    Mechanical thrombectomy in acute stroke – Five years of experience in Poland

    Objectives Mechanical thrombectomy (MT) is not reimbursed by the Polish public health system. We present a description of 5 years of experience with MT in acute stroke in Comprehensive Stroke Centers (CSCs) in Poland. Methods and results We retrospectively analyzed the results of a structured questionnaire from 23 out of 25 identified CSCs and 22 data sets that include 61 clinical, radiological and outcome measures. Results Most of the CSCs (74%) were founded at University Hospitals and most (65.2%) work round the clock. In 78.3% of them, the working teams are composed of neurologists and neuro-radiologists. All CSCs perform CT and angio-CT before MT. In total 586 patients were subjected to MT and data from 531 of them were analyzed. Mean time laps from stroke onset to groin puncture was 250±99min. 90.3% of the studied patients had MT within 6h from stroke onset; 59.3% of them were treated with IV rt-PA prior to MT; 15.1% had IA rt-PA during MT and 4.7% – emergent stenting of a large vessel. M1 of MCA was occluded in 47.8% of cases. The Solitaire device was used in 53% of cases. Successful recanalization (TICI2b–TICI3) was achieved in 64.6% of cases and 53.4% of patients did not experience hemorrhagic transformation. Clinical improvement on discharge was noticed in 53.7% of cases, futile recanalization – in 30.7%, mRS of 0–2 – in 31.4% and mRS of 6 in 22% of cases. Conclusion Our results can help harmonize standards for MT in Poland according to international guidelines

    Mimetics of manganese catalase as synthetic antioxidants

    The coordination chemistry of manganese complexes with heteroaromatic ligands have received considerable attention in modern inorganic chemistry due to its wide range of applications. One of them is related with its potential biological role associated with prevention of oxidative stress injuries. Manganese complexes are one of the most versatile and interesting synthetic systems that can act as artificial mimics of manganese-containing metalloenzymes such as manganese catalase (Mn-CAT). Due to the potential use as catalytic scavengers of H2O2, numerous and diverse, very structurally interesting Mn compounds exhibiting CAT-like activity have been reported so far. In this short review, the relevant features of both, structural, as well as, functional mimics of manganese catalase and their CAT-like activity are presented and some insights into the role of the bridging ligands, endogenous bases, and first- and second-sphere effects on the catalysis are discussed

    Synthesis and structure of novel copper(II) complexes with N,O- or N,N-donors as radical scavengers and a functional model of the active sites in metalloenzymes

    To evaluate the antioxidant activity of potential synthetic enzyme mimetics, we prepared new five copper(II) complexes via a self-assembly method and named them [Cu(2-(HOCH(2))py)(3)](ClO(4))(2) (1), [Cu(2-(HOCH(2))py)(2)(H(2)O)(2)]SiF(6) (2), [Cu(2)(2-(HOCH(2)CH(2))py)(2)(2-(OCH(2)CH(2))py)(2)](ClO(4))(2) (3), [Cu(pyBIm)(3)](BF(4))(2)·1.5H(2)O (4) and [Cu(py(2)C(OH)(2))(2)](ClO(4))(2) (5). The synthetic protocol involved N,O- or N,N-donors: 2-(hydroxymethyl)pyridine (2-(HOCH(2))py), 2-(hydroxyethyl)pyridine (2-(HOCH(2)CH(2))py), 2-(2-pyridyl)benzimidazole (pyBIm), di(2-pyridyl)ketone (py(2)CO). The obtained Cu(II) complexes were fully characterised by elemental analysis, FTIR, EPR, UV-Vis, single-crystal X-ray diffraction and Hirshfeld surface analysis. Crystallographic and spectroscopic analyses confirmed chromophores of both monomeric ({CuN(3)O(3)} (1), {CuN(2)O(4)} (2), {CuN(6)} (4), {CuN(4)O(2)} (5)) and dimeric complex ({CuN(2)O(3)} (3)). Most of the obtained species possessed a distorted octahedral environment, except dimer 3, which consisted of two copper centres with square pyramidal geometries. The water-soluble compounds (1, 3 and 5) were selected for biological testing. The results of the study revealed that complex 1 in solutions displayed better radical scavenging activity than complexes 3, 5 and free ligands. Therefore, complex 1 has been selected for further studies to test its activity as an enzyme mimetic. The chosen compound was tested on the erythrocyte lysate of two groups of patients after undergoing chemotherapy and chemoradiotherapy. The effect of the tested compound (1) on enzyme activity levels (TAS, SOD and CAT) suggests that the selected complex can be treated as a functional mimetic of the enzymes
