    Review on Tomato (Solanum lycopersicum, L.) Improvement Programmes in Ghana

    Tomato is an important component of every Ghanaian meal, and its cultivation contributes significantly to livelihood improvement. The demand for tomato in Ghana outstrips supply, and therefore local production is augmented by imports from neighbouring countries. Despite the importance of tomato in Ghana, past tomato-breeding programmes have been unsystematic and had not led to the development of new varieties that meet the needs of consumers as well as environmental stresses. This review outlined tomato production trends, constraints and past tomato improvement programmes in Ghana, which mainly focused on germplasm collection, morphological and agronomic characterization, molecular evaluation, diversity study, as well as screening germplasm against biotic and abiotic stresses. The established variability and the outcomes of the evaluations against the various biotic and abiotic stresses have not been utilized in the development of new varieties. This work will serve as a reference for developing future tomato-breeding programmes

    Marker-Assisted Selection (MAS): A Fast-Track Tool in Tomato Breeding

    Marker-assisted selection (MAS) is a complementary tool for conventional breeding where a molecular marker linked to a trait is indirectly selected. Many studies conducted have been able to identify and develop markers for traits such as disease and pest resistance and other abiotic stresses. Despite the availability of these markers, the technology has been extensively used in tomato breeding for the identification of some economic traits in particular disease resistance. In developed countries, MAS is utilized routinely in breeding programs, but this cannot be said for developing countries such as Africa. It is high time Africa as a continent looks at the importance of the technology and invests in it. In addition to MAS, other strategies such as marker-assisted backcrossing and recurrent selection have also been employed for breeding in tomato. The use of MAS in crop improvement will not only reduce the cost of developing new tomato varieties but will also increase the precision and efficiency of selection in the breeding program as well as lessen the number of years required to come up with a new crop variety

    Profile and outcomes of hospitalized patients with COVID-19 at a tertiary institution hospital in Ghana

    Background: In high-income countries, mortality related to hospitalized patients with the Coronavirus disease 2019 (COVID-19) is approximately 4-5%. However, data on COVID-19 admissions from sub-Saharan Africa are scanty.Objective: To describe the clinical profile and determinants of outcomes of patients with confirmed COVID-19 admitted at a hospital in Ghana.Methods: A prospective study involving 25 patients with real time polymerase chain reaction confirmed COVID-19 admitted to the treatment centre of the University Hospital, Kwame Nkrumah University of Science and Technology (KNUST), Kumasi, Ghana from 1st June to 27th July, 2020. They were managed and followed up for outcomes. Data were analysed descriptively, and predictors of mortality assessed using a multivariate logistic regression modelling.Results: The mean age of the patients was 59.3 ± 20.6 years, and 14 (56%) were males. The main symptoms at presentation were breathlessness (68%) followed by fever (56%). The cases were categorized as mild (6), moderate (6), severe (10) and critical (3). Hypertension was the commonest comorbidity present in 72% of patients. Medications used in patient management included dexamethasone (68%), azithromycin (96%), and hydroxychloroquine (4%). Five of 25 cases died (Case fatality ratio 20%). Increasing age and high systolic blood pressure were associated with mortality.Conclusion: Case fatality in this sample of hospitalized COVID-19 patients was high. Thorough clinical assessment, severity stratification, aggressive management of underlying co-morbidities and standardized protocols incountry might improve outcomes

    Incidence, prevalence and severity of fall armyworm infestation in Ghana: a case of two maize enclaves in the Ashanti Region of Ghana

    Maize production is an important enterprise in Ghana, providing livelihood for thousands of people. It is challenged by fall armyworm infestation causing destruction to many cultivated lands in production enclaves and threatening food security. Effective control, however, requires information on the actual pest infestation as well as the effect of climate factors on infestation in these enclaves. The study assessed the incidence, prevalence and severity of fall armyworm infestation on maize farms in two major maize enclaves (Ejura and Ejisu) in Ghana. Data was taken on the presence, infestation levels and damage of fall armyworms as well as the climatic conditions in each district. Data collection was done by sampling 50 maize plants each on 40 maize farms in both districts and assessing them for the incidence, larval prevalence, leaf damage and severity. Results showed varying infestation in both districts (p < 0.0001) with Ejisu having a higher prevalence (0.10 ± 0.04 larvae per plant) than Ejura (0.05 ± 0.03 larvae per plant) in the minor season at seedling stage. At the vegetative stage, Ejisu recorded a higher prevalence in both seasons. A low severity was recorded at the seedling stage in both districts for all seasons which, however, varied among seasons at vegetative stage. Climatic variables including rainfall, temperature, relative humidity and wind were found to significantly impact infestation in both districts. The study thus, showed fall armyworm infestation to be a major challenge to maize production in both districts confirming it as a major constraint to maize production in the region

    Effect of gold mining on total factor productivity of farmers: Evidence from Ghana

