    Mapping topography and broad vegetation type to characterise the Boxford meadows SSSI (Unit 2)

    Understanding the dynamic relationship between hydrology and ecology in a complex wetland setting should be considered integral to the sustainable management and conservation of wetland habitats and future water resource planning. Wetland hydrology can exhibit considerable spatial complexity as a result of sub surface and surface heterogeneity. The latter of which may be determined by the relationship between spatial topography variation and broad vegetation distribution. Any study to investigate such a relationship must be at a spatial resolution sufficient to identify patterns in surface topography and vegetation type. In this study state of the art survey technology was used to collect and record for subsequent mapping the topographic and vegetation characteristics of the Boxford lowland chalk groundwater dependent terrestrial ecosystem (GDTE). The resultant survey dataset successfully unveiled distinct patterns in topography and vegetation type. The analysis of the data in a Geographical Information System (GIS) desk confirmed for the first time the presence of paleo-channels and a braided fluvial system within the meadows. In addition the combined survey method gives some indication that the type of vegetation present appears to coincide with some of the more distinctive topographical features. The results demonstrate that combining the field survey campaign alongside desk based GIS analysis is an extremely useful and versatile tool and can provide valuable information to support the decision making process for both further scientific investigation and sustainable habitat management

    Use of routinely collected health data in randomised clinical trials: comparison of trial-specific death data in the BOSS trial with NHS Digital data

    Background: A promising approach to reduce the increasing costs of clinical trials is the use of routinely collected health data as participant data. However, the quality of this data could limit its usability as trial participant data. Methods: The BOSS trial is a randomised controlled trial comparing regular endoscopies versus endoscopies at need in patients with Barrett’s oesophagus with primary endpoint death. Data on death and cancer collected every 2 years after randomisation (trial-specific data) were compared to data received annually (all patients on one date) from the routinely collected health data source National Health Service (NHS) Digital. We investigated completeness, agreement and timeliness and looked at the implications for the primary trial outcome. Completeness and agreement were assessed by evaluating the number of reported and missing cases and any disparities between reported dates. Timeliness was considered by graphing the year a death was first reported in the trial-specific data against that for NHS Digital data. Implications on the primary trial outcome, overall survival, of using one of the data sources alone were investigated using Kaplan-Meier graphs. To assess the utility of cause of death and cancer diagnoses, oesophageal cancer cases were compared. Results: NHS Digital datasets included more deaths and often reported them sooner than the trial-specific data. The number reported as being from oesophageal cancer was similar in both datasets. Due to time lag in reporting and missing cases, the event rate appeared higher using the NHS Digital data. Conclusion: NHS Digital death data is useful for calculating overall survival where trial-specific follow-up is only every 2 years from randomisation and the follow-up requires patient response. The cancer data was not a large enough sample to assess usability. We suggest that this assessment of registry data is done for more phase III RCTs and for more registry data to get a more complete picture of when RCHD would be useful in phase III RCT.     Instream and riparian implications of weed cutting in a chalk river

    Macrophyte growth is extensive in the iconic chalk streams that are concentrated in southern and eastern England. Widespread and frequent weed cutting is undertaken to maintain their key functions (e.g. flood water conveyance and maintenance of viable fisheries). In this study, a multidisciplinary approach was adopted to quantify coincident physico-chemical responses (instream and riparian) that result from weed cutting and to discuss their potential implications. Three weed cuts were monitored at a site on the River Lambourn (The CEH River Lambourn Observatory) and major instream and riparian impacts were observed. Measurements clearly demonstrated how weed cutting enhanced flood flow conveyance, reduced water levels (river and wetland), increased river velocities, and mobilised suspended sediment (with associated chemicals) and reduced the capacity for its retention within the river channel. Potential implications in relation to flood risk, water resources, downstream water quality, instream and riparian ecology, amenity value of the river, and wetland greenhouse gas emissions were considered. Provided the major influence of macrophytes on instream and riparian environments is fully understood then the manipulation of macrophytes represents an effective management tool that demonstrates the great potential of working with nature

We will review the most common malignant pathologies as well as their specific clinical presentations and management. Centers that are able to provide multidisciplinary care for pediatric patients with head and neck pathology can help achieve the best outcomes

    CTdatabase: a knowledge-base of high-throughput and curated data on cancer-testis antigens

