Pulmonary mycobiome of patients with suspicion of respiratory fungal infection – an exploratory study
Objective: This pilot study aimed to characterize the pulmonary mycobiome of patients with suspicion of fungal infection of the respiratory tract as well as to identify potentially pathogenic fungi colonizing/infecting their lungs.
Methods: A cohort of 10 patients was analyzed, including HIV+ patients and patients with active infection caused by Mycobacterium species. Their respiratory samples (bronchoalveolar lavage fluid/ bronchial secretions) were pre-treated with lyticase and proteinase K; DNA was extracted using the High Pure PCR Template Preparation kit following the manufacturer’s instructions. The internal transcribed spacer region 1 (ITS1) and calmodulin gene were amplified by PCR and the resulting amplicons were sequenced using the Illumina MiSeq platform with pair-end reads of 150 bp. The obtained results were analyzed using the PIPITS pipeline as described by Gweon et al. [1]. Operational taxonomic units (OTU) to which less than 0.1% of the total reads attributed were disregarded.
Results: Thirty-seven different OTU were identified from which two belonged to the Plantae kingdom, 11 had less than the 0.1% threshold of the total reads and were therefore disregarded. The remaining 24 different OTU (grouped in 17 phylotypes), were considered as part of the pulmonary mycobiome of patients. Two phyla were identified: Basidiomycota (33.3%) and Ascomycota (54.2%). Regarding the Basidiomycota phylum, reads were classified in three classes (Agaricomycetes, Tremellomycetes and Walleomycetes), while for the Ascomycota phylum four different taxonomical classes were identified: Pneumocystidomycetes, Dothideomycetes, Eurotiomycetes and Saccharomycetes, with the latter being the most frequent class. Twelve fungal genera were identified, being Candida the most frequently detected. The median number of fungal genera detected in patients’ pulmonary mycobiome was six (ranging from two up to nine). The genus Papilotrema and the potentially pathogenic genera Cryptococcus and Pneumocystis were exclusively found in the pulmonary mycobiome of HIV+ + patients. Other potentially pathogenic fungi such as Aspergillus spp., Trichosporon spp., Saccharomyces spp. and Schizophyllum spp. were also detected.
Conclusion: This pilot study illustrates how the pulmonary mycobiome is rich and highly variable in patients with fungal infections. The obtained results suggest that the described metagenomic analysis may possess a great ability to quickly and effectively detect potentially pathogenic fungi in the mycobiome of patients, making it a promising future diagnostic tool. Cinema educativo e construção social da realidade : criando identidades através da leitura e da escrita do mundo com o audiovisual
São reconhecidas as potencialidades do cinema educativo. Contudo, com o advento do computador
pessoal, do multimédia e do mito da interactividade, a sua aplicação entrou em declínio. Hoje, as
tecnologias de rede já suportam a circulação massiva de formatos audiovisuais. Produz-se, cada vez mais, um cinema doméstico (vodcasting) difundido a nível global (Youtube) com as mais variadas
intenções, gerando uma nova economia da imagem na Sociedade do Conhecimento. Por outro lado, os estudos em Novas Literacias encaram o conceito de literacia de uma forma
compreensiva, envolvendo múltiplas destrezas funcionais, referindo-se a um conjunto de actividades significantes, situadas em contextos específicos, e nos quais é fundamental uma atitude crítica relativa aos media e às condiçoes de participação e produção de conhecimento. Este ensaio problematiza o uso da audiovideografia em contexto educativo, enfatizando o cinema educativo na 1ª pessoa, o cinema que é criado pelos estudantes enquanto actividade de apreensão/construção do mundo, dos outros e de si próprios. Advoga um ressurgimento do uso do
cinema em educação quer na sua vertente mais comum de apresentação de conteúdos quer, sobretudo, como forma de construção/expressão da identidade e mediatização dos processos de aprendizagem. Apresenta e discute alguns dados produzidos e recolhidos no contexto do ensino-aprendizagem

Produzir conteúdos para a Internet ou a re-invenção da didáctica na sociedade do conhecimento?
