    Using Group Model Building to Understand Factors That Influence Childhood Obesity in an Urban Environment

    Background: Despite increased attention, conventional views of obesity are based upon individual behaviors, and children and parents living with obesity are assumed to be the primary problem solvers. Instead of focusing exclusively on individual reduction behaviors for childhood obesity, greater focus should be placed on better understanding existing community systems and their effects on obesity. The Milwaukee Childhood Obesity Prevention Project is a community-based coalition established to develop policy and environmental change strategies to impact childhood obesity in Milwaukee, Wisconsin. The coalition conducted a Group Model Building exercise to better understand root causes of childhood obesity in its community. Methods: Group Model Building is a process by which a group systematically engages in model construction to better understand the systems that are in place. It helps participants make their mental models explicit through a careful and consistent process to test assumptions. This process has 3 main components: (1) assembling a team of participants; (2) conducting a behavior-over-time graphs exercise; and (3) drawing the causal loop diagram exercise. Results: The behavior-over-time graph portion produced 61 graphs in 10 categories. The causal loop diagram yielded 5 major themes and 7 subthemes. Conclusions: Factors that influence childhood obesity are varied, and it is important to recognize that no single solution exists. The perspectives from this exercise provided a means to create a process for dialogue and commitment by stakeholders and partnerships to build capacity for change within the community

    The COVID-19 pandemic has resulted in increased need for diagnostic testing using reverse transcriptase real-time PCR (RT-PCR). An exponential increase in demand has resulted in a shortage of numerous reagents in particular those associated with the lysis buffer required to extract the viral RNA. Herein, we describe a rapid collective effort by hospital laboratory scientists, academic researchers and the biopharma industry to generate a validated lysis buffer. We have formulated a 4M Guanidinium thiocyanate (GITC)/ Triton X-100 Lysis buffer which provides comparable results with the recommended reagents. This buffer will ease the burden on hospital labs in their heroic efforts diagnose a large population of patients

    An Empiricist’s Guide to Using Ecological Theory

    A scientific understanding of the biological world arises when ideas about how nature works are formalized, tested, refined, and then tested again. Although the benefits of feedback between theoretical and empirical research are widely acknowledged by ecologists, this link is still not as strong as it could be in ecological research. This is in part because theory, particularly when expressed mathematically, can feel inaccessible to empiricists who may have little formal training in advanced math. To address this persistent barrier, we provide a general and accessible guide that covers the basic, step-by-step process of how to approach, understand, and use ecological theory in empirical work. We first give an overview of how and why mathematical theory is created, then outline four specific ways to use both mathematical and verbal theory to motivate empirical work, and finally present a practical tool kit for reading and understanding the mathematical aspects of ecological theory.We hope that empowering empiricists to embrace theory in their work will help move the field closer to a full integration of theoretical and empirical research

    Transforming Obesity Prevention for CHILDren (TOPCHILD) Collaboration: Protocol for a Systematic Review with Individual Participant Data Meta-Analysis of Behavioural Interventions for the Prevention of Early Childhood Obesity

    INTRODUCTION: Behavioural interventions in early life appear to show some effect in reducing childhood overweight and obesity. However, uncertainty remains regarding their overall effectiveness, and whether effectiveness differs among key subgroups. These evidence gaps have prompted an increase in very early childhood obesity prevention trials worldwide. Combining the individual participant data (IPD) from these trials will enhance statistical power to determine overall effectiveness and enable examination of individual and trial-level subgroups. We present a protocol for a systematic review with IPD meta-analysis to evaluate the effectiveness of obesity prevention interventions commencing antenatally or in the first year after birth, and to explore whether there are differential effects among key subgroups. METHODS AND ANALYSIS: Systematic searches of Medline, Embase, Cochrane Central Register of Controlled Trials, Cumulative Index to Nursing and Allied Health Literature (CINAHL), PsycInfo and trial registries for all ongoing and completed randomised controlled trials evaluating behavioural interventions for the prevention of early childhood obesity have been completed up to March 2021 and will be updated annually to include additional trials. Eligible trialists will be asked to share their IPD; if unavailable, aggregate data will be used where possible. An IPD meta-analysis and a nested prospective meta-analysis will be performed using methodologies recommended by the Cochrane Collaboration. The primary outcome will be body mass index z-score at age 24±6 months using WHO Growth Standards, and effect differences will be explored among prespecified individual and trial-level subgroups. Secondary outcomes include other child weight-related measures, infant feeding, dietary intake, physical activity, sedentary behaviours, sleep, parenting measures and adverse events. ETHICS AND DISSEMINATION: Approved by The University of Sydney Human Research Ethics Committee (2020/273) and Flinders University Social and Behavioural Research Ethics Committee (HREC CIA2133-1). Results will be relevant to clinicians, child health services, researchers, policy-makers and families, and will be disseminated via publications, presentations and media releases.     Unpacking the Behavioural Components and Delivery Features of Early Childhood Obesity Prevention Interventions in the TOPCHILD Collaboration: A Systematic Review and Intervention Coding Protocol