    The potency of the immune response has still to be harnessed effectively to combat human cancers. However, the discovery of T-cell targets in melanomas and other tumors has raised the possibility that cancer vaccines can be used to induce a therapeutically effective immune response against cancer. The targets, cancer-testis (CT) antigens, are immunogenic proteins preferentially expressed in normal gametogenic tissues and different histological types of tumors. Therapeutic cancer vaccines directed against CT antigens are currently in late-stage clinical trials testing whether they can delay or prevent recurrence of lung cancer and melanoma following surgical removal of primary tumors. CT antigens constitute a large, but ill-defined, family of proteins that exhibit a remarkably restricted expression. Currently, there is a considerable amount of information about these proteins, but the data are scattered through the literature and in several bioinformatic databases. The database presented here, CTdatabase (http://www.cta.lncc.br), unifies this knowledge to facilitate both the mining of the existing deluge of data, and the identification of proteins alleged to be CT antigens, but that do not have their characteristic restricted expression pattern. CTdatabase is more than a repository of CT antigen data, since all the available information was carefully curated and annotated with most data being specifically processed for CT antigens and stored locally. Starting from a compilation of known CT antigens, CTdatabase provides basic information including gene names and aliases, RefSeq accession numbers, genomic location, known splicing variants, gene duplications and additional family members. Gene expression at the mRNA level in normal and tumor tissues has been collated from publicly available data obtained by several different technologies. Manually curated data related to mRNA and protein expression, and antigen-specific immune responses in cancer patients are also available, together with links to PubMed for relevant CT antigen articles

    Expression of testicular genes in haematological malignancies

    The gene expression of a new group of tumour antigens known as cancer/testis (CT) antigens is now well-recognized in some solid tumours. However, their expression in haematological malignancies remained unclear. In this study, we have used reverse transcription polymerase chain reaction and Southern blot analysis to examine the presence of transcripts for the three CT antigens, NY-ESO-1, SSX2 and SCP1 in haematological malignant cells. We found that transcripts for SCP1 could be detected in 10% of myeloma, 5.7% of acute myeloid leukaemia and 23% of chronic myeloid leukaemia. In contrast, NY-ESO-1 and SSX2 were not detected in any of the 107 tumour samples. © 1999 Cancer Research Campaig

    Magic-angle spinning 31 P NMR spectroscopy of condensed phosphates in parasitic protozoa: visualizing the invisible

    Abstract We report the results of a solid-state 31 P nuclear magnetic resonance (NMR) spectroscopic investigation of the acidocalcisome organelles from Trypanosoma brucei (bloodstream form), Trypanosoma cruzi and Leishmania major (insect forms). The spectra are characterized by a broad envelope of spinning sidebands having isotropic chemical shifts at V V0, 3 37 and 3 321 ppm. These resonances are assigned to orthophosphate, terminal (K K) phosphates of polyphosphates and bridging (L L) phosphates of polyphosphates, respectively. The average polyphosphate chain length is V V3.3 phosphates. Similar results were obtained with whole L. major promastigotes. 31 P NMR spectra of living L. major promastigotes recorded under conventional solution NMR conditions had spectral intensities reduced with respect to solution-state NMR spectra of acid extracts, consistent with the invisibility of the solid-state phosphates. These results show that all three parasites contain large stores of condensed phosphates which can be visualized by using magic-angle spinning NMR techniques. ß 2002 Published by Elsevier Science B.V. on behalf of the Federation of European Biochemical Societies

While it represents the work of many people connected in various ways to the university, the report does not constitute an official viewpoint of Old Dominion, its president, Brian O. Hemphill, Ph.D., the Board of Visitors, the Strome College of Business or the generous donors who support the activities of the Dragas Center for Economic Analysis and Policy. Over the past year, we have endured the depths of the COVID-19 pandemic, the introduction of vaccines and a return to a semblance of normality in Hampton Roads. Even as the economy recovers in the region, Virginia and the nation, we cannot forget that some have been left behind. Invigorating growth that raises the fortunes of all is the challenge that lies before us. This task will involve difficult discussions about how to diversify our economy in the coming years. Hampton Roads plays a significant role in national security, provides college education to thousands of Virginians and is culturally diverse. Given these realities and the fact that many residents live in one community and work in another, solutions to our regional challenges will require conversations and policies that span jurisdictional boundaries. If there is an overarching lesson to be learned from the pandemic, it is simply that we are all in this together

While it represents the work of many people connected in various ways to the university, the report does not constitute an official viewpoint of Old Dominion, its president, John R. Broderick, the Board of Visitors, the Strome College of Business or the generous donors who support the activities of the Dragas Center for Economic Analysis and Policy. Although our devotion to this work remains steadfast, our enthusiasm, admittedly, has been dampened by the COVID-19 pandemic and the toll it has taken on the region, Commonwealth and nation. In consideration of the national conversation on race and inequality, we have included additional material to shed light on how race affects economic outcomes in Hampton Roads. Hampton Roads plays a significant role in our nation’s national security, provides college education to thousands of Virginians, is working to adapt to sea level rise and is culturally diverse. There are challenges, of course, but, as the adage goes, challenge and opportunity are two sides of the same coin. To adapt, improve and overcome, we must understand where we are and where we want to go. Our work seeks to contribute to this conversation without glossing over the challenges we face
