1.07, P = 0.141), T classification (RR: 0.93, 95% CI: 0.82–1.06, P = 0.293), lymph node metastasis (RR: 0.85, 95% CI: 0.54–1.32, P = 0.461), distant metastasis (RR: 0.97, 95% CI: 0.84–1.10, P = 0.606), vascular infiltration (RR: 1.05, 95% CI: 0.85–1.29, P = 0.611), and TNM stage (RR: 0.93, 95% CI: 0.83–1.04, P = 0.187). However, the overexpression of EpCAM exhibited a significant association with the histological grades (RR: 0.88, 95% CI: 0.80–0.97, P < 0.01).ConclusionBased on pooled HRs, the positive expression of EpCAM was totally correlated to a worse OS and DFS in solid tumors. The expression of EpCAM was related to a worse OS in gastrointestinal tumors and a worse DFS in gastrointestinal tumors and head and neck tumors. Moreover, EpCAM expression was correlated with the histological grade. INTEnsive care bundle with blood pressure reduction in acute cerebral hemorrhage trial (INTERACT3): Study protocol for a pragmatic stepped-wedge cluster-randomized controlled trial
Background: Early intensive blood pressure (BP) lowering remains the most promising treatment for acute intracerebral hemorrhage (ICH), despite discordant results between clinical trials and potential variation in the treatment effects by approach to control BP. As the third in a series of clinical trials on this topic, the INTEnsive care bundle with blood pressure Reduction in Acute Cerebral hemorrhage Trial (INTERACT3) aims to determine the effectiveness of a goal-directed care bundle protocol of early physiological control (intensive BP lowering, glycemic control, and pyrexia treatment) and reversal of anticoagulation, in acute ICH.Methods: INTERACT3 is a pragmatic, international, multicenter, stepped-wedge (4 phases/3 steps), cluster-randomized controlled trial to determine the effectiveness of a multifaceted care package in adult (age ≥ 18 years) patients (target 8360) with acute ICH (\u3c 6 h of onset) recruited from 110 hospitals (average of 19 consecutive patients per phase) in low- and middle-income countries. After a control phase, each hospital implements the intervention (intensive BP lowering, target systolic \u3c 140 mmHg; glucose control, target 6.1-7.8 mmol/L and 7.8-10.0 mmol/L in those without and with diabetes mellitus, respectively; anti-pyrexia treatment to target body temperature ≤ 37.5 °C; and reversal of anticoagulation, target international normalized ratio \u3c 1.5 within 1 h). Information will be obtained on demographic and baseline clinical characteristics, in-hospital management, and 7-day outcomes. Central trained blinded assessors will conduct telephone interviews to assess physical function and health-related quality of life at 6 months. The primary outcome is the modified Rankin scale (mRS) at 6 months analyzed using ordinal logistic regression. The sample size of 8360 subjects provides 90% power (α = 0.05) to detect a 5.6% absolute improvement (shift) in the primary outcome of the intervention versus control standard care, with various assumptions.Discussion: As the largest clinical trial in acute ICH, INTERACT3 is on schedule to provide an assessment of the effectiveness of a widely applicable goal-directed care bundle for a serious condition in which a clearly proven treatment has yet to be established.Trial registration: ClinicalTrials.gov NCT03209258. HMGA2 promotes nasopharyngeal carcinoma progression and is associated with tumor resistance and poor prognosis
Nasopharyngeal carcinoma (NPC), as one of the most prevalent malignancies in the head and neck region, still lacks a complete understanding of its pathogenesis. Presently, radiotherapy, concurrent chemoradiotherapy, and targeted therapy stand as the primary modalities for treating NPC. With advancements in medicine, the cure rates for nasopharyngeal carcinoma have been steadily increasing. Nevertheless, recurrence and metastasis persist as the primary reasons for treatment failure. Consequently, a profound exploration of the molecular mechanisms underlying the occurrence and progression of nasopharyngeal carcinoma, along with the exploration of corresponding therapeutic approaches, becomes particularly imperative in the quest for comprehensive solutions to combat this disease. High mobility group AT-hook 2 (HMGA2) is a pivotal protein capable of altering chromatin structure, regulating gene expression, and influencing transcriptional activity. In the realm of cancer research, HMGA2 exhibits widespread dysregulation, playing a crucial role in nearly all malignant tumors. It is implicated in various tumorigenic processes, including cell cycle regulation, cell proliferation, epithelial-mesenchymal transition, angiogenesis, tumor invasion, metastasis, and drug resistance. Additionally, HMGA2 serves as a molecular marker and an independent prognostic factor in certain malignancies. Recent studies have increasingly unveiled the critical role of HMGA2 in nasopharyngeal carcinoma (NPC), particularly in promoting malignant progression, correlating with tumor resistance, and serving as an independent adverse prognostic factor. This review focuses on elucidating the oncogenic role of HMGA2 in NPC, suggesting its potential association with chemotherapy resistance in NPC, and proposing its candidacy as an independent factor in nasopharyngeal carcinoma prognosis assessment
