    Maternal depression and youth internalizing and externalizing symptomatology: severity and chronicity of past maternal depression and current maternal depressive symptoms

    Maternal depression is a well-documented risk factor for youth depression, and taking into account its severity and chronicity may provide important insight into the degree of risk conferred. This study explored the degree to which the severity/chronicity of maternal depression history explained variance in youth internalizing and externalizing symptoms above and beyond current maternal depressive symptoms among 171 youth (58 % male) ages 8 to 12 over a span of 3 years. Severity and chronicity of past maternal depression and current maternal depressive symptoms were examined as predictors of parent-reported youth internalizing and externalizing symptomatology, as well as youth self-reported depressive symptoms. Severity and chronicity of past maternal depression did not account for additional variance in youth internalizing and externalizing symptoms at Time 1 beyond what was accounted for by maternal depressive symptoms at Time 1. Longitudinal growth curve modeling indicated that prior severity/chronicity of maternal depression predicted levels of youth internalizing and externalizing symptoms at each time point when controlling for current maternal depressive symptoms at each time point. Chronicity of maternal depression, apart from severity, also predicted rate of change in youth externalizing symptoms over time. These findings highlight the importance of screening and assessing for current maternal depressive symptoms, as well as the nature of past depressive episodes. Possible mechanisms underlying the association between severity/chronicity of maternal depression and youth outcomes, such as residual effects from depressive history on mother–child interactions, are discussed.The current work was supported by grants from the National Institutes of Health (MH066077, PI: Martha C.     Mining whole sample mass spectrometry proteomics data for biomarkers: an overview

    In this paper we aim to provide a concise overview of designing and conducting an MS proteomics experiment in such a way as to allow statistical analysis that may lead to the discovery of novel biomarkers. We provide a summary of the various stages that make up such an experiment, highlighting the need for experimental goals to be decided upon in advance. We discuss issues in experimental design at the sample collection stage, and good practise for standardising protocols within the proteomics laboratory. We then describe approaches to the data mining stage of the experiment, including the processing steps that transform a raw mass spectrum into a useable form. We propose a permutation-based procedure for determining the significance of reported error rates. Finally, because of its general advantages in speed and cost, we suggest that MS proteomics may be a good candidate for an early primary screening approach to disease diagnosis, identifying areas of risk and making referrals for more specific tests without necessarily making a diagnosis in its own right.     Voice parameters predict sex-specific body morphology in men and women

    Studies of several mammalian species confirm that formant frequencies (vocal tract resonances) predict height and weight better than does fundamental frequency (F0, perceived as pitch) in same-sex adults due to differential anatomical constraints. However, our recent meta-analysis (Pisanski et al., 2014, Animal Behaviour, 95, 89–99) indicated that formants and F0 could explain no more than 10% and 2% of the variance in human height, respectively, controlling for sex and age. Here, we examined whether other voice parameters, many of which are affected by sex hormones, can indicate additional variance in human body size or shape, and whether these relationships differ between the sexes. Using a cross-cultural sample of 700 men and women, we examined relationships among 19 voice parameters (minimum–maximum F0, mean F0, F0 variability, formant-based vocal tract length estimates, shimmer, jitter, harmonics-to-noise ratio) and eight indices of body size or shape (height, weight, body mass index, hip, waist and chest circumferences, waist-to-hip ratio, chest-to-hip ratio). Our results confirm that formant measures explain the most variance in heights and weights of men and women, whereas shimmer, jitter and harmonics-to-noise ratio do not indicate height, weight or body mass index in either sex. In contrast, these perturbation and noise parameters, in addition to F0 range and variability, explained more variance in body shape than did formants or mean F0, particularly among men. Shimmer or jitter explained the most variance in men's hip circumferences (12%) and chest-to-hip ratios (6%), whereas harmonics-to-noise ratio and formants explained the most variance in women's waist-to-hip ratios (11%), and significantly more than in men's waist-to-hip ratios. Our study represents the most comprehensive analysis of vocal indicators of human body size to date and offers a foundation for future research examining the hormonal mechanisms of voice production in humans and perceptual playback experiments

    The Físchlár-News-Stories system: personalised access to an archive of TV news

    The “Físchlár” systems are a family of tools for capturing, analysis, indexing, browsing, searching and summarisation of digital video information. Físchlár-News-Stories, described in this paper, is one of those systems, and provides access to a growing archive of broadcast TV news. Físchlár-News-Stories has several notable features including the fact that it automatically records TV news and segments a broadcast news program into stories, eliminating advertisements and credits at the start/end of the broadcast. Físchlár-News-Stories supports access to individual stories via calendar lookup, text search through closed captions, automatically-generated links between related stories, and personalised access using a personalisation and recommender system based on collaborative filtering. Access to individual news stories is supported either by browsing keyframes with synchronised closed captions, or by playback of the recorded video. One strength of the Físchlár-News-Stories system is that it is actually used, in practice, daily, to access news. Several aspects of the Físchlár systems have been published before, bit in this paper we give a summary of the Físchlár-News-Stories system in operation by following a scenario in which it is used and also outlining how the underlying system realises the functions it offers