    Gold mining comes with several benefits to developing countries, manifested mainly in the form of employment and revenue, but simultaneously impacts negatively on the immediate environment. It affects the economic structure including agriculture and its productivity. Hence, this study investigated the effect of gold mining on total factor productivity of farmers in Ghana using 110 cocoa farmers from Asutifi North and Asutifi South districts of the Brong Ahafo Region, categorised into mining and non-mining areas respectively. About 83 % of the farmers in the mining areas were affected by gold mining through channels such as land disputes, relocation of farm/residence, high cost of labour, illegal small-scale mining and dust settlement on crops. Also, about 64 % of cocoa farmers in the mining areas lost their farm lands (between 0.4 and 3.64 ha as a result of gold mining. The Tornqvist Total Factor Productivity (TFP) indices for cocoa farmers in the non-mining areas (mean TFP of 1.404) were also statistically higher than those in the mining areas (mean TFP of 0.371). The study concluded that gold mining activities adversely affect productivity of farmers in the catchment areas. The study recommends, among others, that a policy of land-for-land should be in place and effectively implemented to ensure that mining companies in order to enhance and ensure continuity of livelihoods must fully replace lands lost through mining activities

    Plasmodium falciparum isolates from southern Ghana exhibit polymorphisms in the SERCA-type PfATPase6 though sensitive to artesunate in vitro

    <p>Abstract</p> <p>Background</p> <p>In 2005, Ghana replaced chloroquine with artemisinin-based combination therapy as the first-line treatment for uncomplicated malaria. The aim of this work was to determine for the first time, polymorphisms in the putative <it>pfATPase6 </it>and <it>pftctp</it>, <it>pfmdr1</it>, <it>pfcrt </it>genes in Ghanaian isolates, particularly at a time when there is no report on artemisinin resistance in malaria parasites from Ghana. The sensitivity of parasite isolates to anti-malaria drugs were also evaluated for a possible association with polymorphisms in these genes.</p> <p>Methods</p> <p>The prevalence of point mutations in the above <it>Plasmodium falciparum </it>genes were assessed from filter-paper blood blot samples by DNA sequencing. <it>In vitro </it>drug sensitivity test was carried out on some of the blood samples from volunteers visiting hospitals/clinics in southern Ghana using a modified version of the standard WHO Mark III micro-test.</p> <p>Results</p> <p>All successfully tested parasite isolates were sensitive to artesunate; while 19.4%, 29.0% and 51.6% were resistant to quinine, amodiaquine and chloroquine respectively. The geometric mean of IC<sub>50 </sub>value for artesunate was 0.73 nM (95% CI, 0.38-1.08), amodiaquine 30.69 nM (95% CI, 14.18-47.20) and chloroquine 58.73 nM (95% CI, 38.08-79.38). Twenty point mutations were observed in <it>pfATPase6 </it>gene, with no L263E and S769N. All mutations found were low in frequency, except D639G which was observed in about half of the isolates but was not associated with artesunate response (<it>p </it>= 0.42). The <it>pftctp </it>gene is highly conserved as no mutation was observed, while CVIET which is chloroquine-resistant genotype at codon 72-76 of the <it>pfcrt </it>gene was identified in about half of the isolates; this was consistent with chloroquine IC<sub>50 </sub>values (<it>p </it>= 0.001). Mutations were present in <it>pfmdr1 </it>gene but were not associated with artemisinin response (<it>p </it>= 1.00).</p> <p>Conclusion</p> <p>The <it>pfATPase6 </it>gene is highly polymorphic with D639G appearing to be fixed in Ghanaian isolates. These may just be spontaneous mutations as all parasite isolates that were tested displayed satisfactory <it>in vitro </it>response to artesunate. However, there is no improvement in susceptibility of the parasites to chloroquine five years after its proscription.</p

    Mechanistic insights revealed by lipid profiling in monogenic insulin resistance syndromes.

    BACKGROUND: Evidence from several recent metabolomic studies suggests that increased concentrations of triacylglycerols with shorter (14-16 carbon atoms), saturated fatty acids are associated with insulin resistance and the risk of type 2 diabetes. Although causality cannot be inferred from association studies, patients in whom the primary cause of insulin resistance can be genetically defined offer unique opportunities to address this challenge. METHODS: We compared metabolite profiles in patients with congenital lipodystrophy or loss-of-function insulin resistance (INSR gene) mutations with healthy controls. RESULTS: The absence of significant differences in triacylglycerol species in the INSR group suggest that changes previously observed in epidemiological studies are not purely a consequence of insulin resistance. The presence of triacylglycerols with lower carbon numbers and high saturation in patients with lipodystrophy suggests that these metabolite changes may be associated with primary adipose tissue dysfunction. The observed pattern of triacylglycerol species is indicative of increased de novo lipogenesis in the liver. To test this we investigated the distribution of these triacylglycerols in lipoprotein fractions using size exclusion chromatography prior to mass spectrometry. This associated these triacylglycerols with very low-density lipoprotein particles, and hence release of triacylglycerols into the blood from the liver. To test further the hepatic origin of these triacylglycerols we induced de novo lipogenesis in the mouse, comparing ob/ob and wild-type mice on a chow or high fat diet, confirming that de novo lipogenesis induced an increase in relatively shorter, more saturated fatty acids. CONCLUSIONS: Overall, these studies highlight hepatic de novo lipogenesis in the pathogenesis of metabolic dyslipidaemia in states where energy intake exceeds the capacity of adipose tissue

