    Design of an enquiry-based ‘Practical Only’ course for the teaching of basis skills in first year Biology

    First year Biology teaching at the Callaghan Campus of the University of Newcastle has undergone a significant reorganisation in 2006. The rearrangement was conducted with the aims of increasing flexible delivery, improving student learning, reducing overall teaching effort, targeting teaching effort to biology majors and standardising course delivery throughout the university campuses. Key to the reorganisation has been the separation of the practical and lecture components of first year into distinctly separate courses. The practical course runs only in semester 2 and is compulsory for students that intend to graduate with a major in biology. Students that do not intend to continue their biology studies past 1st year are not required nor expected to enrol in the course. Separation of the courses has allowed a renewed focus on basis skills including laboratory and field techniques, the scientific method and practice, report writing and personal interaction. In this paper we present the design of this practical course and explain the process and logic we have used in its construction. Skill acquisition is situated in authentic learning contexts, employing the University campus as a unifying theme. Through an enquiry- based approach, students learn how to think as scientists, posing and testing questions rather than ‘doing the experiment’. The process of building and reinforcing skills (scaffolded learning) and the use of assessment & peer interaction to facilitate the learning process is discussed

    Aquaporins and unloading of phloem-imported water in coats of developing bean seeds

    The definitive version is available at www.blackwell-synergy.comNutrients are imported into developing legume seeds by mass flow through the phloem, and reach developing embryos following secretion from their symplasmically isolated coats. To sustain homeostasis of seed coat water relations, phloem-delivered nutrients and water must exit seed coats at rates commensurate with those of import through the phloem. In this context, coats of developing French bean seeds were screened for expression of aquaporin genes resulting in cloning PvPIP1;1, PvPIP2;2 and PvPIP2;3. These genes were differentially expressed in all vegetative organs, but exhibited their strongest expression in seed coats. In seed coats, expression was localized to cells of the nutrient-unloading pathway. Transport properties of the PvPIPs were characterized by expression in Xenopus oocytes. Only PvPIP2;3 showed significant water channel activity (Pos = 150–200 ”m s−1) even when the plasma membrane intrinsic proteins (PIPs) were co-expressed in various combinations. Permeability increases to glycerol, methylamine and urea were not detected in oocytes expressing PvPIPs. Transport active aquaporins in native plasma membranes of seed coats were demonstrated by measuring rates of osmotic shrinkage of membrane vesicles in the presence and absence of mercuric chloride and silver nitrate.     Activities of the sucrose-cleaving enzymes, acid and neutral invertase and sucrose synthase, were measured in pods and seeds of developing snap bean (Phaseolus vulgaris L.) fruits, and compared with 14C-import, elongation and dry weight accumulation. During the first 10 d post-anthesis, pods elongated rapidly with pod dry weight increase lagging behind by several days. The temporal patterns of acid invertase activity and import coincided closely during the first part of pod development, consonant with a central role for this enzyme in converting imported sucrose during pod elongation and early dry weight accumulation. Later, sucrose synthase became the predominant enzyme of dry weight accumulation and was possibly associated with the development of phloem in pod walls. Sucrose synthase activity in seeds showed two peaks, corresponding to two phases of rapid import and dry weight accumulation; hence, sucrose synthase was associated with seed sink growth. Acid invertase activities in seeds were low and did not show a noticeable relationship with import or growth. All neutral invertase activities, during pod and seed development, were too low for it to have a dominant role in sucrose cleavage. Changes in activities of certain sucrose-cleaving enzymes appear to be correlated with certain sink functions, including import, storage of reserves, and biosynthetic activities. The data supports the association of specific sucrose-cleaving enzymes with the specific processes that occur in the developing pods and seeds of snap bean fruits; for example, acid invertase with pod elongation and sucrose synthase with fruit dry matter accumulation