O texto que se apresenta, resulta de duas intervenções orais feitas, a convite, em dois
eventos: o primeiro em 8 de Junho de 2004, em Aveiro, no Centro de Congressos - Seminário
PORMAT, O Desafio Digital, intervenção intitulada “A re-invenção da didáctica na sociedade do
conhecimento” -; o segundo, em 29 de abril de 2005, em Matosinhos, na Exponor –
Conferência Exponor-conteúdos para e-Learning, intervenção intitulada “A scormização do
conhecimento ou a re-invenção da didáctica…?” (tema proposto pela organização, “econteúdos
para e-Learning”).
No âmbito da educação, produzir e-conteúdos, conteúdos para a Internet, para a web,
para o e-learning e, mais recentemente, objectos de aprendizagem (as designações são
múltiplas) significa re-inventar os recursos pedagógico-didácticos. Tentaremos justificar esta
afirmação procurando dar resposta a três questões: Porquê e para quê criar e-conteúdos? O
que são e-conteúdos? Quem produz os e-conteúdos e em que condições
Can we teach without giving classes?
More than ever, in the 21st century, is essential to acquire competences to learn, to find and manage the information that is needed to solve the anticipated problems or the unpredictable ones. Teaching must be organized towards the acquisition of these competences (basic skills), providing to students activities in which their commitment, responsibility and critical exercise of freedom are essential. I briefly present a case of teaching, at the university, based on negotiation and project work. The teams have created e-portfolios (open portfolios on the internet, blogs) and only 5 of the face-to-face 48 hours were expository (lectures by the professor). Teaching mediation and transactional distance: use of facebook in a master's degree course (B-learning system)
O Ensino a Distância (EAD) continua a ser uma modalidade alternativa ao ensino presencial e nele persiste o problema da Distância Transacional. Não sendo esta distância apanágio do EAD, ela constitui um problema concreto, porém passível de minimização/resolução. O professor pode exercer uma mediação pedagógica caracterizada por um comportamento de suscitador, facilitador e orientador da aprendizagem. Para tal pode servir-se do Facebook para interagir com os estudantes, criando um ambiente de familiaridade e informalidade. Apresenta-se aqui um caso de lecionação num curso de mestrado académico em Educação que funciona em regime b-learning e no qual foi usado o Facebook, para além da plataforma institucional, como fórum de discussão e espaço de partilha de trabalhos e sensibilidades. Para recolha de dados foram usadas as técnicas da observação e análise de documentos. O curso desenrolou-se num ambiente de investigação-ação crítica no qual o professor investigou as suas práticas com a colaboração dos estudantes. Conclui-se que uma mediação plástica exercida pelo professor, servindo-se do sistema de rede social Facebook para criar um ambiente de familiaridade e informalidade, pode contribuir para superar a distância transacional no ensino superior online.Distance Learning continues to be an alternative modality to face-to-face settings and in distance learning the Transactional Distance problem persists. Not being this distance exclusive to distance learning, it is a concrete problem, but it can be minimized/resolved. The teacher can exercise a pedagogical mediation characterized by a behavior such as motivator, facilitator and guide to learning. For this one can use Facebook to interact with students, creating an atmosphere of familiarity and informality. Here we present a case of teaching in an academic master's degree course in Education that operates under a b-learning regime and in which Facebook was used, in addition to the institutional platform, as a forum for discussion and space for sharing of works and sensibilities. As techniques for data collection we used observation and document analysis. The course took place in a critical action research environment in which the teacher investigated his own practices with the collaboration of the students. ePortfolio : the state of the art in Portugal (from the 1st Meeting in Braga, July)
In the current European context, the ePortfolio emerges as an instrument that facilitates mobility,
transparence and acknowledgement of the formal and informal learning developed throughout life.