    West Africa's moist convective environment as observed by the Atmospheric Infrared Sounder (AIRS)

    Knowledge of the seasonal positioning of the Intertropical Discontinuity (ITD) is critical to understanding seasonal moist convective processes and associated rainfall over West Africa. This study constitutes a new analysis of the seasonality of moist convection over West Africa, relative to the ITD, based on NASA's Atmospheric Infrared Sounder (AIRS) measurements from 2003 to 2018. Results show that AIRS resolves the seasonal march of the ITD, including its inherent diurnal-scale variations. AIRS captures the north–south daytime skin temperature dipole around the ITD, with greater relative temperatures to the north, especially during March–August. In the vicinity of the nighttime ITD, AIRS profiles indicate increased instability that is characteristic of nocturnal thunderstorm propagation. For seven Ghana weather stations, we show that AIRS positive moisture and equivalent potential temperature anomalies coincide with observed thunderstorm days. On these thunderstorm days, the mean latitude of the AIRS-derived ITD is displaced 3°, 0.2°, and 2° north of its DJF, MAM, and SON climatological positions, respectively, and 1.2° south in JJA. Among four common thunderstorm initiation indices considered, the K-index is determined to be most skillful.     Association of genetic variants with prostate cancer in Africa: A concise review

    Background: Prostate cancer (PCa) has one of the highest heritability of all major cancers, where the genetic contribution has been documented, and knowledge about the molecular genetics of the disease is increasing. However, the extent and aspects to which genetic variants explain PCa heritability in Africa are limited. Main body: In this review, we summarize studies that highlight how identified genetic variants explain differences in PCa incidence and presentation across ethnic groups. We also present the knowledge gaps in PCa genetics in Africa and why Africa represents an untapped potential ground for genetic studies on PCa. A significant number of genome-wide association studies, linkage, and fine-mapping analyses have been conducted globally, and that explains 30–33% of PCa heritability. The African ancestry has a significant mention in PCa incidence and presentation. To date, the candidate gene approach has replicated 23 polymorphisms including dinucleotide and trinucleotide repeats in 16 genes. CYP17-rs743572, CYP3A4-rs2740574, CYP3A5-rs776746, CYP3A43-rs501275, and haplotype blocks, containing these variants, are significantly associated with PCa among some population groups but not others. With the few existing studies, the extent of genetic diversity in Africa suggests that genetic associations of PCa to African ancestry go beyond nucleotide sequence polymorphisms, to a level of environmental adaptation, which may interpret genetic risk profiles. Also, the shreds of evidence suggest that evolutionary history contributes to the high rates of PCa relative to African ancestry, and genetic associations do not always replicate across populations. Conclusion: The genetic architecture of PCa in Africa provides important contributions to the global understanding of PCa specifically the African-ancestry hypothesis. There is a need for more prostate cancer consortiums to justify the heritable certainties of PCa among Africans, and emphasis should be placed on the genetic epidemiological model of PCa in Africa

    Does increasing treatment frequency address sub-optimal responses to ivermectin for the control and elimination of river blindness?

    Background Several African countries have adopted a biannual ivermectin distribution strategy in some foci to control and eliminate onchocerciasis. In 2010, the Ghana Health Service started biannual distribution to combat transmission hotspots and sub-optimal responses to treatment. We assessed the epidemiological impact of the first 3 years of this strategy and quantified responses to ivermectin over two consecutive rounds of treatment in 10 sentinel communities. Methods We evaluated Onchocerca volvulus microfilarial intensity and prevalence in those aged +/-20 years before the first, second and fifth (or sixth) biannual treatment rounds using skin snip data from 956 participants. We used longitudinal regression modelling to estimate rates of microfilarial repopulation of the skin in a cohort of 217 participants who were followed-up over the first two rounds of biannual treatment. Results Biannual treatment has had a positive impact, with substantial reductions in infection intensity after 4 or 5 rounds in most communities. We identified three communities—all having been previously recognised as responding sub-optimally to ivermectin—with statistically significantly high microfilarial repopulation rates. We did not find any clear association between microfilarial repopulation rate and the number of years of prior intervention, coverage, or the community level of infection. Conclusions The strategy of biannual ivermectin treatment in Ghana has reduced O. volvulus microfilarial intensity and prevalence, but sub-optimal responses to treatment remain evident in a number of previously and consistently implicated communities. Whether increasing the frequency of treatment will be sufficient to meet the World Health Organization’s 2020 elimination goals remains uncertain
