    CAMRI Loss: Improving Recall of a Specific Class without Sacrificing Accuracy

    In real-world applications of multi-class classification models, misclassification in an important class (e.g., stop sign) can be significantly more harmful than in other classes (e.g., speed limit). In this paper, we propose a loss function that can improve the recall of an important class while maintaining the same level of accuracy as the case using cross-entropy loss. For our purpose, we need to make the separation of the important class better than the other classes. However, existing methods that give a class-sensitive penalty for cross-entropy loss do not improve the separation. On the other hand, the method that gives a margin to the angle between the feature vectors and the weight vectors of the last fully connected layer corresponding to each feature can improve the separation. Therefore, we propose a loss function that can improve the separation of the important class by setting the margin only for the important class, called Class-sensitive Additive Angular Margin Loss (CAMRI Loss). CAMRI loss is expected to reduce the variance of angles between features and weights of the important class relative to other classes due to the margin around the important class in the feature space by adding a penalty to the angle. In addition, concentrating the penalty only on the important classes hardly sacrifices the separation of the other classes. Experiments on CIFAR-10, GTSRB, and AwA2 showed that the proposed method could improve up to 9% recall improvement on cross-entropy loss without sacrificing accuracy.Comment: 2022 International Joint Conference on Neural Networks (IJCNN 2022

    Pituitary Volume and Socio-Cognitive Functions in Individuals at Risk of Psychosis and Patients With Schizophrenia

    Objectives: Increased pituitary volume, which probably reflects hypothalamic-pituitary-adrenal (HPA) hyperactivity, has been reported in patients with schizophrenia and individuals at risk of psychosis. On the basis of potential role of abnormal HPA axis function on cognitive impairments in psychosis, we aimed to examine possible relations between the pituitary volume and socio-cognitive impairments in these subjects.Methods: This magnetic resonance imaging study examined the pituitary gland volume in 38 subjects with at-risk mental state (ARMS) [of whom 4 (10.5%) exhibited the transition to schizophrenia], 63 patients with schizophrenia, and 61 healthy controls. Social and cognitive functions of the ARMS and schizophrenia groups were assessed using the Brief Assessment of Cognition in Schizophrenia (BACS), the Schizophrenia Cognition Rating Scale (SCoRS), and the Social and Occupational Functioning Assessment Scale (SOFAS).Results: Both the ARMS and schizophrenia groups had a significantly larger pituitary volume compared to controls. In the schizophrenia group, the pituitary volume was negatively associated with the BACS working memory score. No association was found between the pituitary volume and clinical variables (medication, symptom severity) in either clinical group.Conclusion: Our findings support the notion of common HPA hyperactivity in the ARMS and schizophrenia groups, but abnormal HPA axis function may contribute differently to cognitive deficits according to the illness stages of schizophrenia

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

    Background Elsberg syndrome (ES) is an acute‐onset lumbosacral radiculitis with myelitis caused by a herpes virus infection. Case Presentation We present a case of a 77‐year‐old woman who was admitted with urinary retention prior to genital rash. The patient was diagnosed with ES and treated with intravenous acyclovir 250 mg every 8 h for 1 week. Conclusion Physicians should consider ES in patients with voiding dysfunction, as preceding neurological symptoms may lead to a misdiagnosis. Considering the adverse effects of the antiviral drug, its dosage should be according to the causative virus of the ES as well as the patient's age and medical history

    Reentrant structural and optical properties of organic-inorganic hybrid metal cluster compound ((n-C4H9)(4)N)(2)[(Mo6Bri8Br6a)]

    International audienceWe report the reentrant phase transition of the organic-inorganic hybrid metal cluster (MC) compound (TBA)(2)[(Mo6Br8Br6a)-Br-i] (TBA = ((n-C4H9)(4)N)). Structural studies revealed that (TBA)(2)[(Mo6Br8Br6a)-Br-i] was crystallized in the conventional monoclinic phase with space group P2(1)/n. When the crystal was cooled down to 143 K, we observed a structural phase transition from the conventional phase to an unknown phase with packing structure similar to the conventional one; however, the unit cell was extended to a double size. A reentrant transition to the conventional structure was induced when the crystal was further cooled to 113 K. These reentrant phase transitions occurred without any symmetry changes caused by slight motions of the MCs and TBA(+) ions. Luminescence studies revealed that the phase transition poorly affected the emission spectra; however, the newly found phase exhibited 1.7 times longer emission lifetime than the conventional phase. These results indicate that MC-based compounds are promising and exhibit phase transitions and crystallographic polymorphisms
