    Applying an internal transcribed spacer as a single molecular marker to differentiate between Tetraselmis and Chlorella species

    In the realm of applied phycology, algal physiology, and biochemistry publications, the absence of proper identification and documentation of microalgae is a common concern. This poses a significant challenge for non-specialists who struggle to identify numerous eukaryotic microalgae. However, a promising solution lies in employing an appropriate DNA barcoding technique and establishing comprehensive databases of reference sequences. To address this issue, we conducted a study focusing on the molecular characterization and strain identification of Tetraselmis and Chlorella species, utilizing the internal transcribed spacer (ITS) barcode approach. By analyzing the full nuclear ITS region through the Sanger sequencing approach, we obtained ITS barcodes that were subsequently compared with other ITS sequences of various Tetraselmis and Chlorella species. To ensure the reliability of our identification procedure, we conducted a meticulous comparison of the DNA alignment, constructed a phylogenetic tree, and determined the percentage of identical nucleotides. The findings of our study reveal the significant value of the ITS genomic region as a tool for distinguishing and identifying morphologically similar chlorophyta. Moreover, our results demonstrate that both the ITS1 and ITS2 regions are capable of effectively discriminating isolates from one another; however, ITS2 is preferred due to its greater intraspecific variation. These results underscore the indispensability of employing ITS barcoding in microalgae identification, highlighting the limitations of relying solely on morphological characterization

    Comparative chemical and antimicrobial study of nine essential oils obtained from medicinal plants growing in Egypt

    Essential oils are one of interesting natural products group that are used in different aspects of life due to their various biological activities. This study investigate the antimicrobial activities of 9 herbal essential oils on survival and growth of selected pathogenic and spoilage microorganisms. Essential oils were obtained by hydrodistillation method and were analyzed using GC/MS technique. The oils were tested for their antimicrobial activity against 2 Gram +ve, Staphylococcus aureus (S. aureus) and Listeria innocua (L. innocua), 2 Gram −ve, Pseudomonas aeruginosa (P. aeruginosa) and Salmonella Typhi (S. Typhi) as well as 2 Fungi, Aspergillus niger (A. niger) and Candida albicans (C. albicans), using agar dilution method. Minimum inhibitory concentration (MIC) was determined. The antibiotic susceptibility test was performed against the test organisms by disc diffusion method. Results showed that Cinnamon oil was found effective against all the tested strains (MIC ≤ 1 μl/ml). Peppermint, lemon grass, caraway, anise, fennel and clove showed activity at (MIC ≤ 1 μl/ml) with all the tested organisms except for P. aeruginosa. Lavender oil exhibited antimicrobial activities against 4 strains (S. aureus, L. innocua, A. niger and C. albicans) with MIC (≤1 μl/ml) while geranium oil was inhibitory at (MIC ≤ 1 μl/ml) against S. aureus, S. Typhi, A. niger and C. albicans and with MIC ∼ 2 μl/ml against L. innocua. Although Gram −ve organisms had shown high resistance toward different essential oils, they were found to be susceptible to cinnamon oil even at lower concentration. Cinnamon oil is effective against drug resistant organisms. It can be suggested to use essential oils/constituents as potential natural preservatives and would be helpful in the treatment of various infections

    Allelopathic effects of Sonchus oleraceus L. on the germination and seedling growth of crop and weed species

