Crotoxin from <i>Crotalus durissus terrificus</i> Is Able to Down-Modulate the Acute Intestinal Inflammation in Mice
<div><p>Inflammatory bowel diseases (IBD) is the result of dysregulation of mucosal innate and adaptive immune responses. Factors such as genetic, microbial and environmental are involved in the development of these disorders. Accordingly, animal models that mimic human diseases are tools for the understanding the immunological processes of the IBD as well as to evaluate new therapeutic strategies. Crotoxin (CTX) is the main component of <i>Crotalus durissus terrificus</i> snake venom and has an immunomodulatory effect. Thus, we aimed to evaluate the modulatory effect of CTX in a murine model of colitis induced by 2,4,6- trinitrobenzene sulfonic acid (TNBS). The CTX was administered intraperitoneally 18 hours after the TNBS intrarectal instillation in BALB/c mice. The CTX administration resulted in decreased weight loss, disease activity index (DAI), macroscopic tissue damage, histopathological score and myeloperoxidase (MPO) activity analyzed after 4 days of acute TNBS colitis. Furthermore, the levels of TNF-α, IL-1β and IL-6 were lower in colon tissue homogenates of TNBS-mice that received the CTX when compared with untreated TNBS mice. The analysis of distinct cell populations obtained from the intestinal lamina propria showed that CTX reduced the number of group 3 innate lymphoid cells (ILC3) and Th17 population; CTX decreased IL-17 secretion but did not alter the frequency of CD4<sup>+</sup>Tbet<sup>+</sup> T cells induced by TNBS instillation in mice. In contrast, increased CD4<sup>+</sup>FoxP3<sup>+</sup> cell population as well as secretion of TGF-β, prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) and lipoxin A<sub>4</sub> (LXA<sub>4</sub>) was observed in TNBS-colitis mice treated with CTX compared with untreated TNBS-colitis mice. In conclusion, the CTX is able to modulate the intestinal acute inflammatory response induced by TNBS, resulting in the improvement of clinical status of the mice. This effect of CTX is complex and involves the suppression of the pro-inflammatory environment elicited by intrarectal instillation of TNBS due to the induction of a local anti-inflammatory profile in mice.</p></div

<p>Cell suspensions were prepared from lamina propria of distinct experimental groups after 4 days of TNBS-induced colitis for the analysis of CD4<sup>+</sup>FoxP3<sup>+</sup> cells by flow cytometry. The samples were prepared from a pool of cells from 4–5 animals/group performed in duplicate. Airway inflammation induced by <i>B.</i><i>tropicalis</i> allergens plus eLPS presented a TH1/TH17 pattern of cytokine production.
<p>C57BL/6 (WT) mice were sensitized twice with Bt or Bt containing eLPS co-adsorbed to alum and challenged twice with Bt. Experiments were performed 24 h after the last Bt challenge. Control group consisted of non-manipulated animals. Cytokine IL-4 (<b>A</b>), IL-5 (<b>B</b>), IL-13 (<b>C</b>), IFN-γ (<b>D</b>), IL-17 (<b>E</b>), and VEGF (<b>F</b>) production BAL fluid were measure by ELISA. *Significant difference (<i>p</i><0.05) when compared with control group. <sup>#</sup>Significant difference (<i>p</i><0.05) when compared with Bt group.</p
Development of airway inflammation in the presence of eLPS involves other components than just LPS.
<p>C57BL/6 (WT), MyD88<sup>−/−</sup>, CD14<sup>−/−</sup>, TLR4<sup>−/−</sup>, or TLR2<sup>−/−</sup> mice were sensitized twice with Bt or Bt containing eLPS co-adsorbed to alum and challenged twice with Bt. Control group consisted of non-manipulated animals. The experiments were performed 24 h after the last Bt challenge. Eosinophils (<b>A</b>) and neutrophils (<b>B</b>) in BAL fluid. Results are expressed as mean ±SEM for groups of five mice and are representative of two experiments. *Significant difference (<i>p</i><0.05) when compared with Bt group.</p
Folic Acid-induced chronic kidney injury.
<p>Mice were submitted to folic acid-induced kidney injury (FA) or sodium bicarbonate (Bic) vehicle treatment and after 24 h, received adipose tissue-derived stem cells (AdSC). Kidney tissue and serum samples were extracted 4 weeks after FA for kidney function and protein expression analyses; control (Ctr) mice received no treatment, (Bic) mice received vehicle only (sodium bicarbonate, 0.15 M). <b>(A)</b> Urea biochemistry essays showed no significant difference of long term renal functional between groups (n = 2–7 for each group) <b>(B,C)</b> Kidney sections were viewed under both bright-field (a-e) and polarized light (f-j) microscopy. AdSC-treated mice kidneys displayed reduced deposit of tissue collagen in comparison to FA mice (n = 3–5 for each group) (original magnification: 20x). <b>(D)</b> Bioplex essays show stem cell-treated mice develop ameliorated chronic kidney inflammation (n = 3 for each group). FA-treated mice expressed higher levels of interferon-gamma (IFN-<b>γ</b>) vs. control mice, while in AdSC-treated mice, IFN-<b>γ</b> and eotaxin expression was shown to be reduced in kidney tissue vs. the latter. A tendency towards lower interleukine-2 (IL-2) and MIP-1<b>α</b> expression was also observed for these animals (n = 2–6 for each group).