    INTRODUCTION: Little is known about how early (eg, commencing antenatally or in the first 12 months after birth) obesity prevention interventions seek to change behaviour and which components are or are not effective. This study aims to (1) characterise early obesity prevention interventions in terms of target behaviours, delivery features and behaviour change techniques (BCTs), (2) explore similarities and differences in BCTs used to target behaviours and (3) explore effectiveness of intervention components in preventing childhood obesity. METHODS AND ANALYSIS: Annual comprehensive systematic searches will be performed in Epub Ahead of Print/MEDLINE, Embase, Cochrane (CENTRAL), CINAHL, PsycINFO, as well as clinical trial registries. Eligible randomised controlled trials of behavioural interventions to prevent childhood obesity commencing antenatally or in the first year after birth will be invited to join the Transforming Obesity in CHILDren Collaboration. Standard ontologies will be used to code target behaviours, delivery features and BCTs in both published and unpublished intervention materials provided by trialists. Narrative syntheses will be performed to summarise intervention components and compare applied BCTs by types of target behaviours. Exploratory analyses will be undertaken to assess effectiveness of intervention components. ETHICS AND DISSEMINATION: The study has been approved by The University of Sydney Human Research Ethics Committee (project no. 2020/273) and Flinders University Social and Behavioural Research Ethics Committee (project no.     Electrocardiographic features of immune checkpoint inhibitor associated myocarditis.

    BACKGROUND: Myocarditis is a highly morbid complication of immune checkpoint inhibitor (ICI) use that remains inadequately characterized. The QRS duration and the QTc interval are standardized electrocardiographic measures that are prolonged in other cardiac conditions; however, there are no data on their utility in ICI myocarditis. METHODS: From an international registry, ECG parameters were compared between 140 myocarditis cases and 179 controls across multiple time points (pre-ICI, on ICI prior to myocarditis, and at the time of myocarditis). The association between ECG values and major adverse cardiac events (MACE) was also tested. RESULTS: Both the QRS duration and QTc interval were similar between cases and controls prior to myocarditis. When compared with controls on an ICI (93±19 ms) or to baseline prior to myocarditis (97±19 ms), the QRS duration prolonged with myocarditis (110±22 ms, p CONCLUSIONS: The QRS duration is increased in ICI myocarditis and is associated with increased MACE risk. Use of this widely available ECG parameter may aid in ICI myocarditis diagnosis and risk-stratification

    Biomimetic adhesive containing nanocomposite hydrogel with enhanced materials properties

    Chemically crosslinked polyacrylamide (PAAm) nanocomposite hydrogels were prepared with inorganic nano-silicate, Laponite, and dopamine methacrylamide (DMA). DMA consists of a biomimetic adhesive side chain covalently linked to a polymerizable methacrylate monomer. Copolymerizing DMA into a PAAm hydrogel strongly enhanced the interfacial interaction between the polymer network and Laponite. Nanocomposite hydrogels demonstrated reduced water content and increased materials properties that were dependent on both the Laponite and DMA contents. While increasing Laponite content alone improved materials properties moderately, these improvements were drastically enhanced when DMA is incorporated as measured by both unconfined compression testing and oscillatory rheometry. DMA-containing nanocomposite hydrogels demonstrated increased stiffness as well as excellent energy dissipation capability. Nanocomposite hydrogels with relatively low DMA and Laponite contents (2 to 3 wt% for each) demonstrated maximum compressive stress, elastic modulus, toughness, and storage and loss moduli values that were over an order of magnitude higher than control gels. DMA-containing nanocomposite hydrogels also demonstrated improved fracture resistance to compressive loading, capable of repeated compressed to 80% strain without rest for over 10 times while exhibiting compressive stress of over 1.1 MPa. The catechol side chain of DMA likely formed strong physical bonds with Laponite, which can dissipate fracture energy while minimizing permanent damage to the network architecture

