    Specific ligation to double-stranded RNA for analysis of cellular RNA::RNA interactions

    Double-stranded RNA (dsRNA) is formed in cells as intra- and intermolecular RNA interactions and is involved in a range of biological processes including RNA metabolism, RNA interference and translation control mediated by natural antisense RNA and microRNA. Despite this breadth of activities, few molecular tools are available to analyse dsRNA as native hybrids. We describe a two-step ligation method for enzymatic joining of dsRNA adaptors to any dsRNA molecule in its duplex form without a need for prior sequence or termini information. The method is specific for dsRNA and can ligate various adaptors to label, map or amplify dsRNA sequences. When combined with reverse transcription–polymerase chain reaction, the method is sensitive and can detect low nanomolar concentrations of dsRNA in total RNA. As examples, we mapped dsRNA/single-stranded RNA junctions within Escherichia coli hok mRNA and the human immunodeficiency virus TAR element using RNA from bacteria and mammalian cells

    A Nonabelian Yang-Mills Analogue of Classical Electromagnetic Duality

    The classic question of a nonabelian Yang-Mills analogue to electromagnetic duality is here examined in a minimalist fashion at the strictly 4-dimensional, classical field and point charge level. A generalisation of the abelian Hodge star duality is found which, though not yet known to give dual symmetry, reproduces analogues to many dual properties of the abelian theory. For example, there is a dual potential, but it is a 2-indexed tensor TμνT_{\mu\nu} of the Freedman-Townsend type. Though not itself functioning as such, TμνT_{\mu\nu} gives rise to a dual parallel transport, A~μ\tilde{A}_\mu, for the phase of the wave function of the colour magnetic charge, this last being a monopole of the Yang-Mills field but a source of the dual field. The standard colour (electric) charge itself is found to be a monopole of A~μ\tilde{A}_\mu. At the same time, the gauge symmetry is found doubled from say SU(N)SU(N) to SU(N)×SU(N)SU(N) \times SU(N). A novel feature is that all equations of motion, including the standard Yang-Mills and Wong equations, are here derived from a `universal' principle, namely the Wu-Yang (1976) criterion for monopoles, where interactions arise purely as a consequence of the topological definition of the monopole charge. We study the super-extended HI disk of M83 (∼50 kpc in radius), in particular disc kinematics, rotation and the turbulent nature of the atomic interstellar medium. We define distinct regions in the outer disk (rgal > central optical disk), including ring, southern area, and southern and northern arm. We examine HI gas surface density, velocity dispersion and non-circular motions in the outskirts, which we compare to the inner optical disk. We find an increase of velocity dispersion (σv) towards the pronounced HI ring, indicative of more turbulent HI gas. Additionally, we report over a large galactocentric radius range (until rgal ∼ 50 kpc) that σv is slightly larger than thermal (i.e. > 8 km s-1). We find that a higher star formation rate (as traced by FUV emission) is not always necessarily associated with a higher HI velocity dispersion, suggesting that radial transport could be a dominant driver for the enhanced velocity dispersion. We further find a possible branch that connects the extended HI disk to the dwarf irregular galaxy UGCA365, that deviates from the general direction of the northern arm. Lastly, we compare mass flow rate profiles (based on 2D and 3D tilted ring models) and find evidence for outflowing gas at rgal ∼ 2 kpc, inflowing gas at rgal ~ 5.5 kpc and outflowing gas at rgal ~ 14 kpc. We caution that mass flow rates are highly sensitive to the assumed kinematic disk parameters, in particular, to the inclination.Publisher PDFPeer reviewe

    A Generalized duality symmetry for nonabelian Yang-Mills fields

    It is shown that classical nonsupersymmetric Yang-Mills theory in 4 dimensions is symmetric under a generalized dual transform which reduces to the usual dual *-operation for electromagnetism. The parallel phase transport A~μ(x)\tilde{A}_\mu(x) constructed earlier for monopoles is seen to function also as a potential in giving full description of the gauge field, playing thus an entirely dual symmetric role to the usual potential Aμ(x)A_\mu(x). Sources of AA are monopoles of A~\tilde{A} and vice versa, and the Wu-Yang criterion for monopoles is found to yield as equations of motion the standard Wong and Yang-Mills equations for respectively the classical and Dirac point charge; this applies whether the charge is electric or magnetic, the two cases being related just by a dual transform.