In Portugal, despite the fact that there is a significant tradition in terms of using portfolios in the
educational context, the electronic portfolio has not been object of neither public nor academic
discussion. Several questions are thus raised: What is an ePortfolio? Why build ePortfolios? What
types of ePortfolios are there? In what contexts is its use justified? How would the common citizen
really benefit from it?
In this paper, we critically review the meeting, specifying the accomplishment/evaluation indicators
and analyzing the nature of the texts presented, which, in general, reveal some difficulties both in
terms of the very conceptualization of the portfolio and in the use/knowledge of specific tools for its
implementation.
As far as the Portuguese government’s measures are concerned, there is no public initiative
concerning specifically the use of ePortfolios, despite the visible efforts of modernization that are
being made in the context of the implementation of the Lisbon Strategy and the Technological Plan.
However, some intentions of applying the ePortfolios in what concerns the processes of

Technologies Web et pratiques pédagogiques favorisant l’autonomie des étudiants
Pour que la société du savoir devienne réelle, l’université a besoin de réfléchir sur ses pratiques
d’éducation. Puis, elle doit entreprendre du changement. Ce changement passe forcément par un
usage de l’Internet en tant que dispositif didactique pour l’enseignement-apprentissage. Nous avons
créé, en 2001-2002, un site Web — base de données dynamique, interactive et multimédia — dans
le but de rendre les classes présentielles plus flexibles en ce qui concerne la gestion du temps et de
l’espace, tout en créant un environnement virtuel basé sur de « bonnes pratiques pédagogiques » qui
peuvent être traduites dans les notions de communauté, partage et construction collaborative du
savoir. Nous présentons ici le design d’une étude de cas (méthodologies et stratégies) qui a été mis

Um dispositivo de eLearning no ensino presencial universitário : avaliação do seu valor educativo
No âmbito de uma investigação de desenvolvimento na área da Tecnologia Educativa, foi criado um ambiente de eLearning (web site, base de dados dinâmica, interactiva e multimédia) cuja intenção é suportar qualquer conteúdo de ensino na universidade flexibilizando espácio-temporalmente as aulas presenciais e optimizando o processo de aprendizagem dos alunos. Este ambiente foi implementado e avaliado através de um estudo de caso (2001/02) na Universidade do Minho. Apresentamos agora uma replicação desse estudo, no contexto do Projecto “Transformar a Pedagogia na Universidade: experiências de investigação do ensino e da aprendizagem” (TPU, Universidade do Minho, Centro de Investigação em Educação). O objecto do estudo são os usos desse ambiente com a finalidade de observar/avaliar o seu potencial educativo, de forma partilhada com os colegas, tendo em vista recomendações sobre a utilização de tais recursos. O estudo decorreu no ano lectivo de 2003/2004, na disciplina de Prática Pedagógica II - Tecnologia Educativa (anual, laboratorial) do 3º ano dos cursos de Licenciatura em Ensino de Biologia e Geologia, Português e Francês, Inglês e Alemão, Português e Alemão e História e envolveu 78 alunos e oito colegas.In the context of a research-development study in the area of Educational Technology, an eLearning environment was created (website, dynamic, interactive and multimedia data-base) aimed at supporting any teaching content at the university. In this way, in-class learning can become more flexible in terms of both time and space, thus making the students’ learning process more effective. This environment was implemented and evaluated through a case study (in a natural context, 2001/2002) at the University of Minho. Now we present a replica of that study, undertaken within the Project “Transforming pedagogy at the university - experiences of research into teaching and learning” (TPU, University of Minho, Center for Research into Education). This second study focusses on the evaluation of the educational potential of the environment with the collaboration of peers, and its final aim is to produce recommendations about the use of these kind of resources. The study took place in 2003/2004, within the course of Pedagogical Practice II - Educational Technology (annual, laboratory) in the 3rd year of teaching degrees in Biology/Geology, Portuguese/French, English/German, Portuguese/German, and History.