    We assessed the allelopathic effects of the aqueous extract of Sonchus oleraceus dry shoots on the germination and seedling growth of Trifolium alexandrinum, three weed species (Brassica nigra, Chenopodium murale and Melilotus indicus) and S. oleraceus itself. We assayed four different concentrations of the aqueous extract (w v-1): 1%, 2%, 3% and 4%. To determine whether the effects of the extract were attributable to the presence of allelopathic compounds, its osmotic potential or both, we prepared concentrations of polyethylene glycol (PEG) with osmotic potentials equivalent to those of the aqueous extract. All concentrations of the plant extract completely inhibited the germination and seedling growth of C. murale. The lowest concentration of the plant extract partially inhibited germination and seedling growth of B. nigra, M. indicus and S. oleraceus, whereas the higher concentrations inhibited those parameters completely. The germination of T. alexandrinum was not affected by the aqueous extract at 1% or 2%. In general, the aqueous extracts were more effective in inhibiting seed germination and seedling growth than were the PEG solutions. Phytochemical analyses revealed that phenols and alkaloids were the most abundant compounds in S. oleraceus dry matter. Our results suggest that the aqueous extract of S. oleraceus has an allelopathic effect on some weeds, and its usefulness as a bioherbicide therefore merits further study

    Annual sowthistle (Sonchus oleraceus) has been reported to produce allelopathic effects. Two greenhouse experiments were conducted to estimate the allelopathic potential of both plant residue and root exudates of S. oleraceus on flavonoid composition and nodulation in a leguminous crop, Trifolium alexandrinum, and in two leguminous weeds, Melilotus indicus and T. resupinatum. The results of high performance liquid chromatography-mass spectrometry (HPLC-MS/MS) showed that all three legumes contained six flavonoid aglycones: apigenin, daidzein, kaempferol, luteolin, myricetin and quercetin; and seven flavonoid glycosides: daidzin, genistin, hesperidin, hyperoside, kaempferol-7-O-glucoside, naringin and rutin. In general, both plant residue and root exudates had inhibitory effects on the flavonoid composition and nodulation of the target species. However, residue of S. oleraceus caused a significant increase in both individual and total detected flavonoids in T. alexandrinum. The results suggest that the phytotoxins released from S. oleraceus may restrain the biosynthesis of flavonoids in the target species, whereas the accumulated flavonoids in T. alexandrinum are allelopathic-induced metabolites and suggest a resistance mode in this crop

    Survey to measure the quality of life of patients with tuberculosis in Alexandria, Egypt: a cross-sectional study

    Abstract Background Assessment of quality of life (QoL) in patients with tuberculosis (TB) may improve healthcare providers’ understanding of the disease burden. This study aimed to investigate the QoL of patients with TB in Alexandria, Egypt. Methods This cross-sectional study was conducted in chest clinics and main chest hospitals in Alexandria, Egypt. A structured interview questionnaire was used to collect data from participants through face-to-face interviews from November 20, 2021, until the June 30, 2022. We included all adult patients aged 18 years or above during the intensive or continuation phase of treatment. The World Health Organization (WHO) WHOQOL-BREF instrument was used to measure QoL, which includes the physical, psychological, social relationships, and environmental health domains. Using propensity score matching, a group of TB free population was recruited from the same setting and completed the questionnaire. Results A total of 180 patients participated in the study: 74.4% were males, 54.4% were married, 60.0% were 18–40 years old, 83.3% lived in urban areas, 31.7% were illiterate, 69.5% reported insufficient income, and 10.0% had multidrug-resistant TB. The TB-free population group had higher QoL scores than the TB patients’ group: (65.0 ± 17.5 vs. 42.4 ± 17.8) for the physical domain, (59.2 ± 13.6 vs. 41.9 ± 15.1) for the psychological domain, (61.8 ± 19.9 vs. 50.3 ± 20.6) for the social domain, (56.3 ± 19.3 vs. 44.5 ± 12.8) for the environment domain, (4.0(3.0–4.0) vs. 3.0(2.0–4.0)) for general health, and (4.0(3.0–4.0) vs. 2.0(2.0–3.0)) for the general QoL, P < 0.0001. Patients with TB aged 18–30 years had the highest environmental score compared with the other age groups (P = 0.021). Conclusions TB had a significant negative impact on QoL, with the physical and psychological domains being the most affected. This finding necessitates strategies to improve QoL of patients with to enhance their compliance to treatment
