    Immunological detection of D-β-aspartate-containing protein in lens-derived cell lines

    Alpha-crystallin is the major protein of the mammalian lens and its average molecular weight is approximately 800 kDa. It is composed of two kinds of structurally and functionally related polypeptides, αA-and αB-crystallin subunits, each with a molecular weight of 20 kDa Recently, we prepared a polyclonal antibody against peptide Gly-Leu-D-β-Asp-Ala-Thr-Gly-Leu-D-β-Asp-Ala-ThrGly-Leu-D-β-Asp-Ala-Thr (anti-peptide 3R antibody) that corresponded to three repeats of positions 149-153 in human αA-crystallin [11]. This antibody cross-reacted specifically with D-β-Asp-151-containing αA-crystallin. Because formation of D-Asp is accompanied by isomerization to form the β-Asp (isoaspartate) residue, three isomers of Asp residues, L-β-Asp, D-α-Asp and D-β-Asp isomers, are formed in the protein Cell culture systems are used widely for the analysis of cellular functions related to particular organ systems. For lens research, it is of particular interest to find conditions that reflect the situation within this organ. In order to establish whether the D-β-Asp-containing protein is present in cultured lens cells, we cultured two cell lines, αTN4-1 and N/N1003A, which are commonly used in lens research Conclusions: The results indicate that the N/N1003A cell line expressed a 50 kDa D-β-Asp-containing protein, which may share a common amino acid sequence with αA-and αB-crystallin

    Elucidation of the mechanism of subunit exchange in αB crystallin oligomers

    AlphaB crystallin (αB-crystallin) is a key protein for maintaining the long-term transparency of the eye lens. In the eye lens, αB-crystallin is a “dynamical” oligomer regulated by subunit exchange between the oligomers. To elucidate the unsettled mechanism of subunit exchange in αB-crystallin oligomers, the study was carried out at two different protein concentrations, 28.5 mg/mL (dense sample) and 0.45 mg/mL (dilute sample), through inverse contrast matching small-angle neutron scattering. Interestingly, the exchange rate of the dense sample was the same as that of the dilute sample. From analytical ultracentrifuge measurements, the coexistence of small molecular weight components and oligomers was detected, regardless of the protein concentration. The model proposed that subunit exchange could proceed through the assistance of monomers and other small oligomers; the key mechanism is attaching/detaching monomers and other small oligomers to/from oligomers. Moreover, this model successfully reproduced the experimental results for both dense and dilute solutions. It is concluded that the monomer and other small oligomers attaching/detaching mainly regulates the subunit exchange in αB-crystallin oligomer

    保健師が実施した職域における健康診断の事後相談への評価

    某JAグループの職員4、671名を対象に健康診断を行い、保健師が巡回による事後相談を創意工夫しながら実施している。今回、保健師による事後相談を評価することを目的として、事後相談の対象となった566名(10.5%)に無記名・自記式の調査を行った。その内340名から回答があり(回収率60.1%)、今回の研究対象として分析した。その結果、「個人結果通知票」により"自分の身体の状態がわかる"92.7%、"生活習慣の改善についても認識している"95.7%、であることが確認できた。また、事後相談の場における「保健指導」の内容についても"適切"65.0%、"ほぼ適切"16.2%と良い評価を得た。しかし、生活習慣改善の必要性は理解していても行動変容につながっているとは言えなかった。また、20歳代の対象者は自らの健康課題を十分理解できているとは言えなかった。今後は、行動変容が可能となる職場環境の確保への働きかけ、および、若い世代への働きかけの工夫が重要な課題である

    Low-grade B-cell lymphoma presenting primarily in the bone marrow

    Cases of low-grade B-cell lymphoma presenting primarily in the bone marrow are rare, and its clinicopathology remains unclear. We retrospectively examined patients with low-grade B-cell lymphoma presenting primarily in the bone marrow. Fourteen patients met the inclusion criteria, including 5 with lymphoplasmacytic lymphoma (LPL), 3 with chronic lymphocytic leukemia/small lymphocytic lymphoma, 2 with follicular lymphoma (FL), and 4 with low-grade B-cell lymphoma not otherwise specified (LGBCL-NOS). The median age was 69.5 years (range, 42-89 years), and a slight male predominance was noted (9 men and 5 women, 1.8: 1). Immunohistochemically, all cases were positive for CD20. One case was positive for CD138. Both cases of FL were positive for CD10 and B-cell lymphoma 2 (BCL-2), and immunoglobulin heavy locus (IgH)/B-cell lymphoma 2 rearrangement was observed by fluorescence in situ hybridization. The myeloid differentiation primary response gene (88) leucine to proline mutation was observed in 3 of 5 LPL, 1 of 2 FL, and 2 of 4 LGBCL-NOS patients. Paraproteinemia was observed in 10 patients; IgM and IgG paraproteinemia were observed in 6 and 3 patients, respectively. In this patient series, 3 patients had died at a median follow-up of 36.5 months; the cause of death of 1 LPL patient was malignant lymphoma itself. Thus, low-grade B-cell lymphoma presenting primarily in the bone marrow has various subtypes, and approximately one-third of the patients had LGBCL-NOS. The immunophenotypic features and myeloid differentiation primary response gene (88) leucine to proline mutation data of LGBCL-NOS suggested that some cases present with characteristics similar to those of LPL or marginal zone lymphoma

    G12/13 and Gq mediate S1P2-induced inhibition of Rac and migration in vascular smooth muscle in a manner dependent on Rho but not Rho kinase

    金沢大学医薬保健研究域医学系Aims: The lysophospholipid mediator sphingosine-1-phosphate (S1P) activates G protein-coupled receptors (GPCRs) to induce potent inhibition of platelet-derived growth factor (PDGF)-induced Rac activation and, thereby, chemotaxis in rat vascular smooth muscle cells (VSMCs). We explored the heterotrimeric G protein and the downstream mechanism that mediated S1P inhibition of Rac and cell migration in VSMCs. Methods and results: S1P inhibition of PDGF-induced cell migration and Rac activation in VSMCs was abolished by the selective S1P2 receptor antagonist JTE-013. The C-terminal peptides of Gα subunits (Gα-CTs) act as specific inhibitors of respective G protein-GPCR coupling. Adenovirus-mediated expression of Gα12-CT, Gα13-CT, and Gα q-CT, but not that of Gαs-CT or LacZ or pertussis toxin treatment, abrogated S1P inhibition of PDGF-induced Rac activation and migration, indicating that both G12/13 and Gq classes are necessary for the S1P inhibition. The expression of Gαq-CT as well as Gα12-CT and Gα13-CT also abolished S1P-induced Rho stimulation. C3 toxin, but not a Rho kinase inhibitor or a dominant negative form of Rho kinase, abolished S1P inhibition of PDGF-induced Rac activation and cell migration. The angiotensin II receptor AT1, which robustly couples to Gq, did not mediate either Rho activation or inhibition of PDGF-induced Rac activation or migration, suggesting that activation of Gq alone was not sufficient for Rho activation and resultant Rac inhibition. However, the AT1 receptor fused to Gα12 was able to induce not only Rho stimulation but also inhibition of PDGF-induced Rac activation and migration. Phospholipase C inhibition did not affect S1P-induced Rho activation, and protein kinase C activation by a phorbol ester did not mimic S1P action, suggesting that S1P inhibition of migration or Rac was not dependent on the phospholipase C pathway. Conclusion: These observations together suggest that S1P2 mediates inhibition of Rac and migration through the coordinated action of G 12/13 and Gq for Rho activation in VSMCs. © The Author 2008.