    Suicide and suicide risk

    Although recent years have seen large decreases in the overall global rate of suicide fatalities, this trend is not reflected everywhere. Suicide and suicidal behaviour continue to present key challenges for public policy and health services, with increasing suicide deaths in some countries such as the USA. The development of suicide risk is complex, involving contributions from biological (including genetics), psychological (such as certain personality traits), clinical (such as comorbid psychiatric illness), social and environmental factors. The involvement of multiple risk factors in conveying risk of suicide means that determining an individual’s risk of suicide is challenging. Improving risk assessment, for example, by using computer testing and genetic screening, is an area of ongoing research. Prevention is key to reduce the number of suicide deaths and prevention efforts include universal, selective and indicated interventions, although these interventions are often delivered in combination. These interventions, combined with psychological (such as cognitive behavioural therapy, caring contacts and safety planning) and pharmacological treatments (for example, clozapine and ketamine) along with coordinated social and public health initiatives, should continue to improve the management of individuals who are suicidal and decrease suicide-associated morbidity

    Effects of zinc supplementation on cognitive function in healthy middle-aged and older adults: the ZENITH study

    A randomised double-blind placebo-controlled design was employed to investigate the effects of Zn supplementation on cognitive function in 387 healthy adults aged 55–87 years. Several measures of visual memory, working memory, attention and reaction time were obtained using the Cambridge Automated Neuropsychological Test Battery at baseline and then after 3 and 6 months of 0 (placebo), 15 or 30 mg Zn/d. Younger adults (70 years), and performance improved with practice on some measures. For two out of eight dependent variables, there were significant interactions indicating a beneficial effect (at 3 months only) of both 15 and 30 mg/d on one measure of spatial working memory and a detrimental effect of 15 mg/d on one measure of attention. Further work is required to establish whether these findings generalise to older adults in poorer mental and physical health and with less adequate Zn intake and status than the present sample

    Dysregulated T cell expression of TIM3 in multiple sclerosis

    T cell immunoglobulin- and mucin domain–containing molecule (TIM)3 is a T helper cell (Th)1–associated cell surface molecule that regulates Th1 responses and promotes tolerance in mice, but its expression and function in human T cells is unknown. We generated 104 T cell clones from the cerebrospinal fluid (CSF) of six patients with multiple sclerosis (MS) (n = 72) and four control subjects (n = 32) and assessed their cytokine profiles and expression levels of TIM3 and related molecules. MS CSF clones secreted higher amounts of interferon (IFN)-γ than did those from control subjects, but paradoxically expressed lower levels of TIM3 and T-bet. Interleukin 12–mediated polarization of CSF clones induced substantially higher amounts of IFN-γ secretion but lower levels of TIM3 in MS clones relative to control clones, demonstrating that TIM3 expression is dysregulated in MS CSF clones. Reduced levels of TIM3 on MS CSF clones correlated with resistance to tolerance induced by costimulatory blockade. Finally, reduction of TIM3 on ex vivo CD4+ T cells using small interfering (si)RNA enhanced proliferation and IFN-γ secretion, directly demonstrating that TIM3 expression on human T cells regulates proliferation and IFN-γ secretion. Failure to up-regulate T cell expression of TIM3 in inflammatory sites may represent a novel, intrinsic defect that contributes to the pathogenesis of MS and other human autoimmune diseases

    In vitro studies on CNGRC-CPG2 fusion proteins for ligand-directed enzyme prodrug therapy for targeted cancer therapy

    The sequence asparagine-glycine arginine (NGR), flanked by Cysteine (Cys) residues so as to form a disulfide-bridge (CNGRC), has previously been found to target and bind specifically to aminopeptidase N (APN), which is highly expressed on the surface of tumor cells. The goal of this study was to develop and evaluate the potential of fusion proteins carrying the CNGRC sequence linked to the enzyme carboxypeptidase G2 (CPG2) for targeted cancer therapy. We refer to this strategy as ligand-directed enzyme prodrug therapy (LDEPT). We constructed two forms of the CNGRC-CPG2 fusions, containing one or two copies of the cyclic NGR motif and designated CNGRC-CPG2 (X-CPG2) and CNGRC-CPG2-CNGRC (X-CPG2-X), respectively. binding assays of the purified constructs showed that both X-CPG2 and X-CPG2-X bound with high affinity to cancer cells expressing high levels of APN, compared to their binding to cells expressing low levels of APN. Further studies of the constructs to assess the therapeutic potential of LDEPT were carried out using cells expressing high and low levels of APN. Using methotrexate, it was demonstrated that cancer cell survival was significantly higher in the presence of the fusion proteins, due to the hydrolysis of this cytotoxic drug by CPG2. Conversely, when the prodrug ZD2767P was used, cancer cell killing was higher in the presence of the fused CPG2 constructs than in their absence, which is consistent with CPG2-mediated release of the cytotoxic drug from the prodrug. Furthermore, the doubly-fused CPG2 construct (X-CPG2-X) was significantly more effective than the singly-fused construct (X-CPG2)
