Analyse du contrôle de l'expression du MHC classe II par des enhancers distaux régulés par RFX/CIITA
La présentation d'antigènes par les molécules MHC II (complexe majeur d'histocompatibilité classe II) aux lymphocytes T CD4+ joue un rôle prépondérant dans la réponse immune spécifique. L'expression des gènes des molécules du MHC II est contrôlée principalement au niveau de l'initiation de la transcription par une région régulatrice hautement conservée appelée boîtes S, X1, X2 et Y située en amont du site d'initiation de la transcription. Chez certains malades souffrant du syndrome des lymphocytes dénudés, des mutations dans des protéines spécifiques se liant à ces boîtes empêchent le MHC II de s'exprimer normalement. Ce travail décrit l'analyse de séquences d'ADN architecturalement proches de ces boîtes, à l'intérieur de la région MHC II, et étudie leur fonction via des expériences d'immunoprécipitation et de gène reporteur, permettant d'avancer des hypothèses sur le rôle de ces nouvelles régions identifiées comme régulateur à grande distance de plusieurs gènes des molécules du MHC II
Potential use of microarray patches for vaccine delivery in low- and middle- income countries
Microarray patches (MAPs), also referred to as microneedle patches, are a novel methodology that have the potential to overcome barriers to vaccine delivery in low- and middle-income countries (LMICs), and transform the way that vaccines are delivered within immunization programs. The World Health Organization’s Initiative for Vaccine Research and its partners are working to understand how MAPs could ease vaccine delivery and increase equitable access to vaccines in LMICs. Global stakeholders have been engaged to evaluate technical, economic, and programmatic challenges; to validate assumptions where possible; and to propose areas of focus to facilitate future vaccine-MAP product development. Investigating outbreaks of initially unknown aetiology in complex settings: findings and recommendations from 10 case studies.
BACKGROUND: Outbreaks of unknown aetiology in complex settings pose challenges and there is little information about investigation methods. We reviewed investigations into such outbreaks to identify methods favouring or impeding identification of the cause. METHODS: We used two approaches: reviewing scientific literature and soliciting key informants. Case studies were developed through interviews with people involved and triangulated with documents available from the time of the investigation. RESULTS: Ten outbreaks in African or Asian countries within the period 2007-2017 were selected. The cause was identified in seven, of which two had an unclear mode of transmission, and in three, neither origin nor transmission mode was identified. Four events were caused by infectious agents and three by chemical poisoning. Despite differences in the outbreaks, similar obstacles were noted: incomplete or delayed description of patients, comorbidities confounding clinical pictures and case definitions wrongly attributed. Repeated rounds of data collection and laboratory investigations were common and there was limited capacity to ship samples. DISCUSSION: It was not possible to define activities that led to prompt identification of the cause in the case studies selected. Based on the observations, we conclude that basing case definitions on precise medical observations, implementing initial comprehensive data collection, including environmental, social and behavioural information; and involving local informants could save precious time and hasten implementation of control measures

Méthodologie d’aide à la conception et à l’évaluation de systèmes de production multiperformants : application à l’élevage de bovins laitiers et à la production de poulets de chair
Following the bidding by the General Commission for Future Strategy of the French government, INRA has analyzed the different evolutions possible in French practices and agriculture systems towards a more sustainable production that manages to combine productive, economic, environmental and social performances. A method to help design and evaluating the systems was thus developed. This method is based on over 200 basic agricultural practices whose impact was qualified on 35 performance indicators as well as on a compatibility index between these elementary practices. Two studies based on different uses of this method are presented: i) the step by step conception of a production system by progressively incorporating elementary practices from one entry practice with major effect, to increase food autonomy in dairy cattle production by valorizing grasslands; ii) evaluating the system performances a posteriori by transcribing the action levers and progress margins identified by an expert panel into elementary practices in order to improve competition of the broiler industry through the development of two very different but complementary production systems. The tendencies of the simulated or rebuilt performance evolution of these production systems are discussed. When situations present an ambivalent impact of a practice on one performance, using the detailed description of these practices often allows reducing the uncertainty by considering the situations associated with each type of impact.Suite à un appel d’offre du Commissariat Général à la Stratégie et à la Prospective du gouvernement français, l’INRA a analysé les possibilités d’évolution des pratiques et des systèmes agricoles français vers des systèmes de production plus durables conciliant performances productives, économiques, environnementales et sociales. Une méthodologie d’aide à la conception et à l’évaluation de systèmes a été développée à cette occasion. Elle s’appuie sur un répertoire de plus de 200 pratiques agricoles élémentaires dont l’impact a été qualifié sur 35 indicateurs de performances, ainsi que sur une table de compatibilité entre pratiques élémentaires. Deux études de cas fondées sur des modalités différentes d’utilisation de cette méthodologie sont présentées : i) la conception pas à pas de système de production par incorporation progressive de pratiques élémentaires à partir d’une pratique d’entrée à effet majeur, pour accroître l’autonomie alimentaire en production bovine laitière en valorisant la prairie ; ii) l’évaluation à postériori des performances de systèmes en transcrivant sous forme de pratiques élémentaires des leviers d’action et marges de progrès identifiés à dire d’expert pour améliorer la compétitivité de la filière poulet de chair par la mise en place de deux systèmes de production très contrastés mais complémentaires. Les tendances d’évolution des performances des systèmes de production ainsi simulés ou reconstitués sont discutées. Dans les situations d’impact ambivalent d’une pratique sur une performance, le recours au descriptif détaillé des pratiquespermet le plus souvent de réduire l’incertitude en prenant en compte les situations associées à chaque type d’impact