    Behavioral symptoms of dementia that present management difficulties in nursing homes: staff perceptions and their concordance with informant scales

    The current study aimed to profile behaviors associated with dementia that pose management difficulties for staff and determine whether existing rating scales capture these reported behaviors. Staff in 17 nursing homes described the behavioral symptoms of 229 residents with predominantly moderate-severe dementia associated with management difficulties. Behaviors were categorized by an expert clinical panel and compared to items in four dementia behavior rating scales. Staff reported 59 discrete behavioral symptoms, with physically agitated, aggressive verbal, non-aggressive verbal, and aggressive physical behaviors most common, followed by resistance to care and inappropriate social and sexual behaviors. Results suggested that some scales omit important behaviors reported by staff for residents with particularly challenging behaviors. The current study highlights the clinical complexity faced by nursing home staff in managing residents with behavioral symptoms of dementia. [Journal of Gerontological Nursing, 43(1), 34-43.]

    A Stereoselective Hydroxylation Step of Alkaloid Biosynthesis by a Unique Cytochrome P450 in \u3ci\u3eCatharanthus roseus\u3c/i\u3e

    Plant cytochrome P450s are involved in the production of over a hundred thousand metabolites such as alkaloids, terpenoids, and phenylpropanoids. Although cytochrome P450 genes constitute one of the largest superfamilies in plants, many of the catalytic functions of the enzymes they encode remain unknown. Here, we report the identification and functional characterization of a cytochrome P450 gene in a new subfamily of CYP71, CYP71BJ1, involved in alkaloid biosynthesis. Co-expression analysis of putative cytochrome P450 genes in the Catharanthus roseus transcriptome identified candidate genes with expression profiles similar to known terpene indole alkaloid biosynthetic genes. Screening of these candidate genes by functional expression in Saccharomyces cerevisiae yielded a unique P450-dependent enzyme that stereoselectively hydroxylates the alkaloids tabersonine and lochnericine at the 19-position of the aspidosperma-type alkaloid scaffold. Tabersonine, which can be converted to either vindoline or 19-O-acetylhörhammericine, represents a branch point in alkaloid biosynthesis. The discovery of CYP71BJ1, which forms part of the pathway leading to 19-O-acetylhörhammericine, will help illuminate how this branch point is controlled in C. roseus

    New age constraints support a K/Pg boundary interval on Vega Island, Antarctica: Implications for latest Cretaceous vertebrates and paleoenvironments

    A second K/Pg boundary interval in the northern sector of the Antarctic Peninsula on Vega Island has been proposed, yet current temporal resolution of these strata prohibits direct testing of this hypothesis. To not only test for the existence of a K/Pg boundary on Vega Island but also provide increased age resolution for the associated vertebrate fauna (e.g., marine reptiles, nonavian dinosaurs, and avian dinosaurs), the Vega Island succession was intensively resampled. Stratigraphic investigation of the Cape Lamb Member of the Snow Hill Island Formation, and in particular, the overlying Sandwich Bluff Member of the López de Bertodano Formation, was conducted using biostratigraphy, strontium isotope stratigraphy, magnetostratigraphy, and detrital zircon geochronology. These data indicate a Late Campanian–early Maastrichtian age for the Cape Lamb Member and present three possible correlations to the global polarity time scale (GPTS) for the overlying Sandwich Bluff Member. The most plausible correlation, which is consistent with biostratigraphy, detrital zircon geochronology, sequence stratigraphy, and all but one of the Sr-isotope ages, correlates the base of the section to C31N and the top of the section with C29N, which indicates that the K/Pg boundary passes through the top of the unit. A second, less plausible option conflicts with the biostratigraphy and depends on a series of poorly defined magnetic reversals in the upper part of the stratigraphy that also correlates the section between C31N and C29R and again indicates an inclusive K/Pg boundary interval. The least likely correlation, which depends on favoring only a single Srisotope age at the top of the section over biostratigraphy, correlates the section between C31N and C30N and is inconsistent with an included K/Pg boundary interval. Although our preferred correlation is well supported, we failed to identify an Ir-anomaly, spherules/ impact ejecta, or other direct evidence typically used to define the precise position of a K/Pg boundary on Vega Island. This study does, however, confirm that Vegavis, from the base of the Sandwich Bluff Member, is the oldest (69.2–68.4 Ma) phylogenetically placed representative of the avian crown clade, and that marine vertebrates and nonavian dinosaurs persisted in Antarctica up to the terminal Cretaceous
